SCREENING,IDENTIFICATION,AND ANTIMICROBIAL SPECTRUM of A MARINE BIOCONTROL BACTERIUM STRAIN LHB02
从高度污染的海滩污泥中,分离得到一株对部分水产病原菌有很强拮抗作用的生防细菌lHb02。采用细菌的通用引物进行lHb02的16SrrnA基因片段扩增,得到l511bP的dnA片段,其序列与枯草芽孢杆菌(bACIlluS SubTIlIS)在进化位置上最为接近,结合传统的形态学、生理生化鉴定手段,鉴定该菌株为枯草芽孢杆菌(bACIlluS SubTIlIS)。lHb02的无菌发酵液经硫酸铵沉淀,沉淀物透析除盐后所得的活性物质初提物初步鉴定为一种抗菌蛋白质。与水产主要病原菌进行拮抗试验,结果表明,lHb02对三种弧菌包括哈维氏弧菌(VIbrIOHArVEyI)、溶藻弧菌(V.AlgInOlyTICuS)和鳗弧菌(V.AnguIllAruM)有很强的抑制作用。The bacterium strain LHB02, which exhibited high inhibitory effects against a number of aquatic pathogens, was separated from marine sediments heavily polluted by aquaculture.Using a pair of universal primers, a fragment of LHB02’s 16S rRNA gene was amplified and a 1511bp of its DNA sequence was obtained.Based on its morphological, physiological and biochemical characteristics, LHB02 was identified as Bacillus subtilis.The active substance purified using ammonium sulfate precipitation and subsequent dialysis was identified as a crude antibacterial protein.In the following antimicrobial test, LHB02 showed strong antagonistic effects against three types of vibrios: Vibrio harveyi, V.alginolyticus and V.anguillarum.海洋公益性行业科研专项;201005032
白凤菜总黄酮对肝癌HepG2细胞生长、增殖和凋亡的影响
研究了白凤菜(Gynura formosana Kitam.)总黄酮(TFG)对肝癌HepG2细胞的生长、增殖和凋亡的影响.噻唑兰(MTT)实验结果表明TFG对HepG2细胞体外增殖的抑制作用有浓度和时间依赖效应:处理24h后130和260μg/mL TFG实验组的HepG2细胞凋亡率均显著升高,分别达到(47.00±1.31)%和(76.39±1.39)%(p<0.05);24h的半抑制质量浓度(IC50)为190.80μg/mL.实验组HepG2细胞周期阻滞在S期,细胞迁移率随TFG质量浓度的升高而下降,细胞内活性氧(ROS)水平最终显著下降至对照组的6.9%(p<0.05),细胞中Bax表达量略微上调而Bcl-2表达量明显下调.综上,TFG抑制肝癌HepG2细胞增殖和诱导HepG2细胞凋亡的机制可能与下调细胞ROS水平、重构细胞内还原体系、上调促凋亡蛋白Bax以及下调凋亡抑制蛋白Bcl-2表达相关.国家自然科学基金(31571896);;福建省教育厅科技B类项目(JB14209);;漳州卫生职业学院自然科学项目(ZWYZ201807
