    Necessity of treating subclinical hypothyroidism in early pregnant women

    目的评估早孕妇女亚临床甲状腺功能减退症(subclinical hypothyroidism,SCH)对妊娠结局的影响及药物治疗的必要性。方法收集18 243例在厦门大学附属第一医院产前检查直至分娩的病例,诊断标准为妊娠10~12周产前检查时TT4与FT4正常而TSH≥2.5 m IU/L为SCH,甲状腺过氧化物酶抗体(thyroid peroxidase antibody,TPOAb)≥34 U/L为阳性。根据TSH的升高程度不同、TPOAb情况、能否遵医嘱按时按量服用L-T4使TSH控制在正常范围内进行分组:2.5 m IU/L≤TSH≤5.0 m IU/L且TPOAb阳性且能遵医嘱者为A组,不能遵医嘱者为B组;TSH>5 m IU/L且TPOAb阳性且能遵医嘱者为C组,不能遵医嘱者为D组;2.5 m IU/L≤TSH≤5.0 m IU/L且TPOAb阴性且能遵医嘱者为F组,不能遵医嘱者为G组;TSH>5 m IU/L且TPOAb阴性且能遵医嘱者为H组,不能遵医嘱者为K组。并随机抽取200例同期甲状腺功能正常的病例作为对照组(E组),回顾分析早孕妇女甲状腺功能异常的发生率、SCH对妊娠结局的影响、早孕妇女SCH行左旋甲状腺素片(L-T4)替代治疗的剂量及影响因素。结果早孕妇女SCH检出率为5.20%,甲状腺功能正常率为79.00%。A组早产3.31%(8/242)、妊娠期高血压7.02%(17/242)、胎儿生长受限7.02%(17/242)、出生低体质量儿6.61%(16/242)的发生率均低于B组的8.79%(8/91)、14.29%(13/91)、15.38(14/91)、14.29(13/91)(P均 5 m IU / L,TPOAb was positive and had good compliance; Group D: TSH and TPOAb as Group C but not have good compliance; Group F: 2. 5 m IU / L≤TSH≤5. 0 m IU / L,TPOAb was negative and had good compliance; Group G: as Group F,but not have good compliance; Group H: TSH > 5 m IU / L,TPOAb was negative and had good compliance; Group K: as Group H,but not have good compliance. Randomly selected 200 cases with normal thyroid function at the same period as the control group( Group E). Retrospective analysis the occurrence rate of thyroid gland dysfunction,the influence of SCH to pregnancy outcome,the doses and influencing factors of replacement therapy by L-T4 to SCH of early pregnant women. Results Relevance ratio of SCH in early pregnant women was5. 20%,the ratio of normal thyroid function was 79. 00%. In group A,premature birth 3. 31%( 8 /242),gestational hypertension 7. 02%( 17 /242),fetal growth restriction 7. 02%( 17 /242),low birth weight infant 6. 61%( 16 /242),were lower than that in group B 8. 79%( 8 /91),14. 29%( 13 /91),15. 38( 14 /91),14. 29( 13 /91) respectively( P < 0. 05). In group C,spontaneous abortion 3. 31%( 6 /181),premature birth 3. 89%( 7 /181),gestational hypertension 8. 29%( 15 /181),gestational diabetes mellitus 3. 89%( 7 /181),fetal growth restriction 7. 73%( 14 /181),low birth weight infant 6. 63%( 12 /181),were lower than that in group D 11. 54%( 6 /52),11. 54%( 6 /52),19. 23%( 10 /52),11. 54%( 6 /52),17. 31( 9 /52),15. 38( 8 /52)( P < 0. 05). No significant statistical difference were found in multiple comparison of the above six indexes of group A,B,C. And just the same as the multiple comparison of group F,G,H,K,E. When TSH reach the standard,the dosage of L-T4 in group A and C had significant difference [( 0. 757 ± 0. 378 vs. 1. 106 ± 0. 454) μg / kg,t = 8. 39,P < 0. 001]. There were also significant differences of group F compared with group H[( 0. 443 ± 0. 198 vs. 0. 813 ± 0. 378) μg/kg,t = 8. 22,P < 0. 001],group( A + F) compared with group( C + H) [( 0. 634 ± 0. 358 and 1. 017 ±0. 427) μg / kg,t = 11. 77,P < 0. 001],group A compared with group F [( 0. 757 ± 0. 378 vs. 0. 443 ± 0. 198) μg / kg,t = 9. 85,P < 0. 001 ],group C compared with group H [( 1. 106 ± 0. 45 vs. 0. 813 ± 0. 378) μg / kg,t = 5. 59,P < 0. 001],group( A + C) compared with group( F + H) [( 0. 932 ±0. 463 vs. 0. 693 ± 0. 388) μg / kg,t = 6. 53,P < 0. 001]. Conclusion The early pregnant women with SCH and TPOAb positive could increase the occurrence rate of spontaneous abortion,premature birth,gestational hypertension,gestational diabetes mellitus,fetal growth restriction and low birth weight infant. The early intervening treatment of L-T4 could efficiently decrease the adverse event occurrence ratio of pregnant woman with SCH and positive TPOAb,but no effect for that with SCH and TPOAb negative. The TSH level and condition of TPOAb could affect the replacement therapy dosage of early pregnancy with SCH

