Studies on the Subcellular Localization of Peroxiredoxin V and its Stability and Antioxidant Function in Mitochondria and the Function of Peroxiredoxin I in the Kidney of Cisplatin-injected Mice
Peroxiredoxins (Prxs) are a family of peroxidases that reduces peroxides such as hydrogen peroxide with reactive cysteine residues in their active sites. Mammalian cells express six isoforms of Prx (Prx I to VI). These isoforms vary in subcellular localization, with Prx I, II, and VI being localized mainly in the cytosol, Prx III being restricted to mitochondria, and Prx IV being found predominantly in the endoplasmic reticulum (ER). Prx V gene contains two ATG initiation codons, giving a short (SPrx V) and long form of Prx V (LPrx V), which are proposed to be in cytosol, nucleus, mitochondria, and peroxisomes.
In the first part of this study, I investigated the submitochondrial localization of Prx V and the effect of oxidative stress on the stability of mitochondrial Prx V. Mouse embryonic fibroblasts derived from Prx V knockout (KO) mice were used extensively in this study. I found the presence of Prx V in the intermembrane space (IMS) as well as the matrix of mitochondria. Unlike other Prxs (Prx I-IV) that easily undergo inactivation through the hyperoxidation of their active site cysteine, Prx V was resistant to hyperoxidation. In the cells lacking Prx V, hyperoxidation of Prx III was increased significantly in response to various oxidative stresses, though its concentration is much less than that of Prx III in the organelle, attesting the peroxidase role of Prx V in mitochondria.
LPrx V is expressed as a precursor protein containing an 49-residue N-terminal mitochondria targeting sequence, of which the positively charged, N-terminal 41-residue sequence is cleaved by mitochondrial processing peptidase (MPP) located in the matrix, producing an intermediate Prx V (Octa-Prx V) with an 8-residue hydrophobic sequence. Evidence suggests that the 8-residue peptide is finally cleaved by mitochondrial intermediate peptidase (MIP) to produce mature Prx V. Given that MIP is sensitive to oxidative inactivation, Octa-Prx V accumulates temporarily in cells under oxidative stress as seen in cells lacking Prx III or Srx. Octa-Prx V with destabilizing amino acid residues at its N-terminus is degraded readily according to the N-end rule pathway of protein degradation, thus resulting in the decrease in the amount of Prx V in oxidatively stressed cells.
In the second part, I investigated the potential role of Prx I against cisplatin-induced renal damage in mice. The anticancer drug cis-diamminedichloroplatinum (cisplatin) is a chemotherapeutic agent widely used for solid tumors. Nephrotoxicity of cisplatin is a serious dose-limiting side effect. Oxidative stress is a major cause of cisplatin-induced nephrotoxicity through damaging DNA, membranes and mtiochondria and increasing ER stress. Cisplatin is known to produce reactive oxygen species through Cytochrome P450 2E1 (CYP2E1) and localize at the surface of the ER where CYP2E1 is located.
I found that among six Prx isoforms Prx I is selectively degraded in cisplatin-treated kidney around the time when renal functions are severely damaged. The Prx I degradation was blocked in mouse proximal tubular cells treated with 3-methyladenine, an autophagy inhibitor or in mouse embryonic fibroblast (MEF) cells lacking ATG7. Moreover, increased ROS on ER surface by overexpression of CYP2E1 further accelerated Prx I degradation. These results suggests that Prx I degradation is mediated largely through autophagy that is promoted by cisplatin-induced ER stress. Ablation of Prx I attenuated cisplatin-induced nephrotoxicity and significantly increased the abundance of markers for oxidative stress, ER stress and inflammation in kidney, indicating Prx I palys a protective role against cisplatin-induced nephrotoxicity.;Peroxiredoxins (Prxs)는 활성 부위의 반응성이 높은 cysteine 잔기를 이용하여 hydrogen peroxide와 같은 과산화물질을 환원시키는 peroxidase이다. 포유류에는 여섯 종류의 Prx (Prx I~VI)가 있다. 이 isoform들은 세포 내에서의 위치가 모두 다른데, Prx I, II, VI는 주로 세포 기질에 존재하고 Prx III는 미토콘드리아에만 존재하며 Prx IV는 주로 ER에서 발견되었다. Prx V 유전자는 두 개의 ATG 개시 코돈을 포함하고 있어서 짧고 (SPrx V) 긴 (LPrx V) 형태의 Prx V를 만든다. 그로 인해 Prx V는 세포 기질, 핵, 퍼옥시좀과 미토콘드리아에 존재하는 것으로 알려져 있다.
