跨海集群工程节能减排关键技术研究
港珠澳大桥跨海集群工程囊括跨海桥梁、沉管隧道、离岸人工岛等多个领域。跨海集群工程由于建设期及运营期能耗巨大,国内外尚无节能减排的成套技术指导工程建设,成为制约跨海集群工程持续发展的瓶颈之一。本文依托港珠澳大桥跨海集群工程建设和运营过程,采用理论研究、数值分析及模拟测试等多种方法,开展了集群工程节能减排指标体系和长大沉管隧道通风、照明、人工岛运营节能减排关键技术研究,成果支撑了工程项目节能减排目标的实现
Efficacy of ulinastatin combined with gamma globulin in treating patients with acute purulent peritonitis
目的观察研究乌司他丁联合丙种球蛋白治疗急性化脓性腹膜炎的疗效分析。方法选取厦门大学附属第一医院杏林分院普外科2013年3月~2015年3月收治的确诊为急性化脓性腹膜炎的患者共70例,按随机数字表法分为观察组和对照组,每组各35例,对照组给予常规保守治疗,观察组给予乌司他丁联合丙种球蛋白治疗,治疗结束后,随访并记录2组患者相关指标的变化等情况。结果观察组患者治疗后内毒素含量为(626.9±130.5)pg/mL,低于对照组(867.4±124.2)pg/mL(P<0.05)。观察组患者治疗后有效率高于对照组(97.14%vs.77.14%;χ~2=7.467,P<0.05)。结论乌司他丁联合丙种球蛋白治疗急性化脓性腹膜炎的临床治疗效果确切,整体治疗效果优于常规治疗。Objective To observe the efficacy of ulinastatin combined with gamma globulin in treating patients with acute purulent peritonitis.Methods 70 cases diagnosed with acute purulent peritonitis from March 2013 to March 2015 were randomly divided into observation group and control group,35 cases in each group. The control group received conventional treatment and observation group received ulinastatin combined with gamma globulin treatment. The related indicators were followed up and recorded in two groups post-treatment. Results The endotoxin content post-treatment in observation group was lower than control group[( 626. 9 ± 130. 5) vs.( 867. 4 ± 124. 2) pg / mL,P < 0. 05]. The effective rate in observation group was higher than control group( 97. 14% vs. 77. 14%; χ~2= 7. 467,P < 0. 05). An Investigation of the Professional Development of Primary and Secondary School P. E. Backbone Teachers in Fujian Province
采用文献法、调查法、统计法、逻辑法等研究方法,以参加2013—2015年福建省基础教育万名骨干教师培训中的190名中小学体育骨干教师为调查对象,对教师基本情况和教师专业发展的认识、态度、知识、能力、继续教育等现状进行调查,在此基础上对教师专业发展现状进行分析,总结出我省中小学体育骨干教师专业发展的特点及存在的问题,并提出相应的建议,给我省中小学体育教师培训机构的改革提供依据,同时为我省中小学体育教师专业发展提供参考与借鉴,为进一步深化我省中小学体育教育改革提供人才保障.结果表明:1)目前,福建省中小学体育骨干教师以年富力强者居多,年龄、职称结构较合理;教师专业发展意识强,学历层次由大专逐步向本科、硕士过度,但硕士比例小;2)教师专业发展态度端正,职业的稳定性和责任感强烈,但部分年轻教师缺乏专业发展的自主性;3)教师对专业知识的掌握比较扎实,主要通过互联网、图书资料、专家报告或讲座等方法来获取专业知识;4)专业能力水平方面,教师在体育教学、课外指导、体育保健、现代教育技术运用等方面能力比较强,而创新教育和科研两方面能力相对比较薄弱;5)当下,我省中小学体育骨干教师最需要强化的培训是课题申报技能、论文写作能力、专题讲座能力、教育科研方法、提高学历层次、教学设计能力,其中提高科研能力和水平是教师们最迫切的愿望.2014年福建省中青年教师教育科研项目(社科A类)(JAS14023
Design and analysis on a Gires-Tournois resonator based interleaver

Characteristics of stable L-band SFS using dual-forward synchronous pumping technique
We investigate the characteristics of the dual-forward synchronously pumped L-band erbium-doped superfluorescent fiber source (SFS). The effects of pump ratio and fiber length arrangements on the output characteristics of the L-band SFS in terms of mean wavelength, spectral linewidth, and output power are analyzed. It is shown that the optimized pump ratio and fiber length arrangements provide broadening spectral linewidth and enhanced pumping efficiency, while the synchronous pump ensures stable mean wavelength operation. A new single-forward pumping scheme with a section of unpumped fiber is proposed to achieve a mean wavelength stable L-band SFS with a broadening linewidth of 50.4 nm and a pumping efficiency of 33.5%