    目的初步探索泽泻[Alisma orientalis(Samuel.)Juzep.]降低2型糖尿病小鼠血糖的有效部位。方法采用超声提取法获得泽泻乙醇提取物,再分别用石油醚、乙酸乙酯、水饱和正丁醇和水相进行萃取分离,得到的泽泻乙醇提取物及其各溶剂萃取物应用于腹腔注射烟碱、链脲霉素诱导的2型糖尿病小鼠模型。造模成功后,将动物分组,其中空白对照组(未注射链脲霉素和烟碱的小鼠)和2型糖尿病模型组小鼠连续灌胃0.9%Na Cl溶液21 d(灌胃剂量为10μl/g),测试药物组分别连续灌胃泽泻乙醇提取物、泽泻石油醚萃取物、泽泻乙酸乙酯萃取物、泽泻正丁醇萃取物、泽泻水萃取物21 d(各测试药物灌胃高、中、低剂量分别为100μg/g、50μg/g、25μg/g),阳性对照组连续灌胃罗格列酮溶液21 d(灌胃剂量为10μg/g),每组8只小鼠,筛选具有降低2型糖尿病小鼠血糖的泽泻有效部位。用具有降低2型糖尿病小鼠血糖的有效部位处理3T3-L1前脂肪细胞,观察其对前脂肪细胞分化过程的影响。结果与2型糖尿病模型组比较,连续灌胃泽泻乙酸乙酯萃取物21 d(剂量为100μg/g)降低了2型糖尿病小鼠的血糖,改善了口服葡萄糖耐受试验(oral glucose tolerance test,OGTT)过程中的糖耐量(P<0.01)。1 mg/L和10 mg/L的泽泻乙酸乙酯萃取层均能促进前脂肪细胞的分化过程。结论泽泻乙酸乙酯萃取物降低了2型糖尿病小鼠血糖,促进了3T3-L1前脂肪细胞分化,但其具体作用机制尚需深入研究。Objective To explore the effective parts of Alisma orientalis( Samuel.) Juzep. associated with blood glucose decrease in type 2 diabetic mice.Methods Ethanol extracts from A. orientalis were obtained by ultrasonic extraction. Then,the ethanol extract was extracted and separated with petroleum ether,ethyl acetate,water saturated n-butanol and water respectively. After decompressing concentration,these solvent extractions were applied to type 2 diabetic mice model induced by intraperitoneal injection of streptozotocin and nicotine. After successful modeling,the mice were divided into the blank control group( without injection of streptozotocin and nicotine),model group of type 2 diabetes mellitus,positive control group and test drug groups,8 in each group. The blank control group and model group were treated with 0. 9% Na Cl at the dose of 10μl / g,the test drug groups were treated with corresponding extracts from different solvent extractions described above at the high-,mid-,low-dose of 100,50,25 μg / g respectively. The positive control group was treated with rosiglitazone at the dose of10μg / g. The treatment course was 21 days for intragastric administration. The effective parts of Alisma orientalis( Samuel.) Juzep. associated with blood glucose decrease in type 2 diabetic mice were screened and then the effect of the screened effective part on the differentiation of 3T3-L1 pre-adipocyte was observed.Results Compared with the model group,the blood glucose was decreased after the intragastric administration with ethyl acetate extract of A. orientalis at dose of100μg / g for 21 days and the glucose tolerance during the oral glucose tolerance test( OGTT) was improved( P < 0. 01). The differentiation of pre-adipocyte was promoted when the ethyl acetate extract of A. orientalis at the concentration of 1 mg / L and 10 mg / L.     Identification of Polygonatum from Wuyishan and Its Surrounding Areas by ISSR Molecular Markers and Their HPLC Fingerprints