이 연구의 첫번째 파트에서 Prx V의 미토콘드리아 내부 위치와 산화적 스트레스가 미토콘드리아 Prx V의 안정성에 미치는 영향에 대해 연구하였다. Prx V 결핍 mouse에서 얻은 mouse embryonic fibroblast가 이 연구에서 사용되었다. Prx V가 미토콘드리아 내부에서 matrix 뿐만 아니라 intermembrane space (IMS)에도 존재하는 것을 발견하였다. 활성 부위의 cysteine이 쉽게 과산화되어 비활성화되는 다른 Prxs (Prx I~IV)와 다르게, Prx V는 쉽게 과산화되지 않는다. Prx V가 없는 세포에서 다양한 산화 스트레스에 의해 미토콘드리아에 있는 Prx III의 과산화가 증가하는 것을 보았다. Prx V가 미토콘드리아 내부에서 Prx III에 비해 훨씬 양이 적은 것을 고려했을 때, Prx V가 미토콘드리아에서 중요한 peroxidase로 작용한다는 것을 보여준다.
LPrx V는 positive charge를 띄는 49개 잔기의 아미노 말단 미토콘드리아 표적 서열을 갖는 precursor protein이다. 이 미토콘드리아 표적 서열의 41개 잔기는 matrix에 존재하는 mitochondrial processing peptidase (MPP)에 의해 잘리고, 8개의 소수성 서열을 갖는 intermediate Prx V (Octa-Prx V)가 만들어진다. 여러 증거들을 통해 이 8개의 펩티드는 결국 mitochondrial intermediate peptidase (MIP)에 의해 제거되어 완전히 성숙한 Prx V가 생성되는 것으로 보인다. MIP가 산화적 비활성화에 민감하다는 것을 고려했을 때, 미토콘드리아 특이적인 Prx인 Prx III나 과산화되어 비활성화된 Prx를 재활성화하는 효소인 sulfiredoxin (Srx)가 없는 세포에서 Octa-Prx V는 산화적 스트레스에 의해 세포 내에 일시적으로 축적된다. 아미노 말단에 destabilizing 아미노산 잔기를 갖고 있는 Octa-Prx V는 단백질 분해 과정중 N-end rule pathway에 의해 빠르게 분해되고, 따라서 산화적으로 스트레스를 받은 세포 내에서 Prx V의 양이 줄어들게 된다.
이 연구의 두번째 파트에서 Prx I의 mouse에서의 cisplatin 유도 신장 독성에 대한 잠재적인 역할에 대해 연구했다. 항암제인 cis-diammine dichloroplatinum (cisplatin)은 다양한 solid tumor에 사용되는 화학요법제이다. Cisplatin의 신장 독성은 용량을 제한하는 심각한 부작용의 하나이다. 산화적 스트레스는 cisplatin에 의해 유도된 신장 독성에서 중요한 요인으로 DNA, 세포막, 미토콘드리아에 손상을 입히고 ER stress를 증가시킨다. Cisplatin은 Cytochrome P450 (CYP2E1)을 통해서 활성 산소종을 만드는 것으로 알려져 있고 CYP2E1이 위치한 ER 표면에서도 발견된다.
여섯 종류의 Prx isoform 중에서 Prx I이 cisplatin을 처리한 신장에서 신장기능이 심각하게 손상을 받을 때 선택적으로 분해되는 것을 보았다. Prx I의 분해는 마우스의 근위세뇨관 세포에서 autophagy의 저해제인 3-methyladenine을 처리했을 경우와, autophagy가 일어나지 않는 ATG7 KO MEF 세포에서 저해되었다. 게다가, CYP2E1을 과발현시켜서 ER 표면에서 활성산소 생성을 증가시켰을 때 Prx I의 분해가 촉진되었다. 이러한 결과는 Prx I의 분해가 많은 부분 cisplatin에 의한 ER stress에 의해 유도된 autophagy에 의해 일어난다는 것을 보여준다. The Relationship among the partly family violence by adolescents, their Dissociation and their School Adjustments.
The purpose of this study was to investigate the mediating effects of dissociation between family violence and school adjustment. The subjects in this study were 292 adolescents at middle schools in Kyunggido. A violence scale in CTS(Conflict Tactics Scale), A-DES(Adolescent Dissociative Experiences Scale) and School Adjustment Scale were used to measure. The data were analyzed by frequency, Pearson's correlation and regression using SPSS. Lastly, Baron and Kenny's(1986) model was used to verify mediational effects. The followings are results of the study.
