    Inhibitory effect of NRP-1mAb on the growth of gastric cancer cell BGC-823

    目的观察自主研发的抗NRP-1b1/b2IgG单克隆抗体(NRP-1mAb)对胃癌BGC-823细胞生长的影响,并初步探讨可能的作用机制。方法实验室制备NRP-1mAb,采用SDS-PAGE检测纯度。采用0、25、100、200、400μg/ml NRP-1mAb培养胃癌BGC-823细胞,光学显微镜下观察细胞形态学变化;采用MTT法、集落形成实验、流式细胞术观察胃癌细胞BGC-823的增殖、集落形成和凋亡的情况;Western blotting法检测NRP-1mAb作用后Akt、p38、ERK、JNK信号蛋白磷酸化情况。结果SDS-PAGE检测显示,NRP-1mAb的纯度在95%以上。光学显微镜观察显示,NRP-1mAb作用后,BGC-823细胞形态呈凋亡改变。MTT实验显示,NRP-1mAb抑制BGC-823细胞的增殖作用呈浓度和时间依赖性(P〈0.01)。集落形成实验显示,不同浓度NRP-1mAb均能显著抑制BGC-823细胞的集落形成,其抑制率呈浓度依赖性(P〈0.01)。流式细胞术检测显示,不同浓度NRP-1mAb均明显促进BGC-823细胞凋亡,且大部分集中在早期凋亡。Western blotting检测显示,BGC-823细胞的Akt磷酸化受到抑制,ERK、p38、JNK的磷酸化水平无明显变化。结论NRP-1mAb能抑制胃癌细胞BGC-823的生长、促进凋亡,可能与抑制Akt磷酸化有关。Objective To observe the effect of NRP-1b1/b2 IgG monoclonal antibody( NRP-1) on the growth of gastric cancer cell BGC-823,and to explore the possible mechanism of the antibody. Methods NRP-1mAb was prepared in laboratory,and the purity of antibody was detected by SDS-PAGE. Gastric cancer BGC-823 cells were cultured by 0,25,100,200,400 μg/ml NRP-1 mAb. The morphological changes of BGC-823 cells were observed by microscope. The proliferation,colony formation and apoptosis of gastric cancer BGC-823 cells were observed by MTT assay,colony forming assay and flow cytometry. The phosphorylation of related signal proteins was detected by Western blotting analysis. Results SDS-PAGE test showed that the purity of NRP-1mAb was above95%. Microscopy showed apoptotic changes of BGC-823 cells treated by NRP-1mAb. MTT assay showed that NRP-1mAb could inhibit the proliferation of BGC-823 cells in a time and dose dependent manner( P〈0. 01). Colony forming assay showed that different doses of NRP-1mAb could inhibit the colony formation of BGC-823 cells in a dose dependent manner( P〈0. 01). Flow cytometry showed that different doses of NRP-1mAb could promote the apoptosis of BGC-823 cells mainly at early apoptosis stage. It was found that the level of Akt phosphorylation was decreased after treated by NRP-1mAb,and there was no significant phosphorylation of ERK,p38 and JNK protein. Conclusion NRP-1mAb can inhibit the growth of gastric cancer cell BGC-823 and promote apoptosis,which may be related to the inhibition of Akt phosphorylation.厦门市科技计划创新资助项目(3502z20134026,3502z20144034

    Underwater acoustic speech communication using time reversal and time-frequency differential ofDM methods

    针对水声信道多径、时变、多普勒等恶劣传输特点对水声语音通信的严重影响,本文采用多通道时间反转和时频差分OfdM进行水声语音通信技术方案设计,该方法首先通过多通道时间反转进行时间域和空间域多径聚焦,进而结合时频差分OfdM调制解调抑制残留多径的影响。由于无需采用信道估计和均衡算法,系统实现方便、复杂度低,同时对信道具有一定程度的稳健性。该方法语音压缩编码采用混合激励线性预测编码。仿真实验和海试实验表明了本文方案的有效性。The difficulties of underwater acoustic channel, i.e., multipath, time varying and Doppler shifting pose significant challenges to underwater acoustic speech communication.In this paper, multi-channel time reversal is incorporated with time-frequency differential orthogonal frequency division multiplexing(ofDM)technology to design an underwater acoustic speech communication system, which enables time-frequency domain focusing of multipath by multi-channel time reversal, as well as suppressing of the residual multipath with time-frequency differential ofDM.Thus the employment of complicated channel estimation and equalization are avoided to facilitate the low complexity system implementation.In addition, the mixed-excitation linear predictive(MELP) is employed for speech encoding.The simulation and sea trial demonstrate the effectiveness of the method at presence of time varying multipath underwater acoustic channel.国家自然科学基金资助项目(11274259

