    Culture biology characteristics and haemocytes immune function of Sepiella maindroni

    曼氏无针乌贼(Sepiellamaindroni)曾是舟山渔场四大海产之一。上世纪九十年代以来,由于过度捕捞,自然资源遭到极大破坏。进行人工繁育是对曼氏无针乌贼资源进行保护和恢复的有效方法。本研究主要包括人工养殖条件下的曼氏无针乌贼的繁殖生物学、环境因子耐受性、生长特性、营养成分分析与评价,以及曼氏无针乌贼血细胞的分类及免疫特性等研究,为曼氏无针乌贼的人工增养殖和育种提供理论依据,并为曼氏无针乌贼的规模化生产奠定基础。 在人工养殖条件下,曼氏无针乌贼的繁殖习性发生了重大改变,养殖的曼氏无针乌贼由原来每年的单次到两次的繁育活动。曼氏无针乌贼的卵巢发育与温度有明显相关,较高的温度(21.5-23...Sepiella maindroni once was one of the four famous marine products of Zhoushan fishing ground in 1970s.     采用不同固定剂和染色剂对曼氏无针乌贼的血细胞进行处理,利用组织学的手段对血细胞形态进行了观察、记数和测量.曼氏无针乌贼血细胞浓度平均为2.5×107cells/mL.根据细胞质中颗粒的多少、大小、折光性、染色特性及血细胞的大小、核质比等,将曼氏无针乌贼的血细胞分为二大类:透明细胞和颗粒细胞.透明细胞近圆形,直径7.65~8.28μm,细胞核卵圆形,较大,细胞质中颗粒很少,核质比约0.66,约占总细胞数的45.3%;颗粒细胞圆形,直径9.58~10.42μm,细胞质中有大量嗜酸性颗粒,核质比较小,约0.40,有些具双核,约占总细胞数的54.7%.研究了曼氏无针乌贼血细胞的形态和分类,为曼氏无针乌贼血细胞免疫的深入研究奠定了基础.Study on the morphology of the haemocytes of the cuttlefish Sepiella maindroni was conducted by the method of histological staining in this paper.The average density of haemocytes was 2.5×107 cells/mL.The haemocytes of the cuttlefish S.maindroni were classified into two types,hyalinocytes and granulocytes,according to the characteristics of the haemocytes.Hyalinocytes with the size of 7.65~8.28 μm,were roundness,the nucleolus were also roundness surrounded by relatively small cytoplasm,there were little granule in cytoplast,the nucleus/cytoplasm(N/C) ratio was higher(0.66±0.10),and the proportion of haemocytes was 45.3% in the total haemocytes.Granulocytes with the size of 9.58~10.42 μm were roundness,there were much granules in the endoplasm and the N /C ratio was lower(0.40±0.06),some of the granulocytes had double nuclei,and the proportion of granulocytes was 54.7% in the total haemocytes.This research might be a foundation for further study on haemocytes immune of S.maindroni.科技部重大国际科技合作项目(2006DFA32960);; 浙江省对外合作重大专项(2006C14017)资

    采用SSr方法进行了闽粤群(My)和岱衢群(dQ)及其正交和反交大黄鱼的遗传差异的研究。结果表明,SHAnnOn’S多样性指数为dQ群高于My群,分别为2.14、2.04;反交群高于正交群分别为2.13、2.03;杂交群体中为2.24与其亲体群的相似(2.22)。亲体群间基因分化系数fST值为0.020,低于杂交后代的0.026。AMOVA显示,亲体群和后代群基因差异均很低,且亲体群基因分化度低于和杂交后代群,fST值分别为0.011、0.032。nEI遗传距离显示子代间遗传距离大与亲代代间的遗传距离分别为0.39、0.22。uPgMA聚类将其分为3组,其中dQ群与My群遗传距离最近。可以认为养殖的dQ群与My群大黄鱼间遗传背景差异较低,其中dQ族遗传背景较好,杂交后代群的基因多样性和基因差异性稍有提高,其中反交效果较好。The genetic diversity of 120 accessions of breeding Large Yellow Croaker, Pseudosciaena crocea (Richardson) was analyzed.Using 11 pair of SSR polymorphic markers, a total of 26 alleles were found, which exhibited high level of diversity.The range of polymorphism information content (PIC) was 0.785—0.818, average 0.804.Shannon’s Information index of DQ populations was higher than MY populations and it was 2.14, 2.04 respectively.The filial generation higher than parental generation it was 2.24, 2.22 respectively.And minus-hybrid generation higher than plus-hybrid generation it was 2.13, 2.03 respectively.The Fst of parental generation was lower than finial generation; it was 0.020, 0.026 respectively.The Gene flow was very high according to Fst value.The UPGMA clustering analysis at the genetic similarity grouped the four populations into three groups.The minus-hybrid stock was the most far away from others populations.AMOVA in filial generation higher than parental generation it P value was 0.01, 0.03 respectively.国际科技合作项目;2006DFA32960号;国家公益性行业科研专项;200903029-06

