Methods; The existing problems in POCT quality management were analyzed. Taking; portable blood glucose meters as an example,the quality management; system of POCT-Internet of things was designed based on the all aspects; of data acquisition,transmission, server management and storage,etc.; Results The POCT quality management system based on the Internet of; things and medical cloud technology was designed. The system could; implement the management of POCT operators and devices,monitor the; realtime quality control data and reports,and facilitate the; administrators to analyze the quality problems in the whole process of; POCT management.     Biological effects of Axin gene expression in glioma C6 cells

    目的研究Axin基因对神经胶质瘤细胞C6生物学特性及相关蛋白的影响。方法采用噻唑蓝(MTT)比色法测定细胞生长曲线,应用流式细胞仪(FCM)测定细胞周期的变化,并用平板克隆形成实验对细胞克隆形成能力进行了检测。应用末端TdT酶标记技术(TUNEL)观察细胞凋亡及比率。采用免疫细胞化学染色法检测Ki67、CyclinD1、p53的表达。结果转染Axin基因使细胞周期在G1期阻滞;细胞增殖能力和细胞克隆形成能力均显著降低;细胞凋亡率增高;Ki67及CyclinD1低表达,p53高表达。结论转染Axin基因后胶质瘤细胞出现了一定的增殖抑制现象,诱导细胞凋亡,Axin基因可能通过上调p53基因表达和下调Cy-clinD1表达而抑制胶质瘤细胞的生长。 【英文摘要】 Objective:To observe the effects of Axin gene expression on the biological characteristics of glioma cells. Methods:The proliferation ability of transfected cells was examined by MTT assay, cell growth curve and colony formation assay. The change of cell cycle was analyses by flow cytometry (FCM).The apoptosis was measured by TUNEL staining. Immunostaining was used to detect the expression of Ki67, CyclinD1 and p53 in C6 cells and transfected cells.     综合医院与结核病防治机构合作提高肺结核患者发现率的研究

    目的探索提高肺结核患者发现率的新途径,加快患者发现进程,实现世界卫生组织(WHO)提出的全球结核病控制阶段性目标。方法选择湖北省2002年度涂阳肺结核登记率低于全省平均水平的40个县,在每个县选择1家门诊量最大的综合医院和结核病防治中心作为研究项目实施单位。通过对综合医院相关科室医务人员进行培训和督导,从而在综合医院建立肺结核可疑症状者诊断、疫情报告和转诊工作规范。利用月报和季度报表收集资料,采用χ2检验对率和构成比进行统计分析,处理过程通过SAS 8.1实现。结果2003年10月至2004年9月期间,40个县累计发现新涂阳肺结核患者12 193例,比项目实施前同期增加了3 965例;新涂阳患者登记率由项目前同期的33.6/10万提高到49.6/10万(χ2=760.8,P<0.01),患者发现率由57.8%提高到85.5%。40个县综合医院门诊对肺结核可疑症状者查痰率达到48.6%,涂阳检出率为15.6%,患者转诊率为29.7%。医院门诊登记的11 303例可疑症状者中,共查出涂阳肺结核患者1 663例,占40个县涂阳患者发现总数的11.2%。结论加强综合医院与结核病防治中心的协调与合作,特别是加强医院肺结核患者和肺结核可疑症状者的转诊和追踪调查,能够快速地提高新涂阳肺结核患者发现水平

    Soil heterotrophic respiration in Casuarina equisetifolia plantation at different stand ages