    Two-dimensional Model Predictive Iterative Learning Control Scheme Based on a Two-dimensional Performance Model

    将迭代学习控制(ITErATIVE lEArnIng COnTrOl,IlC)系统看作一类具有2维动态特性的控制系统,根据模型预测控制(MOdEl PrEdICTIVE COnTrOl,MPC)和性能参考模型控制思想,提出了一种基于2维性能参考模型的2维模型预测迭代学习控制系统设计方案.在该控制系统设计方案中,可以通过选择适当的2维性能参考模型来构造2维动态变化的设定值信号和预测控制信号,从而引导迭代学习控制系统收敛到合理的控制性能,并有效避免系统性能收敛过程中控制输入可能发生的剧烈波动.通过对控制系统的结构分析可知,所得的迭代学习控制器本质上是由沿时间指标的参考模型预测控制器和沿周期指标的迭代学习控制器组成,闭环系统的收敛性等价于一个2维滤波系统的稳定性.数值仿真结果证明了该设计方案的有效性和鲁棒性.By representing an iterative learning control(ILC) system as a two-dimensional system and using the philosophy of model predictive control(MPC) and performance model reference control,a two-dimensional performance model based model predictive iterative learning control scheme is proposed in this paper.Through the design of two-dimensional dynamics of the performance model to generate more proper reference trajectories and predictive control signals for each cycle,the convergence pattern of the iterative learning control system can be guided to avoid the issue of possibly violent oscillation of input signal.The structure analysis indicates that the resulted control is composed of the time-wise performance model based model predictive control and cycle-wise iterative learning control,and that the convergence of the closed loop control system is equivalent to the stability of a two-dimensional filter.Numerical simulations illustrate the effectiveness and robustness of the proposed control scheme.国家自然科学基金(61174093)资助~

    Online Soft-measurement for Thermal Efficiency of Circulating Fluidized Bed Boilers

    提出循环流化床(Cfb)锅炉热效率的在线软测量方法,该方法将效率分为煤燃烧效率和传热效率两部分,根据Cfb锅炉动态机理模型在线计算锅炉未燃烧碳量,在此基础上计算煤燃烧效率,再通过反平衡法和正平衡法计算锅炉传热效率,最后得到锅炉热效率.应用该方法对某440T/H循环流化床锅炉热效率进行了计算,分析了影响该锅炉热效率的主要因素.结果表明:影响锅炉热效率的最主要因素是烟气中的飞灰含量;该软测量方法适用于Cfb锅炉,能有效指导锅炉调控,从而提高锅炉热效率.An online soft-measuring method for thermal efficiency of the circulating fluidized bed boiler(CFB)is being proposed.The method divides the efficiency into two parts,coal combustion efficiency and heat transfer efficiency.On-line calculation for the unburned carbon content of the boiler is carried out according to the dynamic mechanism model of the CFB boiler,after that combustion efficiency of the coal is calculated.Then heat transfer efficiency is calculated by indirect heat balance and direct heat balance,finally the boiler thermal efficiency is obtained.The thermal efficiency of certain 440 t/h CFB was calculated by the method,and the main factors which influence the thermal efficiency of the boiler were analyzed.Results show that the main factor which influences the thermal efficiency of the boiler is the content of fly ash in the flue gas.The soft-measuring method is suitable for CFB boiler,which can effectively direct the control of the boiler,thus improving the thermal efficiency of the boiler.福建省科技基金资助项目(2007J0165