    Influence of different material plastic plates on the planula larvae settlement of moon jellyfish Aurelia sp.1

    The influence of different material plastic plates(DMPP),including high density polyethylene(HDPE),low density polyethylene(LDPE),polyvinyl chloride(PVC),polystyrene(PS),polyethylene terephthalate(PET),and polyurethane(PUR),and the relationship between the roughness of plates and the settlement rates on the settlement of the larval moon jellyfish Aurelia sp.1 were investigated in situ.As result,it showed firstly that polyps of Aurelia sp.1 found on the DMPP during the early autumn indicates that the increasing artificial structures in coastal waters might have provided much suitable habitats for the settlement and proliferation of polyps.Secondly,the larvae showed a clear settling selection on the DMPP.The settlement rates of larvae were higher in the plastic plates made of PET,PUR and PVC than other three materials.The settlement rate of larvae was the highest in plastic plates made of PUR (3.21 ± 0.74 ind./cm~2),while the lowest in plastic plates made of HDPE (0.26 ± 0.08 ind./cm~2).Thirdly,the settlement rate of larvae was not significant related to the roughness value (Ra) of the materials.Therefore,the surface roughness of a material is not the key factor influencing the settlement rate of the larvae.The difference in biofilm formed on the plastic plate may also influence the settlement of larvae.These findings may provide important information on the causes and control of moon jellyfish blooms in coastal waters of China

    用组织学和免疫学方法对曼氏无针乌贼(Sepiella maindroni)的血细胞进行形态观察和免疫活性研究。根据血细胞大小和美兰染色结果,将曼氏无针乌贼的血细胞分为2类:较小的血细胞圆形,直径7.65~8.28μm,细胞核卵圆形,核质比约0.66,约占细胞总数的45.3%;较大的血细胞圆形,直径9.58~10.42μm,细胞核肾形,核质比约0.40,有些细胞具双核,约占细胞总数的54.7%。饥饿乌贼血细胞数量较正常乌贼有明显下降(t-检验,P0.05)。对曼氏无针乌贼血细胞形态和功能的研究,为探索曼氏无针乌贼免疫防御机理提供了基础资料。Morphological and phagocytosis properties of the haemocytes of the cuttlefish Sepiella maindroni were studied by histology and immunology methods. The results showed that the haemocytes of the S. maindroni could be divided into two types according to the characteristics of the haemocytes: the lesser haemocytes with the diameter of 7.65~8.28 μm, were round, the nucleolus were also round,surrounded by relatively small cytoplasms, there were little granules in cytoplast, the ratio of nucleus/cytoplasm (N/C) was higher (0.66 ± 0.10), and the proportion of haemocytes was 45.3% in the total haemocytes; the biggish haemocytes with the size of 9.58~10.42μm, were round, there were much granules in the endoplasm and the N /C ratio was lower (0. 40±0.06) , some of the granulocytes had double nuclei, and the proportion of granulocytes was 54.7% in the total haemocytes. The density of haemocytes of starved S. maindroni was lower than that of the control ones (t- test, P0.05).     主要研究了不同施肥处理(肥料种类不同和施肥量配比不同)对温室樱桃番茄果实和土壤硝酸盐含量变化的影响.结果表明:不同施肥处理果实中硝酸盐含量随追肥次数的增加,逐渐加大,到采收中期达生育期内最大值,最终略有累积;果实中硝酸盐含量为49.6~273.0 m g.k-g 1,亚硝酸盐含量为0.011~0.054 m g.kg-1.第4次追肥后果实与土壤(0~40 cm土层)中硝酸盐含量在追肥后6~9 d达最大值,以后开始下降.果实内硝酸盐的累积量与土壤硝酸盐含量成显著正相关.合理的氮肥施用量不会造成果实内硝酸盐含量的过量累积;有机肥配施适量化肥对改善果实品质、提高果实产量、调节土壤硝酸盐含量变化有着很好的效果
