    应用ARIMA模型预测厦门市2017年乙肝月发病率

    目的分析厦门市乙型肝炎发病的流行病学特征,建立ARIMA模型并预测2017年厦门市乙肝月发病率,为厦门市乙肝防治提供理论依据。方法对2005-2016年厦门市乙肝报告发病数据进行统计描述,通过ARIMA模型预测2017年的乙肝月发病情况。结果厦门市2005-2016年乙肝报告年均发病率为45.70/10万,乙肝年发病率呈逐渐下降趋势;男性乙肝发病高于女性;各年龄组乙肝发病水平存在差异;15岁以下儿童乙肝发病率显著低于其他高年龄组的发病水平。建立模型ARIMA(1,1,1)×(0,1,0)12,模型的拟合效果较好,2017年厦门市乙肝月发病率预测值区间为[0.92/10万,4.76/10万]。结论厦门市2005-2016年乙肝发病总体呈现下降趋势,经预测2017年厦门市乙肝发病率与前几年相比波动不大,有轻微下降趋势,可用ARIMA模型对乙肝的短期发病情况进行预测

    Development and Application of a Novel Neutralization Test for Echovirus 25

    目的:建立一种新型的快速、高通量的埃可病毒25型(ECHO25)中和抗体检测方法,并初步评价其在ECHO25中和抗体筛选和血清流行病学调查中的应用价值。方法:应用免疫荧光方法筛选ECHO25高亲和性抗体并将其作为检测单抗,结合酶联免疫斑点检测技术(ELISPOT)建立ECHO25中和抗体检测方法;使用不同效价的血清评价该方法的准确性;采用所建立的中和方法对ECHO25单克隆抗体、临床血清样品进行检测。结果:建立了快速检测ECHO25中和抗体的Nt-ELISPOT方法,以ECHO25单克隆抗体5B9作为检测抗体;相比经典的中和实验方法 Nt-CPE,该方法可显著缩短检测时间(从5~7 d缩短至1 d以内),检测结果具有较好的一致性;采用所建立的Nt-ELISPOT方法首次筛选获得3株对ECHO25具有较好中和能力的单克隆抗体;临床血清样品检测结果显示厦门地区可能存在ECHO25的流行。结论:该方法可以应用于中和抗体筛选和血清学的临床辅助诊断,为ECHO25的防治研究提供支持。Objective: To establish a rapid and high-throughput neutralization test for echovirus 25(ECHO25),and evaluate its application in neutralizing antibody screening and seroepidemiological surveys. Methods: Immuno-fluorescence assay was applied to screen a high affinity antibody, which was used as the detection antibody forECHO25, and a rapid neutralization test was established based on enzyme- linked immunospot assay(Nt-ELISPOT). The accuracy of this method was evaluated by detecting serum samples with different titer. Monoclonalantibodies against ECHO25 and clinical serum samples were detected via the established neutralization test. Results: A rapid method to detect neutralizing antibody against ECHO25 was established and an anti-ECHO25 anti-body, 5B9, was used as the detection antibody. The detection period could be shortened significantly comparedwith the classical neutralization test(Nt- CPE)(from five to seven days to less than one day), and the Nt-ELISPOT had good consistency with the Nt- CPE. Meanwhile, three neutralizing antibodies for ECHO25 werescreened firstly by this method. The detection results of clinical serum samples showed that infection of ECHO25 might be popular in Xiamen. Conclusion: This method can be used in neutralizing antibody screening and seroepi-demiological surveys, and it may provide support for the control of ECHO25.国家自然科学基金(81371817,81401669

    Quantification of Methylation at CMV Promoter of pEGFP-C3 Vector by Gradient Touch-Down PCR Sequencing

    目的改进亚硫酸氢钠测序法,并在CMV基因启动子甲基化检测中进行验证。方法提取PEgfP-C3质粒重组人肝癌细胞株HEPg2 dnA,亚硫酸氢钠化学修饰,针对修饰后质粒基因CMV启动子序列设计特异引物并结合梯度降落PCr扩增,T-A载体克隆、测序,目标区域甲基化定量。结果 PEgfP-C3质粒基因约600bP的CMV启动子区甲基化水平可以精确定量,检测结果与标准品一致,重复测量结果稳定。结论改进后亚硫酸氢钠测序法能明显减少非特异性扩增,提高PCr效率,更适于基因甲基化状态的检测。Objective To improve the sodium bisulfite sequencing method and validate it in quantification of methylation in CMV promoter.Methods DNA was extracted from recombinant HepG2 hepatoma cell line with plasmid pEGFP-C3,and chemically modified with sodium bisulfite,the gene-specific primers were designed according to the modified CMV promoter sequence and conducted PCR amplification with gradient touch-down PCR,then the DNA methylation level in the target areas was quantitated after T-A cloning and sequencing.Results The DNA methylation level of the 600 bp CMV promoter within pEGFPC3 plasmid could be quantified accurately,and be consistent with that of the methylated DNA standards,repeated measurements indicated stable quantification result with this method.Conclusion The improved sodium bisulfite sequencing method can reduce non-specific amplification significantly and improve PCR efficiency,therefore has more potential for quantitative detection of gene methylation status.国家自然科学基金资助项目(20907047);中央级公益性科研院所基本科研业务专项(2008KYYW05

    Flexible decapyrrylcorannulene hosts

    球形笼状的富勒烯是上个世纪末最重要的科学发现之一,但对富勒烯的精确几何结构的认识却困难重重,原因是单晶中球形分子的取向往往是无序的,需通过笼外衍生或通过八乙基金属卟啉-富勒烯超分子主客体组装来固定富勒烯的取向,然后利用常用的单晶衍射分析技术来精确表征富勒烯的几何结构。然而许多富勒烯新结构因无法与卟啉主体形成高质量的单晶至今仍无法利用X射线衍射技术进行结构分析,直接制约了对富勒烯形成机理及结构-性能关系的深入认识。功能团簇材料创新研究群体的谢素原、张前炎课题组另辟蹊径地从曲面结构的十氯碗烯C20Cl10出发,合成了十吡咯取代的碗烯分子C20(C4H4N)10。结构分析表明该分子的结构特征是碗烯的碳框架与十个吡咯基团通过单键相连。实验还证明,用甲基去取代吡咯3, 4-位置的氢并不利于富勒烯与碗烯衍生物形成有序的超分子组装体,理论研究进一步诠释了十个吡咯‘手指’的集体贡献比单个碗烯‘手掌’更大的原因。该研究工作是功能团簇材料创新研究群体长期积累,并由校内外十多位研究人员共同努力完成。徐云彦(2014级硕士生)、田寒蕊(2014级博士生)和李姝慧(2016年进站博士后)为该论文共同第一作者。【Abstract】The assembly of spherical fullerenes, or buckyballs, into single crystals for crystallographic identification often suffers from disordered arrangement. Here we show a chiral configuration of decapyrrylcorannulene that has a concave ‘palm’ of corannulene and ten flexible electron-rich pyrryl group ‘fingers’ to mimic the smart molecular ‘hands’ for self-adaptably cradling various buckyballs in a (+)hand-ball-hand(−) mode. As exemplified by crystallographic identification of 15 buckyball structures representing pristine, exohedral, endohedral, dimeric and hetero-derivatization, the pyrryl groups twist with varying dihedral angles to adjust the interaction between decapyrrylcorannulene and fullerene. The self-adaptable electron-rich pyrryl groups, susceptible to methylation, are theoretically revealed to contribute more than the bowl-shaped palm of the corannulene in holding buckyball structures.