Cloning and Expression of the Leukotoxin Gene SH from Fusobacterium necrophorum
坏死梭杆菌白细胞毒素是坏死杆菌病的主要致病因子,白细胞毒素基因(lkT)是其编码基因。以分离到的国内牛源坏死梭杆菌fn(A)菌株f4基因组dnA为模板,应用PCr方法扩增白细胞毒素基因SH片段,克隆至PMd18-T载体上,以bAMHⅠ和HIndⅢ酶切的目的片段SH与相应酶切的PET32A载体连接构建PET32A-SH重组表达质粒,经转化E COlI bl21(dE3)后用IPTg进行蛋白诱导,SdS-PAgE检测重组蛋白表达情况。结果表明:扩增基因序列大小为1800bP,SdS-PAgE检测重组蛋白有效表达,表达得到大小为80.2kdA的目的蛋白,采用镍柱亲和层析方法纯化SH重组蛋白,获得了纯度达95%的重组蛋白;经WEST-Ern-blOT证实,该蛋白对抗坏死杆菌阳性血清具有反应活性。The leukotoxin of Fusobacterium necrophorum(FN) is considered to be one of the main virulence factors.The lkt gene encodes for FN.In this study,the SH fragment of lkt gene was amplified by PCR using the F4 genome as the template,which was isolated from the Chinese Fusobacterium necrophorum strain.The fragment was then cloned to the pMD18-T vector for sequencing.Thereafter,the SH fragment was subcloned into the multiple cloning sites of the pET32 to construct pET32a-SH recombinant plasmid,which was then trans-formed into E.coli BL21(DE3) with IPTG induction for expression.SDS-PAGE was used to analyze the recombinant protein.The results showed that the SH fragment of about 1800 bp was amplified and was about 80.2 kDa.The fusion protein was purified by Ni-NTA affinity chromatography under denature conditions,and their purity was above 95%.Western-blot analysis indicated the SH fragment had anti-genicity against Fusobacterium necrophorum.“十五”国家科技攻关子课题(2002BA518A04);吉林省科技发展计划项目(20070570);吉林市科技发展计划项目(200805
Cloning and Expression of the Leukotoxin BSBSE Gene from Fusobacterium necrophorum
以牛源坏死梭杆菌FNn株为试材,根据GenBank上已发表的坏死梭杆菌AF312861标准菌株的lktA序列设计1对引物,利用PCR技术扩增出1 131 bp的坏死梭杆菌白细胞毒素BSBSE基因。将PCR产物插入pGEM-T Easy vector中,经双酶切鉴定正确后进行序列测定。分析表明该BSBSE序列与GenBank上已发表的坏死梭杆菌AF312861标准菌株的lktA序列的核苷酸同源性为99%,推导出的氨基酸序列同源性为98%。为研究BSBSE的免疫原性,构建了原核表达载体pMAL-p2X-BSBSE,用IPTG诱导在大肠杆菌中表达。结果表明,BSBSE基因在大肠杆菌中进行了高效特异性融合表达,融合蛋白分子量约为84.5×103,其中41.5×103为BS-BSE基因表达的蛋白质,43.0×103为MBP融合标签,Western-blotting检测表明该表达产物有免疫原性。According to the sequence of announced lktA gene in Fusobacterium necrophorum,a pair of primers were designed.The BSBSE gene was amplified by PCR.The product was cloned into pGEM-T Easy vector.When nucleotide sequence and deduced amino acid sequence were compared with homologous sequence of the FN AF312861 lktA of GenBank,the homologue of the mucleotide sequence is 99% and the homologue of the amino acid sequence is 98%.The BSBSE fragment was inserted into expression vector pMAL-p2X and the plasmid pMAL-p2X-BSBSE were expressed in E.coli BL21 by IPTG induction.The SDS-PAGE analysis indicated the weight of the fusion protein was about 84.5.0×103,which included the 41.5×103 protein expressed from BSBSE gene and 43.0×103 fusion MBP tag.The recombinant BSBSE-pMAL-p2X production has Immunogenicity with western-blotting.The cloning and expression of the BSBSE gene established the foundation of further research on the function and application of the BSBSE gene.“十五”国家科技攻关子课题(2002BA518A04);; 中国农业科学院特产研究所科研基金项目(Tcs2005-03