First, the results of adolescent's family violence were perceptual mild degree.
Second, there was a significant correlation of family violence, dissociation and school adjustment. Family violence and dissociation were significant positive correlation, family violence and school adjustment were significant negative correlation. Dissociation and school adjustment were significant negative correlation.
Third, dissociation mediated the relationship between family violence and school adjustment.
These results contribute to the understanding about the effects of the dissociation between family violence and school adjustment. Additionally, it was confirmed the mediating effects of dissociation between family violence and school adjustments. Given the consistent findings to dissociation, more research is needed to better understand effective intervention strategies to effectively family violence and school adjustments.;해리는 외상 적 사건에 대한 신체적 탈출이 불가능할 때 심리적으로 도망하는 것을 의미하는 것으로서 일생에 한번 이상 경험할 수 있는 것이다. 외상을 경험한 후 자신을 보호하기 위해 방어하려는 수단으로 사용하는 것은 적응적이지만 지속적으로 해리에 의존하는 것은 부적응적이다. 가정폭력적인 환경에서 청소년들이 해리 경험을 통해 대처하였음을 검증하였고 많은 시간을 보내는 학교에서도 해리하고 있는 것을 알 수 있었다.
본 연구는 청소년의 직·간접 가정폭력 경험이 학교적응에 있어서 해리의 매개효과를 검증하고자 하였다.
경기도 S시에 위치한 D 중학교 292명을 대상으로 가정폭력수준 검사, 해리수준 검사, 학교적응 검사를 사용하여 조사하였다. 가정 폭력을 어느 정도 경험하는지를 분석하기 위해 빈도분석을 하였고 가정 폭력과 해리 및 학교 적응간의 관계를 알아보기 위해 상관분석을 실시하였다. 그리고 청소년이 지각한 가정폭력과 학교적응과의 관계에서 해리의 매개효과를 알아보기 위하여 Baron과 Kenny(1986)가 제안한 매개효과 검증 절차를 사용하였다.
본 연구 결과를 요약하면 다음과 같다.
첫째, 청소년의 직·간접 가정폭력 경험을 분석한 결과 많은 청소년들이 가벼운 수준에서 가정폭력을 경험한다고 지각하고 있다. 특히 일부 청소년들의 경우 언어폭력에서는 가벼운 수준뿐만 아니라 심한 수준으로까지 지각하고 있는 것으로 나타났다.
둘째, 가정폭력과 해리 및 학교 적응간의 관계를 살펴보았을 때 가정 폭력과 해리는 정적 상관관계를 보였으며 가정 폭력과 학교 적응에서는 부적 상관관계를 보였다. 그리고 해리와 학교 적응에서도 부적 상관관계를 보였다.
셋째, 가정 폭력과 학교적응과의 관계에서 해리의 매개효과를 검증한 결과 해리가 유의미한 매개변인 역할을 하는 것으로 검증 되었다. 해리를 통제했음에도 불구하고 가정 폭력과 학교 적응에 미치는 직접 효과가 여전히 유의미해 가정 폭력과 학교 적응간의 관계에서 해리는 부분 매개를 한다는 결과를 얻었다.
본 연구는 가정 폭력 경험으로 해리되어 학교 적응에 어려움을 주는 특성을 명확하게 이해하는데 도움을 주었고 상담 및 심리 치료 분야에서 해리의 특성을 좀 더 살펴 볼 수 있었다는 점에서도 의의가 있었다.Ⅰ. 서론 = 1

Purpose The dynamics of serum alpha-fetoprotein (AFP) level have been found to be a useful predictor of therapeutic responsiveness
in patients with hepatocellular carcinoma (HCC). We evaluated whether AFP changes were able to accurately reflect
imaging-based responses and predict prognosis in patients receiving therapies including immune-checkpoint inhibitors (ICIs).
Methods A total of 108 HCC patients with baseline serum AFP ≥ 20 ng/mL who received ICI-based treatment were included.
We evaluated AFP-based responses, coupled with radiographic responses by RECIST, at 6?10 (time-point 1, TP1) and
14?18 weeks (time-point 2, TP2) of therapy in terms of the change of AFP from baseline, with a > 20% decrease or increase
in level corresponding to the AFP response and progression, respectively. We examined the correlations between AFP and
imaging-based responses, and the prognostic implications of the AFP-based measure.
Results Based on AFP change, there were 24 and 20 responders and 74 and 24 progressors at TP1 and TP2, respectively.