    C4ISR系统是一个规模庞大、层次分明的离散动态系统,文章提出了一种面向C4ISR系统的UML和Petri网结合的建模分析方法。该方法采用从上到下的层次分析方法,用扩展的UML模型对系统组织结构和系统的静态特征进行描述,用扩展的Petri网模型描述和分析系统动态特性,通过相应的规则将Petri网模型转化成状态图和序列图。新方法可以对系统要求进行验证,同时还可以编程实现。最后通过该方法,对一个C4ISR系统进行了建模分析。C4ISR system is a discrete dynamic system with large scale and clearly hierarchy,this paper presents the modeling and analysis method combining UML with Petri net for C4ISR system.This approach adopts a hierarchical analytical method with top-down techniques,describes the whole systemic structure and static property through extended UML model,shows systemic dynamic characteristic using extended Petri net then transforms to the correspondence state diagram and sequence diagram.New methods verify system requirement and easily realize its code.At last the approach is illustrated by its application to C4ISR system.国家“十五”计划基金资助项目(40405010101C

    Steady-state simulation of fixed bed for coal gasification using ASPEN PLUS

    用ASPEn PluS软件对固定床煤气化过程进行模拟,采用带fOrTrAn气化动力学子程序的串联全混流反应器来代替gIbbS反应器,能够更好地反映气化炉的真实反应情况,结果表明该模型与实际固定床煤气化的反应结果吻合较好。利用该模型研究了串联釜数对碳转化率及出口温度的影响,研究表明:随着釜数的增加,碳转化率和出口温度更加接近于实际数据;在保证模拟精度的前提下,较少的釜数有利于减少计算量。ASPEN PLUS is used to simulate the fixed bed for coal gasification process.The gasification reaction kinetics is expressed by FORTRAN subroutine in RCSTRs,and then the RSCTRS in series are used to substitute the Gibbs reactor.So the reaction condition of the gasifier can be described more accurate in the simulation and the results of the simulation are in agreement with the actual fixed bed of the coal gasification process.The proposed model was applied to study the effect of the number of RCSTRs on carbon conversion and outlet temperature.The results show that carbon conversion and outlet temperature are closer to the actual data with the number increase of RCSTRs.However,less number of RCSTRs helps to reduce the computation time.With consideration of the accuracy and computation time of the simulation for coal gasification,it is suitable to use 6 RCSTRs in series to describe the fixed bed according to the results of the simulation.中央高校基本科研业务费专项资金资助项目(2010121047

    Study of solderable lead free tin-bismuth alloy electrodeposition

    提出了一种酸性锡铋合金电沉积工艺。研究了镀液中硫酸铋含量和电流密度对镀层铋含量、电流效率及镀层表面形貌的影响。另外 ,通过循环伏安曲线的测量研究了锡铋合金的电沉积过程 ,结果表明 ,镀层铋含量随硫酸铋含量的增大而增大 ,铋在 - 0 3V时开始析出 ,当电位达到 - 0 6V时 ,锡和铋共同沉积。A process for acid lead free tin-bismuth alloy electrodeposition was advanced. The effect of bismuth sulfate content in plating bath and current density on current efficiency, bismuth content and morphology of obtained tin-bismuth alloy was studied. In addition, electrochemical behavior of tin-bismuth alloy deposition was studied by cyclic voltammetry.     采用循环伏安(CV)和电化学石英晶体微天平(EQCM)方法研究了酸性介质中铜阳极溶出和阴极沉积过程以及丙烯基硫脲(AT)对该过程的影响.结果表明,铜阳极溶出和阴极沉积过程的M/n分别为32.0和34.2g/mol,都是两电子过程,其间未检测到Cu(Ⅰ)中间产物.AT改变了铜阳极溶出和阴极沉积的历程.在含AT的溶液中,铜阳极溶出和阴极沉积过程的M/n分别为61.9和65.4g/mol,可指认铜阳极溶出产物为CuAT+,并提出了AT存在下Cu阳极溶出和阴极沉积过程的反应机理;从电极表面质量定量变化的角度提供了Cu阳极溶出和阴极沉积过程的新数据
