Estimating Submarine Groundwater Discharge into the Jiaozhou Bay Using ~(226)Ra
海底地下水排泄(SGD)是全球水循环的一个组成部分,其输送的溶解物质不仅参与海洋的生物地球化学循环,而且影响近岸海域的生态环境。为了评估胶州湾海底地下水排泄状况,通过建立胶州湾内海水中226Ra的质量平衡模型来计算海底地下水排泄通量。胶州湾海水中226Ra的源主要有河流的输入、沉积物扩散输入和地下水的输入,海水系统在稳定状态下,这几种源应该与湾内海水和湾外海水的混合损失达到平衡。除了将地下水输入作为未知项外,对其他源和汇逐个进行量化,计算得知:2011年9—10月胶州湾的海底地下水排泄通量为7.85×10~6m~3·d~(-1);2012年4—5月胶州湾的海底地下水排泄通量为4.72×106m~3·d~(-1)。在此基础上,对地下水输入胶州湾的营养盐进行了评价。Submarine groundwater discharge( SGD) is a part of global water cycle,which delivers a large amount of solutes to the biogeochemical cycle and affects the coastal ecological environment. The ~(226)Ra mass balance model was established to estimate the submarine groundwater discharge to the Jiaozhou Bay. The ~(226)Ra sources include river transport,sediment diffusion and SGD,these sources should be in balance with the mixing loss of the bay water and open seawater when the seawater system is in the stable state. Except for the SGD was unknown item,other sources and sinks were quantified one by one. Calculated results showed that the SGD fluxes during September to October in 2011 was 7. 85 × 106m~3·d~(-1),April to May in 2012 was 4. 72 × 106m~3·d~(- 1). Based on the SGD fluxes,the SGD derived nutrient fluxes were estimated.国家自然科学基金项目(41072174)~
粗糙铂电极上甲酸吸附氧化的电化学原位表面增强拉曼光谱研究
采用循环伏安法和电化学原位表面增强拉曼光谱 (SERS)技术研究甲酸的解离吸附与氧化行为 .首次报道了甲酸吸附、解离和氧化的电化学原位SERS谱 ,发现甲酸在粗糙铂电极上能自发解离吸附 ;首次成功地获得了粗糙铂电极上甲酸吸附解离的强吸附中间体CO和活性中间体COOH的表面增强拉曼光谱 ,同时首次检测到甲酸氧化最终产物CO2 的拉曼光谱信号 ,从分子水平证实甲酸解离吸附反应的双途径机理
过氧化物还原酶3参与肾透明细胞癌发生与发展的分子机制
【目的】探讨肾透明细胞癌组织中硫氧还蛋白依赖的过氧化物还原酶3(PRDX3)的表达与肾透明细胞癌发生发展的关系。【方法】本研究首先在16例肾透明细胞癌组织及癌旁组织中验证了PRDX3的表达。根据表达量构建了786-O的PRDX3过表达细胞系及敲低表达细胞系,考察了PRDX3过表达和敲低表达后,肿瘤细胞的生长情况。通过pull-down试验联合LC-MS/MS技术寻找PRDX3的相互作用蛋白。初步探究了PRDX3与过氧化物酶1(PRDX1)的相互作用关系及PRDX3参与肾透明细胞癌发生发展的机制。【结果】16例肾透明细胞癌组织样本的western blot检测结果表明,有14例样本PRDX3下调表达,下调均值1.78倍左右。成功筛选到PRDX3过表达的786-O单克隆细胞系和对照细胞系[786O-PRDX(3+)和786O-PRDX(3-)]和PRDX3敲低表达的786-O单克隆细胞系和无意义序列对照细胞系(786O-PRDX3KN和786O-PRDX3NCi)。qPCR和western blot结果表明,相比786O-PRDX(3-)组,786O-PRDX(3+)组在mRNA水平过表达约2.1倍,在蛋白水平过表达约1.8倍;相比786O-PRDX3NCi组,786O-PRDX3KN组在mRNA水平低表达约0.48倍,在蛋白水平低表达约0.51倍。CCK-8试验表明,相比786O-PRDX(3-),786O-PRDX(3+)细胞生长速率明显降低。但敲低PRDX3的表达后,敲低组和其对照组细胞的生长速率却无显著性差异。Pull-down试验发现PRDX3可能与PRDX1存在相互作用。结合质谱分析及数据库检索,发现PRDX3通过二硫键与PRDX1结合并发挥作用,分别通过plink软件鉴定到二者可信的结合位点,初步探讨了PRDX3参与肾透明细胞癌发生发展的可能机制。【结论】PRDX3在肾透明细胞癌组织中下调表达,可能参与了肿瘤的发生发展。提高PRDX3的表达水平,能显著降低肿瘤细胞的生长速率。基于PRDX3蛋白,可能开发针对肾透明细胞癌的靶向药物
In-situ SERS study on the electro-oxidation with HCOOH on a roughened platinum electrode