The AFP responders yielded radiological objective responses in 90.9% (10/11) and 93.8% (15/16) of the cases at TP1 and
TP2, respectively, compared with only 1.4% and none, respectively, of the AFP progressors at the corresponding times. The
agreement between progression by RECIST and increased AFP level at the two time-points was 93.8% and 95.0%, respectively.
The accuracy of the AFP-based criterion for predicting radiologic response/progression was comparable at TP1 and
TP2. Both “AFP responder” and “AFP progressor” at TP1 or TP2 independently predicted the overall survival of patients
(adjusted hazard ratios [95% confidence intervals], 0.360 [0.174?0.743] and 0.315 [0.117?0.850]; and 2.525 [1.362?4.679]
and 3.908 [1.563?9.769], respectively).

Meta-Analysis of the Accuracy of Abbreviated Magnetic Resonance Imaging for Hepatocellular Carcinoma Surveillance: Non-Contrast versus Hepatobiliary Phase-Abbreviated Magnetic Resonance Imaging
Simple Summary Ultrasonography is recommended as a standard surveillance modality, but the performance of surveillance ultrasound for detecting early-stage hepatocellular carcinoma (HCC) is limited. Motivated to provide a more sensitive method, abbreviated magnetic resonance imaging (AMRI) protocols have been introduced for HCC surveillance. We aimed to systematically determine the diagnostic performance of surveillance AMRI for detecting HCC. This meta-analysis of 10 studies comprising 1547 patients found that the pooled sensitivity and specificity of surveillance AMRI for detecting HCC were 86% and 96%, respectively. Hepatobiliary phase contrast-enhanced AMRI showed significantly higher sensitivities for detecting HCC than non-contrast AMRI (87% vs. 82%), but significantly lower specificities (93% vs. 98%). Therefore, surveillance AMRI had overall good diagnostic performance for detecting HCC and might be clinically useful for HCC surveillance. In addition, AMRI protocol should be selected with consideration of the advantages and disadvantages of each protocol. We aimed to determine the performance of surveillance abbreviated magnetic resonance imaging (AMRI) for detecting hepatocellular carcinoma (HCC), and to compare the performance of surveillance AMRI according to different protocols. Original research studies reporting the performance of surveillance AMRI for the detection of HCC were identified in MEDLINE, EMBASE, and Cochrane databases. The pooled sensitivity and specificity of surveillance AMRI were calculated using a hierarchical model. The pooled sensitivity and specificity of contrast-enhanced hepatobiliary phase (HBP)-AMRI and non-contrast (NC)-AMRI were calculated and compared using bivariate meta-regression. Ten studies, including 1547 patients, reported the accuracy of surveillance AMRI. The pooled sensitivity and specificity of surveillance AMRI for detecting any-stage HCC were 86% (95% confidence interval (CI), 80-90%; I-2 = 0%) and 96% (95% CI, 93-98%; I-2 = 80.5%), respectively. HBP-AMRI showed a significantly higher sensitivity for detecting HCC than NC-AMRI (87% vs. 82%), but significantly lower specificity (93% vs. 98%) (p = 0.03). Study quality and MRI magnet field strength were factors significantly associated with study heterogeneity (p <= 0.01). In conclusion, surveillance AMRI showed good overall diagnostic performance for detecting HCC. HBP-AMRI had significantly higher sensitivity for detecting HCC than NC-AMRI, but lower specificity

Treatment-naive patients with preserved hepatic reserve (Child?Pugh score ≤ 7) who received TACE plus sorafenib (n = 91) or TACE alone (n = 109) for locally advanced HCC with macrovascular invasion were retrospectively evaluated. Propensity score matching (PSM) was used to correct selection bias, and 63 pairs were created. In the entire study population, the median progression-free survival (PFS) and overall survival (OS) with TACE plus sorafenib were better than those with TACE alone. After PSM, the median PFS (7.0 vs. 4.3 months; p = 0.017) and OS (17.5 vs. 12.8 months; p = 0.049) were again significantly longer with TACE plus sorafenib than with TACE alone. Stratified Cox regression analysis and doubly robust estimation revealed that treatment type was significantly associated with both PFS and OS. In the subgroup analysis, TACE plus sorafenib did not show a significant survival benefit for patients with main portal vein or inferior vena cava invasion. Major complications were similar in both groups (p = 0.330). In conclusion, TACE plus sorafenib showed better survival outcomes than TACE alone in patients with locally advanced HCC
