    Sequence-Based Mapping and Genome Editing Reveal Mutations in Stickleback Hps5 Cause Oculocutaneous Albinism and the casper Phenotype.

    Here, we present and characterize the spontaneous X-linked recessive mutation casper, which causes oculocutaneous albinism in threespine sticklebacks (Gasterosteus aculeatus). In humans, Hermansky-Pudlak syndrome results in pigmentation defects due to disrupted formation of the melanin-containing lysosomal-related organelle (LRO), the melanosome. casper mutants display not only reduced pigmentation of melanosomes in melanophores, but also reductions in the iridescent silver color from iridophores, while the yellow pigmentation from xanthophores appears unaffected. We mapped casper using high-throughput sequencing of genomic DNA from bulked casper mutants to a region of the stickleback X chromosome (chromosome 19) near the stickleback ortholog of Hermansky-Pudlak syndrome 5 (Hps5). casper mutants have an insertion of a single nucleotide in the sixth exon of Hps5, predicted to generate an early frameshift. Genome editing using CRISPR/Cas9 induced lesions in Hps5 and phenocopied the casper mutation. Injecting single or paired Hps5 guide RNAs revealed higher incidences of genomic deletions from paired guide RNAs compared to single gRNAs. Stickleback Hps5 provides a genetic system where a hemizygous locus in XY males and a diploid locus in XX females can be used to generate an easily scored visible phenotype, facilitating quantitative studies of different genome editing approaches. Lastly, we show the ability to better visualize patterns of fluorescent transgenic reporters in Hps5 mutant fish. Thus, Hps5 mutations present an opportunity to study pigmented LROs in the emerging stickleback model system, as well as a tool to aid in assaying genome editing and visualizing enhancer activity in transgenic fish

    Short-Term Belowground Responses to Thinning and Burning Treatments in Southwestern Ponderosa Pine Forests of the USA

    Microbial-mediated decomposition and nutrient mineralization are major drivers of forest productivity. As landscape-scale fuel reduction treatments are being implemented throughout the fire-prone western United States of America, it is important to evaluate operationally how these wildfire mitigation treatments alter belowground processes. We quantified these important belowground components before and after management-applied fuel treatments of thinning alone, thinning combined with prescribed fire, and prescribed fire in ponderosa pine (Pinus ponderosa) stands at the Southwest Plateau, Fire and Fire Surrogate site, Arizona. Fuel treatments did not alter pH, total carbon and nitrogen (N) concentrations, or base cations of the forest floor (O horizon) or mineral soil (0–5 cm) during this 2-year study. In situ rates of net N mineralization and nitrification in the surface mineral soil (0–15 cm) increased 6 months after thinning with prescribed fire treatments; thinning only resulted in net N immobilization. The rates returned to pre-treatment levels after one year. Based on phospholipid fatty acid composition, microbial communities in treated areas were similar to untreated areas (control) in the surface organic horizon and mineral soil (0–5 cm) after treatments. Soil potential enzyme activities were not significantly altered by any of the three fuel treatments. Our results suggest that a variety of one-time alternative fuel treatments can reduce fire hazard without degrading soil fertility

    Several solar cells having dissimilar spectral response curves and cell construction were measured at various locations in the United States to determine sensitivity of cell performance to atmospheric water vapor and turbidity. The locations selected represent a broad range of summer atmospheric conditions, from clear and dry to turbid and humid. Cell short circuit current under direct normal incidence sunlight, the intensity, water vapor and turbidity were measured. Regression equations were developed from the limited data base in order to provide a single method of prediction of cell current sensitivity to the atmospheric variables

    POPCORN: a Supervisory Control Simulation for Workload and Performance Research

    A multi-task simulation of a semi-automatic supervisory control system was developed to provide an environment in which training, operator strategy development, failure detection and resolution, levels of automation, and operator workload can be investigated. The goal was to develop a well-defined, but realistically complex, task that would lend itself to model-based analysis. The name of the task (POPCORN) reflects the visual display that depicts different task elements milling around waiting to be released and pop out to be performed. The operator's task was to complete each of 100 task elements that ere represented by different symbols, by selecting a target task and entering the desired a command. The simulated automatic system then completed the selected function automatically. Highly significant differences in performance, strategy, and rated workload were found as a function of all experimental manipulations (except reward/penalty)

    The relation between time of presentation and the sleep disturbing effects of nocturnally occurring jet aircraft flyovers

    The research describes the sleep disturbing effects of jet aircraft flyover noise as it occurs at different times during the night. The results indicate that individuals respond statistically more, in terms of greater cortical desynchronization, during the first and last thirds of the night. During the middle third, while there is still a significant amount of cortical desynchronization in response to the jet aircraft noise, this is always less than that which occurs earlier or later in the night. The implications of this data are that it might be possible to reduce the disruptive effects of at least certain extrinsic sonic stimulation by appropriate scheduling

    The fast contribution of visual-proprioceptive discrepancy to reach aftereffects and proprioceptive recalibration

    Adapting reaches to altered visual feedback not only leads to motor changes, but also to shifts in perceived hand location; “proprioceptive recalibration”. These changes are robust to many task variations and can occur quite rapidly. For instance, our previous study found both motor and sensory shifts arise in as few as 6 rotated-cursor training trials. The aim of this study is to investigate one of the training signals that contribute to these rapid sensory and motor changes. We do this by removing the visuomotor error signals associated with classic visuomotor rotation training; and provide only experience with a visual-proprioceptive discrepancy for training. While a force channel constrains reach direction 30o away from the target, the cursor representing the hand unerringly moves straight to the target. The resulting visual-proprioceptive discrepancy drives significant and rapid changes in no-cursor reaches and felt hand position, again within only 6 training trials. The extent of the sensory change is unexpectedly larger following the visual-proprioceptive discrepancy training. Not surprisingly the size of the reach aftereffects is substantially smaller than following classic visuomotor rotation training. However, the time course by which both changes emerge is similar in the two training types. These results suggest that even the mere exposure to a discrepancy between felt and seen hand location is a sufficient training signal to drive robust motor and sensory plasticity.York University Librarie

    Percolating cluster of center vortices and confinement

    We study the role of percolating clusters of center vortices in configurations of an Ising gauge theory in 3D. It is known that low energy features of gauge theories can be described in terms of an ``effective string picture'', and that confinement properties are associated with topologically non-trivial configurations. We focus our attention upon percolating clusters of center vortices, and present numerical evidence for the fact that these objects play a preminent role in confinement phenomenon, since their removal sweeps off confinement altogether.