医院门诊药房全自动整盒发药设备使用优劣势分析
目的提高全自动整盒发药设备在门诊药房的使用效果,为国内其他医院引进该设备提供参考。方法结合智能化门诊药房的实际工作和全自动整盒发药设备的运行情况,评价该设备使用的优劣势。结果与结论全自动整盒发药设备的引进,改变了药师的工作环境,规范了药品的使用管理,但仍需要通过规范操作来减少某些弊端,同时加强管理,促进设备的高效运作,使其发挥最大优势
基于复合型耦合器的新型透射式GT交错复用器及色散补偿
提出了基于复合型耦合器的透射式GT腔,并将其串入马赫-曾德尔干涉仪(MZI)的两臂上,得到了一种新型的交错复用器。推导了该器件的理论表达式,并进行了数值模拟。模拟结果表明,该交错复用器具有相当好的通带平坦性,大的通带带宽、截止度和阻带带宽。对该交错复用器进行了优化分析,发现腔数固定时,纹波带宽比越大(频谱越接近矩形方波),隔离度将越小。分析结果表明,可以通过增加腔的数量来解决纹波带宽与隔离度的矛盾,即交错复用器的频谱性能随着腔数的增加得以改善。相对于环形谐振腔辅助的交错复用器,该交错复用器不需要添加额外的相移器就能获得近似矩形的频谱,并且在整个工作波段范围具有很好的频谱均匀性。对于该器件的色散问题,提出了采用级联环形腔作为色散补偿器的补偿方案。模拟结果表明,级联环形腔能够很好地补偿该交错复用器的色散
Biological characteristics of mesenchymal stem cells from bone marrow versus adipose tissue in rats
背景:除骨髓外,人们已从胎盘组织、脐带血肌肉组织、脂肪组织中分离到了间充质干细胞。目的:比较大鼠脂肪和骨髓间充质干细胞生物学特性和免疫调节功能的差异。方法:脂肪间充质干细胞和骨髓间充质干细胞分别来自bn大鼠的脂肪和骨髓。体外分离、纯化脂肪和骨髓来源间充质干细胞,进行细胞形态、表面标志、生长动力学、分化潜能鉴定;混合淋巴细胞反应比较两种细胞的免疫调节特性。结果与结论:脂肪间充质干细胞与骨髓间充质干细胞光镜和透射电镜下形态相似,第3代的脂肪间充质干细胞和骨髓间充质干细胞均高表达Cd29,Cd90,低表达Cd34,Cd45,Cd11b;第3,4,5代脂肪间充质干细胞增殖速度明显快于骨髓间充质干细胞;两者都具有低免疫原性,可以抑制异基因抗原引起的T淋巴细胞增殖,且这种抑制作用与细胞数目成正相关,等量脂肪间充质干细胞和骨髓间充质干细胞抑制作用差异无显著性意义。结果证实脂肪间充质干细胞具有和骨髓间充质干细胞同样的低免疫原性和免疫调节功能。BACKGROUND:Except bone marrow,people have collected mesenchymal stem cells (MSCs) from placenta tissue,cord blood muscle tissue and adipose tissue.OBJECTIVE:To compare the difference in biological characteristics and immunoloregulation of rat adipose-derived MSCs (ADSCs) and bone marrow MSCs (BMSCs).METHODS:ADSCs and BMSCs were obtained respectively from adipose and bone marrow of BN rats.ADSCs and BMSCs were isolated and purified in vitro to determine cell morphology,surface marker,growth kinetics and differentiation potential.Immunoloregulation of ADSCs and BMSCs was compared in mixed lymphocyte response RESULTS AND CONCLUSION:Morphology was similar between ADSCs and BMSCs under an optical microscope and transmission electron microscope.ADSCs and BMSCs at passage 3 highly expressed CD29 and CD90,weakly expressed CD34,CD45 and CD11b.The proliferation speed of ADSCs at passages 3,4 and 5 was obviously faster than BMSCs.Both ADSCs and BMSCs possessed low immunogenicity,could suppress allogenic antigen-induced T lymphocyte proliferation.Moreover,this inhibitory effect was positively associated with cell number.There were no significant differences in inhibitory effects of an equal volume of ADSCs and BMSCs.Results verified that ADSCs had the same low immunogenicity and immunoloregulation as BMSCs.卫生部科学研究基金——福建省卫生教育联合攻关计划资助(WKJ2005-2-020);厦门市科技计划社会发展项目(3502Z20064020)资助课题名称:外周血干细胞移植诱导肝移植术后免疫耐受的实验研究---
