Effect of Ginseng Saponin on the Circadian Rhythms in Rabbit and Mouse

Changes in circadian rhythms of the body temperature, the circulating leukocytes level and the pentobarbital induced sleeping times were observed in rabbit and mouse treated with ginseng saponin. The animals had been maintained for one week under the artificial illumination extending from 06 : 00 to 18 : 00 hours alternating with 12 hours of darkness, And thereafter the lighting schedule was completely reversed. Rabbits kept on a routine lighting regimen showed" a circadian variation in temperature characterized by a highest temperature at 14 : 00 hours and a noctural ' fall to low level. On reversing the light regimen the routine temperature rhythm adapted to the new lighting schedule by 3 days after the reversal and it was not affected by ginseng saponin. The highest level of the circulating total leukocytes, neutrophiles and lymphocytes in rabbit were reached in the afternoon followed by a evening fall. On reversal of the lighting schedule, however, the routine' circadian variations of the leukocytes were not adapted to the reversed lighting regimen until 7 days. All,' the circadian variations in circulating leukocytes levels' were also not affected by ginseng saponin in any case. Rabbit showed a minimum value in circulating. eosinophiles at 08:00 hours and an evening rise. When the lighting schedule was reversed, the adaptation of the routine eosinophile rhythm took 3 days" to occur. The occurrence of the adaptation was similar in fashion both in control and in ginseng saponin. treated animals. In male DDO mice maintained on a routine day and night lighting regimen, the longest duration of sleep induced by pentobarbital occured at 14:00 hours' and the shortest occurred between 22:00 and 02:00 hours. Studies on the cardiotonic effect of Aconiti tuber

The positive inotropic and the hemodynamic effects of Aconiti tuber butanol fraction were studied in excised rat auricle and in intact anesthetized rabbit. In contrast with digitalis cardiac glycoside, Aconiti tuber butanol fraction showed strong positive inotropic effect on rat auricle, which was not influenced by varying concentrations of potassium in Krebs solution. High calcium concentrations in the nutrient medium reduced the positive inotropic effect of Aco.. niti tuber butanol fraction, while low calcium shoo wed to increase the positive inotropic action. This synergistic effect of calcium with Aconiti tuber butanol fraction was similar to that of digitalis glyco.. side with calcium in a digitalis susceptible cardiac muscle preparation. In the hemodynamics of anesthetized rabbit heart rate, left ventricular diastolic pressure and systemic arterial pressure were slightly reduced by Aconiti tuber butanol fraction. The clinical efficacy of Vibramycin Precon PT-122 M on various infections in OB-GYN was studied. The aims of this study are determination of local and systemic tolerance of PT-122M intravenous administration and also determination of side effect as well as the clinical efficacy. 1. The over-all efficacy of this intravenous formulation of Vibramycin was good and satisfactory in 92% of patients studied. 2. Most of patients(82%) were considered to be tolerated to this intravenous Vibramycin. 3. Several side effects were observed. but in most patients the disease process and treatment were not influenced by side effects, except six patients in whom the treatment had to be discontinued

Effect of Propranolol on the Ca++-regulation of Cardiac Mitochondria

Propranolol is one of clinically useful antiarrhythmic agents and elcetrophysiologically classified as group II. And the negative inotropic effect which is not related to adrenolytic effect has been demonstrated with high concentration of propranolo l. On the other hand , it has been well known that the calcium plays a central role in excitation-contraction coupling process of myocardium and also in electrophysiological changes of cell membrane. Authors studied the effect of propranolol on calcium uptake and release in mitochondria prepared from porcine myocardium to investigate the mechanism of action of propranolol on myocardium. The results are summerized as follows; 1. The maximum calcium uptake of mitochondria was 50.30 nmoles per mg protein with the rate of 0.65 nmole/mg/sec. and the half-maximum velocity of Ca서-uptake was achieved at about 11. 67μM Caκ 2. The calcium uptake of mitochondria was noncompetitively inhibited by propranolol with dose dependent manner (Ki; 6.2 x lO-'M) . 3. The calcium efflux from the internal pool of mitochondria was rapid by the treatment of 50μM dinitrophenol (9.60 nmole/mg/min.) , andit was not affected by propranolo l. 4. The rate of sodium-induced calcium efflux appeared to be a function of [Na+]' and about 8.80 mM Na+ was required to elicit half-maximum velocity of calcium efflux; The maximum velocity was 7.43 nmole/mg protein per minute 5. The mitochondrial NaηCa얘 exchanges were noncompetitively inhibited by propranolol with dosedependent manner (Ki; 2.91 x lO-'M) .
