    Genetic Diversity of Thunnus thynnus and Thunnus albacares Stocks by AFLP

    利用选择性扩增片断长度多态性分子标记技术(AMPlIfIEd frAgMEnT lEngTH POlyMOrPHISM,AflP),分析了采自日本九州海域蓝鳍金枪鱼(THunnuS THynnuS)和台湾海域黄鳍金枪鱼(THunnuS AlbACArES)野生群体的遗传多样性水平.结果表明:9对选择性引物在2种金枪鱼中共扩增出675个位点(100--750 bP),其中蓝鳍金枪鱼和黄鳍金枪鱼多态位点分别为388个和368个.蓝鳍金枪鱼和黄鳍金枪鱼的多态位点比例、SHAnnOn遗传多样性指数分别为57.48%和54.52%,0.330 1和0.301 8.AMOVA分子方差分析显示:种间遗传分化中,83.79%的遗传变异由种间贡献,而16.21%的变异分布于种内个体之间.同其他鱼类比较,2种金枪鱼显示了较为丰富的遗传多样性水平,其种质资源处于较好的水平.研究结果将为我国蓝鳍金枪鱼和黄鳍金枪鱼的资源保护与合理利用提供重要的理论依据.Thunnus is a important commercially marine fish species,which is widely distributed.Thunnus has been intensively studied in China,including its basic biological characteristics in fishery and artificial reproduction.However,very few studies on Thunnus were focused on genetic diversity,which has become an obstacle to further researches on the protection,management,rational exploitation and sustainable utilization of Thunnus resources.The genetic diversity of wild Pacific blue-fin tuna(Thunnus thynnus)and Pacific yellow-fin tuna(Thunnus albacare)were investigated by using amplified fragment length polymorphic(AFLP) in the present paper.AFLP fingerprinting was regarded as one of the most useful techniques in nowadays for DNA fingerprint analysis.The results showed that a total of 675 DNA amplification bands ranging from 100--750 bp were produced using 9 pairs of primer combinations,and 35--54 polymorphic loci were detected per primer combination.The percentage of polymorphic loci and Shannon's information index of the blue-fin and yellow-fin tuna were 57.48%,0.330 1 and 54.52%,0.301 8,respectively.Both populations were at the same high level of genetic diversity.AMOVA analysis(83.79%) indicated that a certain extent of differentiation in two species.There were 83.79% of variance was among the populations and 16.21% of variance was within a population.Compared with other fishes,both tuna species possessed rich population genetic diversities.The information of genetic diversities in different populations will give us theoretical guidance in breeding and genetic improvement.福建省科技重大专项(2004NZ03)资

    采用同源克隆的方法、利用rACE技术从黄鳍金枪鱼(THunnuS AlbACArES)肝脏中克隆了其MIf(TAMIf)的CdnA序列。TAMIf的CdnA全长706 bP含一个345 bP的开放阅读框,编码一个长115氨基酸的蛋白质。信号肽预测分析表明TAMIf没有信号肽。序列分析与进化分析表明,黄鳍金枪鱼与近海养殖鱼类MIf的氨基酸序列高度相似、进化地位相近,预示着深海鱼类TAMIf与近海养殖鱼类具有相似的生物学功能。研究结果是对深海鱼类MIf基因信息资料的重要补充。The cDNA sequence of an MIF homologue(TaMIF) was cloned from the liver of Thunnus albacares by a homologous cloning strategy and RACE approach.The full-length cDNA of TaMIF was 706 bases long and contained an ORF of 345 bases encoding a protein of 115 amino acid residues.Predication of the signal peptide indicated that TaMIF lacks a signal peptide.Sequence analysis revealed a high structural similarity and phylogenetic analysis indicated a close relationship between TaMIF and other fish MIF,implying that TaMIF maybe have a simliar function to offshore cultured fish MIFs.The result is the complement of the gene information to open-sea fish MIF gene.福建省科技重大专项(2004NZ03

    Effect of Compound Pollution of Zn-Cd in Purple Soil on Lettuce under Simulating Acid Rain

    通过盆栽实验研究了酸雨影响下紫色土Zn-Cd复合污染对莴笋的影响。结果表明:酸雨影响下,Zn为低水平时,Zn-Cd复合污染对莴笋表现为拮抗增产;Zn为高水平时,复合污染对莴笋表现为协同减产。重金属累积量在Zn-Cd复合污染时表现为Cd对Zn的作用和Zn对Cd的作用恰好相反:在Zn为低浓度时,Zn促进了莴笋对Cd的吸收,Cd抑制了莴笋对Zn的吸收;在Zn为高浓度时,Zn抑制了莴笋对Cd的吸收,Cd促进了莴笋对Zn的吸收。酸雨对莴笋的生长起抑制作用,莴笋生物量随着酸雨pH值的升高而增加,而莴笋地上部分重金属的富集能力也随之增强,并且Cd的富集能力明显大于Zn,莴笋可食部分毒性也随之增大。Pot experiments under simulating acid rain were conducted to study the effect of Cd-Zn compound polluted on lettuce(Lactuca sativa var.angustana Irish) in purple soil.The results showed that Zn counteracted Cd to improve the yield when the concentration of Zn was low,Zn also cooperated with Cd to reduce the yield when the concentration of Zn was high.The Cd-Zn compound pollution indicated that Zn promoted the absorption of Cd by the lettuce,and Cd restrained the absorption of Cd by the lettuce,under the condition of low Zn level;Zn restrained the absorption of Cd by the lettuce,and Cd promoted the absorption of Cd by the lettuce,under the condition of high Zn level.Acid rain restrained the growth of lettuce.Biology quantity of lettuce and cumulating coefficient of Zn, Cd in lettuce increased when pH of the acid rain increased,and cumulating ability of Cd was bigger than that of Zn,which resulted in heavy metal toxicity of edible part increasing.重庆市生态环境保护行动计划项
