    Detection of BCR-ABL fusion gene by multiplex RT-PCR combining with capillary electrophoresis

    目的建立多重逆转录-聚合酶链反应(rT-PCr)联合毛细管电泳法检测bCr-Abl融合基因,以期应用于慢性粒细胞白血病(CMl)辅助诊断。方法采用重叠延伸法构建bCr-Abl融合基因阳性模板,设计并优化检测bCr-Abl融合基因的多重rT-PCr联合毛细管电泳体系,对检测体系进行初步的性能评估。结果多重rT-PCr技术检测bCr-Abl融合基因(E1A2、E13A2和E14A2)的最低检测限在102~103拷贝/μl;50例临床确诊CMl的患者应用该法检测bCr-Abl融合基因的阳性率为86.0%,其中E13A2型(20.0%),E14A2型(66.0%);本方法与骨髓细胞培养染色体核型分析相比,阳性符合率为97.6%,阴性符合率为75.0%,总符合率为94.0%,2种方法具有较高的一致性(kAPPA=0.765,P>0.05)。结论成功建立检测bCr-Abl融合基因的多重rTPCr联合毛细管电泳方法。该方法操作简单快捷、特异性好、灵敏度高,适合于临床上CMl的辅助诊断和分子分型。Objective To establish a method for detecting BCR-ABL fusion gene by multiplex reverse transcription polymerase chain reaction( RT-PCR),and to provide a useful tool for chronic myeloid leukemia( CML) auxiliary diagnosis.Methods The construction of BCR-ABL fusion gene as positive template was made by overlap extension method.The method of multiplex RT-PCR combining with capillary electrophoresis was designed and optimized.Its primary performance was evaluated.Results The lower detection limit for BCR-ABL fusion gene( ela2,e13a2 and e14a2) by multiplex RT-PCR was 102-103 copies / μL.The positive rate of this method to detect 50 CML patients was 86.0%( e13a2: 20.0% and e14a2: 66.0%).Compared with the results of chromosome karyotype analysis,the positive coincidence rate was 97.6%,and the negative coincidence rate was 75.0%.The 2 methods have high consistency( Kappa = 0.765,P > 0.05).Conclusions The method of multiplex RT-PCR combining with capillary electrophoresis to detect BCR-ABL fusion gene is simple and rapid with good specificity and sensitivity and is very suitable for the clinical detection of CML.福建省自然科学基金计划资助项目(2012J01414); 厦门市科技局计划项目资助课题(3502Z20113013

Noether's theorem and its inverse theorem of Vacco systems with unilateral constraints are given,which are based upon the invariant properties by introducing the generalized quasi-symmetry of the infinitesimal transformation for the transformation group G r of r -parameter. And Noether's theorem dealing with the Vacco system of bilateral constraints is its corollary.     Performance of the fuel injector in supersonic combustor

    Enhancing the fuel-air mixing is critical for scramjet combustor performance. The performance of four different aerodynamic ramp injectors was reported in this paper. The experiments were conducted in a direct-connected scramjet test facility. The concentration profiles were obtained by gas sampling and chromatogram analysis. The pictures of the flow field were obtained by using laser scatter. It would be used to analyze the flow field generated by the aerodynamic ramp, then optimize the layout of the injectors.     Inflammation-related signaling pathways of differently-methylated DNA from benign and malignant thyroid tumors in one patient

    目的:研究同一患者体内甲状腺良性瘤与癌组织在dnA甲基化方面的差异,从而揭示dnA甲基化在肿瘤发生发展中的重要作用。方法:使用基因甲基化芯片技术对同一患者的并发甲状腺癌、甲状腺良性瘤组织样本的基因组甲基化水平进行检测,对筛选出的差异化显著的基因进行生物信息学分析得到功能基因的信号通路,并针对与甲状腺癌明显相关的信号通路进行分析讨论。结果:甲状腺的良性瘤与癌在dnA甲基化水平上存在显著差异;参与恶性肿瘤相关的信号通路包括T细胞受体的信号通路,JAk-STAT信号通路与细胞因子-细胞因子受体相互作用信号通路等炎症相关信号通路。结论:研究得出炎症的相关生物信息学通路与甲状腺肿瘤的发生发展密切相关,为甲状腺恶性肿瘤的发生发展机制的阐明与潜在药物靶点的选择提供新的思路与方向。Objective To determine the possible differences in DNA methylation of benign and malignant thyroid tumors and its role in tumorgenesis and tumor progression.Methods Infinium human methylation 450 bead chip platform was utilized to screen the whole-genome of benign and malignant thyroid tumor tissues surgically removed from a patient for the determination of their DNA methylation levels.The genes with significant differences in methylation were analyzed for their functional pathways with bioinformatics.Results There were significant differences in DNA methylation between benign and malignant thyroid tumor.The inflammation-related pathways, such as T cell, Jak-STAT and cytokine-cytokine receptor signaling pathways involved in the biological process of tumorgenesis and tumor progression.Conclusions The differences in DNA methylation between benign and malignant reveal a close relationship between inflammation and tumorgenesis and tumor progression and provide a novel method for investigation of thyroid cancer development and target drug discovery.国家自然科学基金(编号:81272445); 厦门市科技局国家对台科技合作项目(编号:350Z20121153

    Preliminary analysis of newly diagnosed acute myeloid leukemia from 2005 to 2012 in Zhongshan Hospital Xiamen University