    采用ISSR分子标记鉴定方法及HPLC色谱方法对武夷山及周边地区5份黄精植物样本进行DNA分子标记鉴定及指纹图谱分析,并通过聚类分析软件分别探讨其遗传相关聚类图谱。ISSR分子标记法能较好地对5份黄精植物样本进行区分,HPLC指纹图谱分析同样表明5份黄精样本化学成分存在一定的差别。DNA分子标记及HPLC指纹图谱分析的遗传相关聚类分析结果相似,表明武夷山及周边地区的野生黄精种质资源存在明显的差别。The purpose of this project was to identify and analyze the genetic relationships of 5 kinds of Polygonatum in Wuyishan and its surrounding areas. Using ISSR marker and HPLC molecular identification method for chromatographic methods analysis of DNA markers for identification and fingerprinting of 5 Polygonatum samples, the genetic correlation map of Polygonatum was studied by cluster analysis software. ISSR molecular marker method could be used to distinguish Polygonatum of Wuyishan and its surrounding areas. HPLC fingerprint analysis also showed that there were some differences in the composition of Wuyishan and its surrounding distribution. The results of genetic correlation cluster analysis were similar by ISSR and HPLC.     槐花提取物降血糖活性研究

    Effects of the Xitongxiao prescription on apoptosis and proliferation of chondrocytes in knee

    目的:探讨膝痛消方对促进软骨细胞增殖和抑制IL-1β诱导的软骨细胞凋亡的作用。方法:体外培养膝关节软骨细胞,用阿利新蓝染色方法检测软骨细胞蛋白多糖,用Ⅱ型胶原免疫荧光染色检测软骨细胞Ⅱ型胶原的表达,以鉴定软骨细胞;用MTS检测软骨细胞的增殖;用IL-1β诱导软骨细胞凋亡,AnnexinV/PI双染检测细胞凋亡。结果:膝关节剥离得到的细胞形态为多角形或梭形,阿利新蓝与Ⅱ型胶原免疫荧光染色均呈阳性,证明分离培养的细胞为软骨细胞。MTS实验结果发现膝痛消方提取物处理软骨细胞后检测到的OD值随浓度增加而增加且显著高于对照组。此外,IL-1β处理诱导软骨细胞凋亡,而膝痛消方提取物能够有效地抑制IL-1β引起的细胞凋亡。结论:膝痛消方能促进软骨细胞的增殖,抑制IL-1β诱导的软骨细胞的凋亡。以上发现为膝痛消方临床上治疗膝关节骨性关节炎提供药理作用依据。Objective:To investigate the effects of the Xitongxiao prescription on the cell proliferation and the inhibition of IL-1β-induced apoptosis of chondrocytes.Methods:To identify chondrocytes,the articular chondrocytes were cultured in vitro;staining with alcian blue was used to detect the protein polysaccharide,and staining with collagen immunofluorescence was used to examine the expression of type II collagen in chondrocytes.The proliferation of chondrocytes was determined by MTS.Apoptosis of chondrocytes was induced by IL-1β,and the effect of apoptosis was tested by Annexin V/PI double staining.Results:The morphology of the cells obtained from the knee joint was polygonal or fusiform.The tested results of Alcian blue and collagen immunofluorescence staining were positive.These together certified that the isolated cells were knee chondrocytes.MTS assay results showed that the detected OD values were dosedependently increased and they were significantly higher than those of the control group.In addition,the apoptosis of chondrocytes was induced by IL-1βtreatment.While the extract of the Xitongxiao prescription could effectively inhibit the IL-1β-induced apoptosis of chondrocytes.Conclusion:The Xitongxiao prescription could promote the proliferation of chondrocytes and inhibit the apoptosis of chondrocytes induced by IL-1β.The above findings may provide one of pharmacological mechanisms of the Xitongxiao for clinical treatment of knee osteoarthritis.厦门市科技计划项目(3502z20144031);福建省自然科学基金计划资助项目(2015J01065)