The dissociative adsorption and oxidation behavior of HCOOH on Pt was investigated by cyclic voltammogram (CV) and in-situ surface enhanced Raman spectroscopy (SERS) techniques. The in-stiu SERS of HCOOH adsorption, dissociation and oxidation on rough Pt is reported. It is found that HCOOH can spontaneously dissociate. The surface Raman spectra of CO, the strongly adsorbed intermediate and COOH, the weakly adsorbed intermediate of the dissociative adsorption of HCOOH were successfully obtained for the first time. At the same time, the Raman spectra of the finally oxidized product CO2 of HCOOH was also firstly detected. SURVEY OF MIXED INFECTIONS OF FISH-BORNE TREMATODES IN PINYUAN COUNTY AND DNA(RAPD)ANALYSIS
目的 调查广东省平远县棘隙吸虫及其他鱼源性吸虫混合感染情况。方法 对鱼类宿主取鱼鳃和鱼肉等组织 ,镜检囊蚴 ;用水洗过筛沉淀法粪检虫卵 ,部分虫卵阳性动物解剖检查。采用DNA -RAPD技术 ,比较福建棘隙吸虫广东株与福建株。结果 查出华支睾吸虫 (Clonorchissinensis)、东方次睾吸虫 (Metorchisorientalis)、福建棘隙吸虫 (Echinochasmusfujianensis)、日本棘隙吸虫 (E japonicus)、抱茎棘隙吸虫 (E perfoliatus)和钩棘单睾吸虫 (Haplorchispumilio) 6种。第二中间宿主为麦穗鱼等 11种 ,感染率 48 3% (182 / 377) ,有 92 4%的阳性鱼混合感染 2种以上吸虫囊蚴。保虫宿主狗、猫感染率 79 2 % (19/ 2 4) ,每只阳性动物均有 2种以上吸虫混合感染。 15个引物在福建棘隙吸虫广东株与福建株两者共获DNA片段 185个 ,共享度 (F) =0 984,遗传距离 (D) =0 0 16。结论 当地为人兽共患的多种鱼源性吸虫混合感染区。形态学观察和分子生物学检测结果相印证 ,福建棘隙吸虫广东株与福建棘隙吸虫福建株为同一虫种。Aim To seek the possible existence of mixed infections of Echinochasmus fujianensis and other fish-borne tremtodes in Pinyuan county,Guangdong.Methods Metacercariae of trematodes were detected from gills,muscles and some other tissues of freshwater fish and examintd under microscopy Sedimentation pocedures were used to observe parasite ova in stool Adult worms were obtained from definitive animal hosts upon dissection Experimental animal infection were performed from metacercariae of E fujianensis strains in Fujian and Guangdong,and it's DNA analysis Random amplified Polymorphic DNA(RAPD).Results Six species of trematodes were found,including Clonorchis sinensis,Metorchis orientalis,E fujanensis, E japonicus,E perfoliatus and Haplorchis pumilio Eleven species of freshwater fish comprising Pseudorasbora parve were identified as the second intermediate hosts,with an average positive rate of 48 33%(182/377) The overall mixed infection animal were contracted with 2 or more species Dogs and cats were detected to be reservoir hosts,the total infection rate being 79 2%(19/24).92 4% of infecded fish were contracted with 2 or 6 speies of the metacercariae of trematodes 185 DNA fragments were obtained by 15 primers,F=0 894 and D=0 016 between E fujianensis Guandong and Fujian strain Conclusion There are six species of fish-borne trematodes which may cause infections in human and animal in the regions E fujianensis Guangdong stain and Fujian strain are the same species福建省科技计划基金!资助项目 (No 98-Z - 1 56);; 福建省“百千万人才”人选培养资