    目的分析厦门大学附属中山医院近年来急性髓性白血病的发病情况与趋势,从中了解本地区急性髓性白血病的流行趋势,为制定科学的防治方法提供依据。方法回顾性分析2005~2012年厦门大学附属中山医院就诊的初发急性髓性白血病病例资料,统计分析年龄、性别、亚型及疗效的差异。结果新发急性髓性白血病患者的男女比例相当,无差异,25至59岁的中青年为高发病人群,其中35至39岁年龄段最高发,75到79岁出现老年人发病小高峰;M2、M3、M5是发病人数最多的三个亚型,且在未成年人、成年人、老年人不同年龄段组的发病比例有显著差异;2005~2012年新增患者人数具有上升趋势。结论 2005~2012年本院新发急性髓性白血病人数呈上升趋势,不同年龄段新发病例疗效差异显著可能与疾病亚型相关。Objective To explore the morbidity and trend of acute myeloid leukemia of Zhongshan Hospital Xiamen University in recent years, and to provide support for its prevention and intervention.Methods The retrospective analysis of acute myeloid leukemia patient of Zhongshan Hospital Xiamen University from 2005 to 2012 was made to analyze the differences among age, gender, subtype, and curative effect.Results The ratio of newly diagnosed acute myeloid leukemia between male and female was nearly same.The incidence of young and middle aged population between 25 to 59 years old was high.Among them, the highest incidence appeared in people aged 35 to 39.There was a small peak of incidence in old population aged 75 to 79.M2, M3, M5 were the most common subtypes of all patients, and there was a significant difference among age groups of minor, adult, and senior citizens.The newly diagnosed acute myeloid leukemia of each year was upward trend from 2005 to 2012.Conclusion The trend of newly diagnosed acute myeloid leukemia appeared upward.There was a significant difference of curative effects in newly diagnosed group of different ages may be caused by the specific subtype.福建省自然科学基金计划资助项目(2012J01414); 厦门市科技局计划项目资助课题(3502Z20113013); 福建省卫生厅青年课题(2013-2-92

    空间生命科学是随着人类空间探索活动,特别是载人空间探索而产生和发展的新兴交叉学科,它涵盖了较为广泛的研究范围.在过去半个多世纪,国际上在该领域取得了许多重要发现和研究成果,不仅支撑了载人空间探索任务,同时也服务了地球人类的生活.随着我国载人航天和深空探索活动的不断发展,特别是我国载人空间站工程的启动,未来20年将是我国空间生命科学发展的黄金时期.基于我国的载人空间站和返回式科学卫星实验平台,开展空间生命科学研究,获取新知识、创新新技术,进一步服务于人类空间探索活动、服务国家经济和社会发展,这就需要我们从学科发展的战略高度,系统深入地进行研究思考.在全面回顾国内外空间生命科学的发展历史及现状的基础上,本文对我国空间生命科学的战略需求、发展方向和关键科学问题等进行了梳理、分析和展望,以期为我国空间生命科学的发展提供思路和借鉴.As an emerging interdiscipline, space life science, generated and developed with human space, especially manned space exploration,, covering very wide research fields.In the past more than half a century, many significant findings and achievements have been achieved in this field, which not only supports the missions of manned space exploration, but also provides service for the life on the earth.With the continuous development of Chinese manned space and deep space exploration activities, especially the startup of Chinese manned space station project, the next two decades will be the prime time for the development of Chinese space life science.To conduct space life science, obtain new knowledge and innovative technology and provide further service for human space exploration, national economic and social development basing on Chinese manned space station and return type scientific satellite experiment platform, which need us to study and thinking systematically from the strategic perspective of subject development.By reviewing the historical and current development of space life science at home and abroad, this essay analyzes and prospects the strategic needs, key scientific problems and development direction of Chinese space life science, in order to provide inspiration and reference for its development.国家重点基础研究发展计划(编号:2011CB710903); 中国科学院技术科学部学科发展战略研究课题(我国空间科学发展战略研究)资助项

    Measurement of temperature, velocity and water vapor concentration in a scramjet combustor based on nearinfrared diode laser absorption

    A multi-channel Tunable Diode Laser Absorption Spectroscopy (TDLAS) system was designed and constructed for flow parameters diagnostics in a scramjet combustor. Two fiber coupled distributed feedback (DFB) lasers with narrow line width were used to probe two H2O absorption features (7185.597cm-1,7444.35cm-1+7444.37cm-1(c ombined)) by using direct absorption Time-Division-Multiplexing (TDM) strategy at a 4-kHz repetition rate. Laser light was split into five beams and transmitted across the test region. Two motorized precision translation stages were used to move the collimators during the test, so that the three beams located near the cavity and at the exit of the combustor can scan the cross sections respectively. Flow parameters could be obtained simultaneous which included average temperature, water vapor concentration and velocity at the entrance of the combustor, the distribution of temperature, water vapor concentration at a cross section near the cavity, the distribution of temperature, water vapor concentration and velocity at the exit cross section of the combustor. The parameters of the flow entering and exiting the combustor could be used to evaluate the performance of the direct-connected scramjet facility and the combustion efficiency of the combustor. The parameters at the cross section in the combustor could also be used to analysis combustion characteristics in the combustor

    径向变形量系数对谐波齿轮传动啮合特性的影响

    径向变形量系数是衡量柔轮弹性变形的重要参数之一,选取公切线双圆弧齿廓为谐波传动柔轮齿廓,基于谐波传动包络啮合理论,采用MATLAB编程计算,研究了径向变形量系数对共轭区域、共轭齿廓、啮合轨迹等性能的影响。研究结果表明,径向变形量系数对长轴与短轴附近轮齿的径向变形、长短轴过渡段轮齿的切向变形与法向转角影响较大;随着径向变形量系数减小,共轭区域增大,而共轭区域之间的空白区域减小,且共轭曲线一齿廓向上偏移,共轭曲线二齿廓向下偏移,存在有效共轭区域及有效共轭齿廓;合理选择径向变形量系数是避免谐波传动啮合干涉并提高啮合性能的重要方式之一
