    Transcriptome profile analysis of cell proliferation molecular processes during multicellular trichome formation induced by tomato Wo v gene in tobacco

    Trichomes, developing from the epidermis of nearly all terrestrial plants, provide good protection from environmental stress. Regulation of trichomes in Rosids has been well characterized. However, little is known about the cell proliferation molecular processes during multicellular trichome formation in Asterids. Ectopic expression of Wov in tobacco and potato induces much more trichome formation than wild type. To gain new insights into the underlying mechanisms during the processes of these trichomes formation, RNA-seq was employed for the young primary leaf tissues in Wov transgenic and wild-type tobacco. We identified differentially expressed genes which are related to various biological processes and molecular functions. Here, we provide details of experimental methods, RNA-seq data (available at Gene Expression Omnibus database under GSE72310).     A simulation study on the measurement of D0-D0bar mixing parameter y at BES-III

    We established a method on measuring the \dzdzb mixing parameter yy for BESIII experiment at the BEPCII e+ee^+e^- collider. In this method, the doubly tagged ψ(3770)D0D0\psi(3770) \to D^0 \overline{D^0} events, with one DD decays to CP-eigenstates and the other DD decays semileptonically, are used to reconstruct the signals. Since this analysis requires good e/πe/\pi separation, a likelihood approach, which combines the dE/dxdE/dx, time of flight and the electromagnetic shower detectors information, is used for particle identification. We estimate the sensitivity of the measurement of yy to be 0.007 based on a 20fb120fb^{-1} fully simulated MC sample.Comment: 6 pages, 7 figure

    The CDEX-1 1 kg Point-Contact Germanium Detector for Low Mass Dark Matter Searches

    The CDEX Collaboration has been established for direct detection of light dark matter particles, using ultra-low energy threshold p-type point-contact germanium detectors, in China JinPing underground Laboratory (CJPL). The first 1 kg point-contact germanium detector with a sub-keV energy threshold has been tested in a passive shielding system located in CJPL. The outputs from both the point-contact p+ electrode and the outside n+ electrode make it possible to scan the lower energy range of less than 1 keV and at the same time to detect the higher energy range up to 3 MeV. The outputs from both p+ and n+ electrode may also provide a more powerful method for signal discrimination for dark matter experiment. Some key parameters, including energy resolution, dead time, decay times of internal X-rays, and system stability, have been tested and measured. The results show that the 1 kg point-contact germanium detector, together with its shielding system and electronics, can run smoothly with good performances.     Impact of Limited Statistics on the Measured Hyper-Order Cumulants of Net-Proton Distributions in Heavy-Ion Collisions

    Hyper-order cumulants C5/C1C_5/C_1 and C6/C2C_6/C_2 of net-baryon distributions are anticipated to offer crucial insights into the phase transition from quark-gluon plasma to hadronic matter in heavy-ion collisions. However, the accuracy of C5C_5 and C6C_6 is highly contingent on the fine shape of the distribution's tail, the detectable range of which could be essentially truncated by low statistics. In this paper, we use the fast Skellam-based simulations, as well as the Ultrarelativistic Quantum Molecular Dynamics model, to assess the impact of limited statistics on the measurements of C5/C1C_5/C_1 and C6/C2C_6/C_2 of net-proton distributions at lower RHIC energies. Both ratios decrease from the unity baseline as we reduce statistics, and could even turn negative without a pertinent physics mechanism. By incorporating statistics akin to experimental data, we can replicate the net-proton C5/C1C_5/C_1 and C6/C2C_6/C_2 values comparable to the corresponding measurements for Au+Au collisions at sNN=\sqrt{s_{NN}} = 7.7, 11.5 and 14.5 GeV. Our findings underscore a caveat to the interpretation of the observed beam energy dependence of hyper-order cumulants.Comment: 6 pages, 7 figure

    The LAMOST Survey of Background Quasars in the Vicinity of the Andromeda and Triangulum Galaxies -- II. Results from the Commissioning Observations and the Pilot Surveys

    We present new quasars discovered in the vicinity of the Andromeda and Triangulum galaxies with the LAMOST during the 2010 and 2011 observational seasons. Quasar candidates are selected based on the available SDSS, KPNO 4 m telescope, XSTPS optical, and WISE near infrared photometric data. We present 509 new quasars discovered in a stripe of ~135 sq. deg from M31 to M33 along the Giant Stellar Stream in the 2011 pilot survey datasets, and also 17 new quasars discovered in an area of ~100 sq. deg that covers the central region and the southeastern halo of M31 in the 2010 commissioning datasets. These 526 new quasars have i magnitudes ranging from 15.5 to 20.0, redshifts from 0.1 to 3.2. They represent a significant increase of the number of identified quasars in the vicinity of M31 and M33. There are now 26, 62 and 139 known quasars in this region of the sky with i magnitudes brighter than 17.0, 17.5 and 18.0 respectively, of which 5, 20 and 75 are newly-discovered. These bright quasars provide an invaluable collection with which to probe the kinematics and chemistry of the ISM/IGM in the Local Group of galaxies. A total of 93 quasars are now known with locations within 2.5 deg of M31, of which 73 are newly discovered. Tens of quasars are now known to be located behind the Giant Stellar Stream, and hundreds behind the extended halo and its associated substructures of M31. The much enlarged sample of known quasars in the vicinity of M31 and M33 can potentially be utilized to construct a perfect astrometric reference frame to measure the minute PMs of M31 and M33, along with the PMs of substructures associated with the Local Group of galaxies.     DKK3 overexpression attenuates cardiac hypertrophy and fibrosis in an angiotensin-perfused animal model by regulating the ADAM17/ACE2 and GSK-3β/β-catenin pathways

