    Label-free colorimetric detection of biological thiols based on target-triggered inhibition of photoinduced formation of AuNPs

    A label-free colorimetric method for the detection of biological thiols (biothiols) was developed.This method is based on prevention of the photoinduced reduction of auric ions (Au(III)) in thepresence of amino acids (acting as a reducing agent) by biothiols; the photoinduced reduction isinhibited due to the strong interaction of the biothiols with Au(III). In this method, the samplewas first incubated in an assay solution containing Au(III) and threonine; the sample solutionwas then exposed to 254 nm UV light. For samples without biothiols, this process led to thephotoreduction of Au(III) followed by growth of gold nanoparticles accompanied by the visuallydetectable development of a red coloration typified by an absorption peak at ca 530 nm.Conversely, in the presence of biothiols, reduction of Au(III) to Au(0) was prevented byentrapment of Au(III) within the biothiols via the thiol group. The solution thus remainedcolorless even after UV irradiation, which was used as an indicator of the presence of biothiols.Using this strategy, biothiols were very conveniently analyzed by monitoring color changes ofthe samples with the naked eye or a UV–vis spectrometer. The strategy based on this interestingphenomenon exhibited high selectivity toward biothiols over common amino acids and wassuccessfully employed for reliable quantification of biothiols present in human plasma,demonstrating its great potential for clinical applications.

    본 발명은 폐암 검출용 동물 바이오센서를 이용한 냄새 변별 훈련장치 및 그 용도에 관한 것으로, 더욱 상세하게는 이동공간이 구비되어 있는 상기 훈련장치에서 동물 바이오센서인 탐지쥐가 표적물인 폐암 환자의 호흡에서 나오는 특정 표적 가스를 지각하고 특정한 신호행동을 보이도록 훈련시킴으로써 동물 그 자체를 질병 진단에 사용할 수 있는 비침습적 폐암 검출용 냄새 변별 훈련장치 및 그 용도에 관한 것이다. 본 발명에 따른 폐암 검출용 동물 바이오센서를 이용한 냄새 변별 훈련장치는 암 진단에 제한되지 않고, 결핵, 식중독균을 포함하는 질병의 진단 및 환경 오염물질 모니터링 등에 유용하게 이용할 수 있다.국

    학위논문(석사) - 한국과학기술원 : 생명화학공학과, 2002.8, [ ix, 60 p. ]Physiological and environmental characteristics of succinate-producing Actinobacillus succinogenes sp. 130Z were studied to develop the efficient biological system for succinic acid production. High cell density culture system by external membrane recycle bioreactor was also developed. In high-cell concentration cultivation of Actinobacillus succinogenes, volumetric productivity of succinic acid reached over 6.5 g/L/h. Several factors which affect cell growth and succinic acid production were examined by 100 ml flask and 1L batch cultures. Specific growth rates in the flask cultivation were almost same with increasing glucose concentration up to 40 g/L glucose, but slightly decreased when 80 g/L glucose was used. Cell growth and succinic acid production were also affected by initial pH in flask cultivation. The range between pH 7 and pH 7.5 was the best condition on a view of cell growth and succinic acid production. But the succinic acid yield between pH 6 and pH 6.5 showed higher results than the data between pH 7 and pH 7.5. Other carbon sources including lactose, sucrose, maltose, fructose, mannitol and mannose could be utilized as alternatives to glucose. Among the eight different complex nitrogen sources, only soytone could be utilized for succinic acid production. Magnesium carbonate in the medium could increase succinic acid production. In 1 L batch cultures, volumetric productivity of succinic acid increased when more yeast extract was used. With 60 g/L glucose and 5 g/L yeast extract, cells grew up to OD660OD_660 (optical density) of 3.5 and succinic acid productivity reached 0.46 g/L/h. When 60 g/L glucose and 25 g/L yeast extract were used, cell concentration was over OD660OD_660 of 9 and succinic acid productivity reached 1.34 g/L/h. Cell concentration and the amount of succinic acid attainable were not dependent on the concentration of phosphorous sources in the medium. Effects of external supply of H2H_2 were examined.