    以中国亚热带木麻黄沿海防护林为研究对象,在2006年5月-2007年4月,利用lI-8100土壤呼吸自动观测系统对不同林龄(幼林、中林、成林)木麻黄人工林生态系统的土壤异养呼吸特征进行了监测。结果表明,不同林龄木麻黄林地土壤异养呼吸季节动态均呈单峰曲线,最大值出现在6~7月份,最小值则出现在12~1月份。土壤温度和水分对土壤异养呼吸的季节变化存在显著影响,并有明显的交互作用,进行单因素方差分析发现,土壤异养呼吸季节变化与5CM深的土温存在着较好的指数相关关系(P成林>幼林。随着林龄的增大,土壤异养呼吸对土壤温度的敏感性增强。土壤异养呼吸对土壤总呼吸的贡献在幼林、中林、成林中分别达到71.89%、71.02%和73.53%。幼林、中林、成林样地土壤异养呼吸CO2年释放量分别为29.072、38.964和30.530T·HM-2·A-1。The soil respiration rates (Rh) in 6-year-old (young), 17-year-old (middle-age), 31-year-old (mature) Casuarina equisetifolia coastal plantations were measured using an LICOR-8100 automated soil CO2 flux system from May 2006 to April 2007.Results show that Rh displayed an obvious seasonal pattern across the observed years.The maximum values of Rh occurred at June and July and the minimum at December and January.Soil temperature and soil moisture as well as their interaction had significant effects on the monthly dynamics of Rh.The analysis by one-way ANOVA showed that Rh had a significantly exponential relation (pmature plantation>young-age plantation.With the increase of growth age of plantation, the Q10 of Rh increased.The contribution of Rh to total soil surface CO2 flux was 71.89%, 71.02% and 73.53% for the young, middle-age and mature plantation, respectively.It was estimated that the annual CO2 fluxes from Rh were 29.07, 38.964 and 30.530 t·ha-1·a-1 for the young, middle-age and mature plantation, respectively.supportedbyKeyProjectsintheNationalScience&TechnologyPillarProgramduringtheEleventhFive-yearPlanPeriod(Nos.2006BAD03A14-01);ImportantScience&TechnologySpecificProjectsofFujianprovince(2006NZ0001-2);supportedbytheKeyLaboratoryofsouthmountaintimberculti-va

    目的:合成25-羟基维生素d3人工完全抗原,并制备抗25-羟基维生素d3的特异性抗体。方法:将25-羟基维生素d3进行化学修饰加入羧基活性基团,合成具有半抗原结构特征的25-羟基维生素d3-半琥珀酸酯。采用碳二亚胺法,将25-羟基维生素d3-半琥珀酸酯,分别与牛血清白蛋白(bSA)和卵清蛋白(OVA)偶联,合成人工完全抗原25-羟基维生素d3-半琥珀酸酯-bSA和25-羟基维生素d3-半琥珀酸酯-OVA。通过紫外吸收光谱,SdS-PAgE和MAldI-TOf进行偶联鉴定。用25-羟基维生素d3-半琥珀酸酯-bSA免疫小鼠,获得抗25-羟基维生素d3抗体免疫血清。结果:25-羟基维生素d3-半琥珀酸酯与bSA的偶联比为(12±0.16)∶1,免疫小鼠后获得高效价(效价为6.25x10-4)的抗体,且标准品浓度在37.5~600 ng/Ml范围具有显著的竞争性线性关系,检测的灵敏度为37.5 ng/Ml。结论:成功合成了25-羟基维生素d3人工完全抗原,制备出25-羟基维生素d3的抗体且其线性关系显著,灵敏度较高,为进一步研制检测25-羟基维生素d3的试剂盒奠定了基础。AIM: To synthesize the 25-hydroxyvitamin D3 artificial complete antigen and to prepare the specific antibody against 25-hydroxyvitamin D3.METHODS: The active group carboxyl was introduced into 25-hydroxyvitamin D3 and formed 25-hydroxyvitamin D3-hemisuccinate which possessed the structure of the hapten by chemical modification.The EDC method was applied to conjugate 25-hydroxyvitamin D3-hemisuccinate to bovine serum albumin as an artificial immunogen.The coating antigen 25-hydroxyvitamin D3-hemisuccinate-OVA was obtained in the same way.Ultraviolet,SDS-PAGE and MALDI-Tof were used to identify 25-hydroxyvitamin D3-hemisuccinate-BSA.RESULTS: BALB/c mice were immunized with 25-hydroxyvitamin D3-hemisuccinate-BSA to generate the polyclonal antibody of the 25-hydroxyvitamin D3 worth high titer and the immunogen,25-hydroxyvitamin D3-hemisuccinate-BSA,was successfully prepared with coupling ratio(12±0.16)∶1(N=3) coupling.CONCLUSION: The high titer and good sensitivity of anti-25-hydroxyvitamin D3 antibody are produced in sera immunized BALB/c mice,which made it possible to develop a clinical diagnostics for illness