    Aims Cardiac pressure and humoral factors induce cardiac hypertrophy and fibrosis, which are characterized by increased stiffness, reduced contractility and altered perfusion. Angiotensin II (AngII) is well known to promote this pathology. Angiotensin-converting enzyme (ACE) 2, which cleaves AngII and forms Ang-(1–7), exerts protective anti-hypertrophy and anti-fibrosis effects. A disintegrin and metalloproteinase 17 (ADAM17), a membrane-bound enzyme reported to cleave ACE2, may participate in the pathological process of AngII perfusion-induced heart damage. However, researchers have not clearly determined whether dickkopf-3 (DKK3) regulates the ADAM17/ACE2 pathway and, if so, whether DKK3-mediated regulation is related to the glycogen synthase kinase-3β (GSK-3β)/β-catenin pathway. In this study, we explored whether DKK3 overexpression ameliorates the development of AngII-induced cardiac fibrosis and hypertrophy through the ADAM17/ACE2 and GSK-3β/β-catenin pathways. Methods Mice were injected with a DKK3-overexpressing adenovirus or vehicle and then infused with AngII or saline using subcutaneously implanted mini-pumps for four weeks. Hearts were stained with hematoxylin-eosin, Masson's trichrome and immunohistochemical markers for histology. Primary fibroblasts were treated with the adenovirus and AngII and then examined using western blotting, EdU (5-ethynyl-2′-deoxyuridine) assays and immunofluorescence. Additionally, siRNA silencing was performed to study the role of DKK3 and the involved pathways. Results AngII-induced cardiac hypertrophy and interstitial and perivascular fibrosis were less severe in DKK3-overexpressing mice than in control mice. Moreover, the expression levels of fibrotic genes, such as collagen I and III, and the hypertrophic genes atrial natriuretic peptide (ANP) and beta-myosin heavy chain (β-MHC) were decreased. DKK3 overexpression also exerted a protective effect by inhibiting ADAM17 phosphorylation, thus increasing ACE2 expression and subsequently promoting AngII degradation. Furthermore, this process was mediated by the inhibition of GSK-3β and β-catenin and decreased translocation of β-catenin to the nucleus. On the other hand, the DKK3 knockdown by siRNA achieved opposite results. Conclusion DKK3 overexpression substantially alleviated AngII infusion-induced cardiac hypertrophy and fibrosis by regulating ADAM17/ACE2 pathway activity and inhibiting the GSK-3β/β-catenin pathway

    Existence of Tidal Tails for the Globular Cluster NGC 5824

    Context. Several dynamically cold streams have been associated with certain globular clusters (GCs) based on orbital energies and angular momenta. Some of these streams are surprisingly far apart from their progenitors and one such pair is Triangulum and NGC 5824. Triangulum can be considered as a piece of NGC 5824 leading tail since the cluster's future orbit matches with the stream's track well. The existence of the leading tail for NGC 5824 is the motivation behind the search for its trailing tail. Aims. Our goal is to confirm the connection between Triangulum and NGC 5824 and seek the trailing tail of the cluster. Methods. The selection of member stars of Triangulum is made through various cuts in metallicity, proper motions (PMs), radial velocity and color-magnitude diagram (CMD). The selected members are compared in phase space to a mock stream which models the disruption of NGC 5824. We then try to detect the trailing tail of the cluster based on a modified matched-filter technique. Stars are assigned weights using their color differences from the cluster's locus in CMD. These weights are further scaled based on stars' departures from expected PMs of the model stream. Results. A total of 26 member stars for Triangulum are obtained and 16 of them are newly identified. These members are consistent with the mock stream in the phase space and their metalicity and position on the CMD are in good agreements with NGC 5824. By applying the matched-filter, a tenuous trailing tail of the cluster is detected, spanning \sim 50^\circ long on sky. The signature matches with the mock stream's trajectory well. Conclusions. Our results support that Triangulum stream acts as a part of the leading tail for NGC 5824. On the trailing side, we have detected a 50^\circ tail extended from the cluster. The existence of both leading and trailing tails for the GC NGC 5824 is verified.Comment: Accepted for publication in A&

    Distilling Artificial Recombinants from Large Sets of Complete mtDNA Genomes

    BACKGROUND: Large-scale genome sequencing poses enormous problems to the logistics of laboratory work and data handling. When numerous fragments of different genomes are PCR amplified and sequenced in a laboratory, there is a high imminent risk of sample confusion. For genetic markers, such as mitochondrial DNA (mtDNA), which are free of natural recombination, single instances of sample mix-up involving different branches of the mtDNA phylogeny would give rise to reticulate patterns and should therefore be detectable. METHODOLOGY/PRINCIPAL FINDINGS: We have developed a strategy for comparing new complete mtDNA genomes, one by one, to a current skeleton of the worldwide mtDNA phylogeny. The mutations distinguishing the reference sequence from a putative recombinant sequence can then be allocated to two or more different branches of this phylogenetic skeleton. Thus, one would search for two (or three) near-matches in the total mtDNA database that together best explain the variation seen in the recombinants. The evolutionary pathway from the mtDNA tree connecting this pair together with the recombinant then generate a grid-like median network, from which one can read off the exchanged segments. CONCLUSIONS: We have applied this procedure to a large collection of complete human mtDNA sequences, where several recombinants could be distilled by our method. All these recombinant sequences were subsequently corrected by de novo experiments--fully concordant with the predictions from our data-analytical approach