    The Communities of Ammonia-oxidizing Organisms in Pearl River Estuary Sediments

    [目的]对珠江口海岸带沉积物中的氨氧化细菌和古菌的组成进行分析,并进行定量研究。[方法]用构建克隆文库和Q-PCr定量的方法对珠江口沉积物中氨氧化细菌和古菌AMOA基因的含量和多样性特征进行研究。[结果]在2个沉积物表层,氨氧化古菌的含量是细菌的9和22倍,揭示氨氧化古菌在珠江口的氨氧化过程中起主导作用;系统发育分析表明大多数古菌和细菌的AMOA基因序列与不可培养的源于河口区和污染区域的环境克隆子序列有较高的同源性;细菌AMOA序列可分成5个类群(CluSTEr A、b、C、d和E),均属于nITrOSOMOnAS类群,其中CluSTEr A是主要类群(72.1%);古菌AMOA序列分析表明来自于表层的序列有52.2%属于“水/沉积物“簇,47.8%属于“土壤/沉积物“簇,而沉积物底层厌氧区,检测到的古菌AMOA基因93.3%属于“土壤/沉积物“簇,6.7%属于“水/沉积物“簇,且AMOA基因数量略高于表层。[结论]该研究有助于了解珠江口区域氮的循环过程,为氮的富营养化处理提供重要的理论依据。[Objective] This study aimed to investigate the abundance and composition of ammonia-oxidizing organism in Pearl River Estuary sediment.[Method] Based on amoA genes,we detected the abundance and composition of ammonia-oxidizing organism in Pearl River Estuary sediment by using quantitative real-time polymerase chain reaction(Q-PCR),cloning and sequencing approaches.[Result] The results of Q-PCR presented that ammonia-oxidizing archaea were more abundant than ammonia-oxidizing bacteria in the top of sediment cores,with AOA to AOB ratios 8.96(site Q5) and 3.69(site Q7).It suggested that ammonia-oxidizing archaea maybe play more important roles than ammonia-oxidizing bacteria in the process of ammonia oxidation in the Pearl River Estuary sediment.In the top sediment layer of Q7,bacterial amoA-like gene sequences were dominated by Nitrosomonas-like sequence types,which could be classified into five groups(clusters A,B,C,D,and E).Interestingly,archeal amoA-like gene was successfully amplified while bacterial amoA-like gene failed to be detected.These archeal amoA-like genes fell into two groups "water column/sediment" cluster and "soil/sediment" cluster.Most of the sequences(93.3%) in the bottom sediment layer of Q7 fell into "soil/sediment" cluster.[Conclusion] This study helps to realize the cycle of nitrogen in Pearl River Estuary region,and thus to provide theoretical support for the treatment of nitrogen eutrophication.国家973前期项目(2006CB708200);国家自然科学重点基金项目(40532011

    Effects of NaCl Stress on Photosynthesis of Three Bamboos

    采用盆栽试验法,对花叶唐竹(SInObAMbuSA TOOTSIk f.luTEOlO-AlbO-STrIATA)、小琴丝竹(bAMbuSA MulTIPlEX‘AlPHOnSE-kArr’)和刺黑竹(CHIMOnObAMbuSA nEOPurPurEA)进行不同浓度盐(nACl)胁迫(Ck、0.1%、0.3%、0.5%、0.7%、1.0%)处理,用美国lI-COr公司生产的lI-6400便携式光合作用分析系统测定其各项光合作用相关指标,分析研究其中重要的4项指标:净光合速率(Pn)、气孔导度(gS)、胞间二氧化碳浓度(CI)和蒸腾速率(E),以探究nACl胁迫对3种竹子叶片光合作用的影响。结果表明:低盐胁迫(0.1%nACl处理)提高了小琴丝竹和刺黑竹叶片净光合速率、气孔导度和蒸腾速率,而对花叶唐竹影响不大,但随着盐胁迫浓度的增加,3种竹类植物叶片的净光合速率、气孔导度和蒸腾速率呈逐渐下降的趋势;3种竹类植物胞间二氧化碳浓度随盐胁迫浓度的提高变化规律不明显。Four important indices of photosynthesis,including net photosynthetic rate(Pn),stomatal conductance(Gs),transpiration rate(E) and intercellular CO2(Ci) of the three bamboos of Sinobambusa tootsik f.luteolo-albostriata,Bambusa multiplex ‘Alphonse-karr’ and Chimonobambusa neopurpurea were studied by LI-6400 Portable Photosynthesis System and treatments by different concentrations of NaCl(CK-0.0%,S1-0.1%,S2-0.3%,S3-0.5%,S4-0.7% and S5-1.0%) in order to discuss the effects of NaCl stress on the photosynthesis of bamboos.The results showed that: low concentrations of NaCl stress(0.1%) promoted the Pn,Gs and E of Bambusa multiplex ‘Alphonse-karr’ and Chimonobambusa neopurpurea,but little effect on Sinobambusa tootsik f.luteolo-albo-striata;from 0.3% NaCl stress to 1.0% NaCl stress,with the increasing of salinity,Pn,Gs and E of all three bamboos were decreased;Ci of all three bamboos was not in good relationship with the increasing of salinity.厦门市科学技术局科研项目(No.3502Z20102003); 厦门市市政园林局科研项目(No.YK-2000-02
