    用焦锑酸钾沉淀法对宁夏枸杞柱头和花粉中的钙离子分布进行了研究.结果显示,宁夏枸杞柱头表皮有一覆盖层,其中有许多含钙沉淀颗粒的小泡,当花粉落到柱头后从覆盖层中吸水,在萌发孔的表面上聚集了较多的钙沉淀颗粒.同时,花粉内部出现许多含钙的小液泡,使花粉体积增大,内部产生膨压,花粉萌发;生长在覆盖层中的花粉管顶端穿过覆盖层小泡时,附近聚集了较多的钙沉淀颗粒,在花粉管壁上也附着较多的细小钙沉淀颗粒.萌发的花粉粒中由大液泡占据,在其亚顶端的细胞质中,聚集较多钙沉淀颗粒的线粒体膨大形成了一些含钙沉淀颗粒的小液泡,由这些小液泡融合形成的大液泡,将花粉管细胞质挤到其顶端,使其极性生长.这是首次发现在植物柱头覆盖层中有钙离子的现象,从体内证明了钙离子在花粉萌发过程中的现象.讨论了枸杞柱头组织中钙的分布和花粉管的萌发与生长的关系.The stigma of Lycium barbarum is wet type and some material to be secreted to the surface of stigma to form a layer of tectum at anthesis.There were many vesicles in the tectum and numerous calcium precipitated grana were accumulated in the tectum vesicles.When pollen grains landed onto the stigma they began to hydrate and form many small vacuoles in which some calcium precipitated grana located,which made a swelling pressure in pollen.During pollen germinating in the tectum,the pollen tubes often traversed tectum vesicles and displayed many calcium precipitated grana on the tube wall,suggesting it to absorb calcium of the tectum vesicles.In the base of pollen tube some mitochondria with calcium precipitated grana swelled to form small vacuoles which fused to form a large vacuole.The pollen tube cytoplasm was pushed into the top of tube to make sure directional growth.This is first time to report that stigma tectum containing rich calcium in flowering plants.The calcium distribution in the stigma and pollen germination and tube growth were also discussed.国家自然科学基金项目(30670126

    Screening, Identifying of Cellulose-Decomposing Strain L-06 and Its Enzyme-Producing Conditions

    从用于堆肥的水稻秸秆中初筛出一株高效纤维素降解菌L-06,根据18SrRNA基因序列和菌株形态分析,初步鉴定该菌为斜卧青霉(Penicillium decumbens)。研究了液体发酵培养基中氮源、碳源、表面活性剂、培养温度、起始pH以及接种量对该菌株各纤维素酶活力的影响。在最适条件下,该菌的β-葡萄糖苷酶(BGL)、外切纤维素酶(CBH)于培养第3天酶活力分别达到1662u/mL和2770u/mL;内切纤维素酶(EG)、滤纸糖化力(FPase)于培养第4天酶活力分别达到18064u/mL和4035u/mL。在产酶优化实验中,该菌的EG和CBH在pH10的培养条件下分别保持了70%和43%的酶活性;在50oC培养条件下EG和CBH分别保持了68%和59%的酶活性。表现出了耐热,耐碱的特性。Cellulases are relatively costly enzymes that are sold in large volumes for use in different industrial applications, and a significant reduction in cost will be important for their commercial use in biorefineries. The production of cellulase is a major factor in the hydrolysis of cellulosic materials. Hence it is essential to make the process economically viable. A strain (L-06) with high cellulase activity was screened from rice straw compost and classified as Penicillium decumbens by the analysis of its morphology and 18S rRNA gene sequences. Different conditions of liquid fermentation medium including nitrogen source, carbon source, surfactant, temperature, initial pH, inoculation quantity for the production of cellulase had been studied. The maximal β-1, 4-glucosidase(BGL) activity was 1662 u/mL which is 1.49 times of the previous and the maximal exo-β-1, 4-glucanases(CBH) activity was 2770 u/mL which is 1.36 times of the previous, cultured in the optimal condition for three days. And the maximal endo-β-1, 4-glucanases (EG) activity was 18064 u/mL which is 1.87 times of the previous and the maximal filter paper enzyme(FPase) activity was 4035 u/mL which is 1.47 times of the previous , cultured in the optimal condition for four days. In the optimization experiments, the EG and CBH in the culture condition (pH10) maintained 70% and 43% activity.     天然淡水冰单轴压缩强度及其温度和应变率效应

    本文报道了在不同温度和不同应变率下天然淡水冰单轴压缩强度的实验结果。指出S—l型柱状晶粒冰的压缩强度随着温度的降低而增加,极限压缩强度在应变率为10~(-4)S(-1)时有一个最大值。本实验的应变率范围是10~(-5)—10~0s~(-1)。10~(-1)—10~0s~(-1)的应变率范围是在冲击速度lm/s左右的实验条件下获得的,这个速度接近于浮冰与海上结构物相撞的情况。以本实验为基础,有希望发展对海冰的实验技术
