A Conceptual Model for Quality of Life Among People with Type 2 Diabetes in the Philippines
[Background] The increased prevalence of chronic diseases is a social issue in developing countries. [Methods] To create a conceptual model representing the quality of life of low-income people with type 2 diabetes in the Philippines, 117 low-income adult participants in a public support group were studied, and the Physical Component Summary (PCS) and Mental Component Summary (MCS) scores on quality of life (QOL) were analyzed in terms of their relationships with the following factors: basic attributes, physical factors (glycated hemoglobin level, complications, comorbidities, and symptoms), social factors (support, education, and financial status), and cognitive factors (knowledge, attitude, self-efficacy, and self-management behavior). [Results] Based on correlation coefficients among variables and goodness-of-fit test results through a path analysis, 2 models representing causal relationships were created, both of which showed sufficient goodness-of-fit. [Conclusion] Glycated hemoglobin levels, knowledge of blood glucose levels, self-efficacy, and self-management behavior (exercise) influenced PCS scores, while age, glycated hemoglobin levels, neuropathy, knowledge of insulin, self-efficacy, and self-management behavior (exercise) influenced MCS scores. The influence of self-efficacy was prominent in both cases, providing an important insight for healthcare professionals to develop effective support methods.journal articl
Report of an Attempt to Visualize the Value of the Starry Sky and Potential as a Regional Resource
Under the activity of “Hoshinochi lab" aiming for collaboration across barrier between different organizations, we studied the possibility of “starry sky" as a regional resource creating a new added value. The purpose of the study is to make a proposal that leads to the improvement of regional vitality for the next generation based on the effects observed by visualization of the value of starry sky. Here, "visualization" has a various meaning from physical measurement of value of starry sky based on night sky brightness to activities making people conscious the wide variety of value of starry sky. Solution Structure of Ternary Complex of Berberine Bound to a dGMP–Fill-In Vacancy G‑Quadruplex Formed in the PDGFR‑β Promoter
The G-quadruplexes (G4s) formed in
the PDGFR-β gene promoter are transcriptional
modulators and amenable to small-molecule
targeting. Berberine (BER), a clinically important natural isoquinoline
alkaloid, has gained increasing attention due to its potential as
anticancer drug. We previously showed that the PDGFR-β gene promoter forms a unique vacancy G4 (vG4) that can be filled
in and stabilized by guanine metabolites, such as dGMP. Herein, we
report the high-resolution NMR structure of a ternary complex of berberine
bound to the dGMP-fill-in PDGFR-β vG4 in potassium solution.
This is the first small-molecule complex structure of a fill-in vG4.
This ternary complex has a 2:1:1 binding stoichiometry with a berberine
molecule bound at each the 5′- and 3′-end of the 5′-dGMP-fill-in
PDGFR-β vG4. Each berberine recruits the adjacent adenine residue
from the 5′- or 3′-flanking sequence to form a “quasi-triad
plane” that covers the external G-tetrad of the fill-in vG4,
respectively. Significantly, berberine covers and stabilizes the fill-in
dGMP. The binding of berberine involves both π-stacking and
electrostatic interactions, and the fill-in dGMP is covered and well-protected
by berberine. The NMR structure can guide rational design of berberine
analogues that target the PDGFR-β vG4 or dGMP-fill-in vG4. Moreover,
our structure provides a molecular basis for designing small-molecule
guanine conjugates to target vG4s
Thermodynamic Stability and Folding Kinetics of the Major G-Quadruplex and Its Loop Isomers Formed in the Nuclease Hypersensitive Element in the Human c-Myc Promoter: Effect of Loops and Flanking Segments on the Stability of Parallel-Stranded Intramolecular G-Quadruplexes
Overexpression of the c-Myc proto-oncogene is associated with a broad spectrum of human cancers. Nuclease hypersensitivity element III1 (NHE III1) of the c-Myc promoter can form transcriptionally active and silenced forms, and the formation of DNA G-quadruplex structures has been shown to be critical for c-Myc transcriptional silencing. The major G-quadruplex formed in c-Myc NHE III1 is a mixture of four loop isomers, which have all been shown to be biologically relevant to c-Myc transcriptional control. In this study, we performed a thorough thermodynamic and kinetic study of the four c-Myc loop isomers in a K+ solution. The four loop isomers all form parallel-stranded G-quadruplexes with short loop lengths. While the parallel-stranded G-quadruplex has been known to favor short loop lengths, our results show that the difference in thermodynamic and kinetic properties of the four loop isomers, and hence between the parallel G-quadruplexes with similar loop lengths, is more significant than previously recognized. At 20 mM K+, the average difference in the Tm values between the most stable loop isomer 14/23 and the least stable loop isomer 11/20 is more than 10 °C. In addition, the capping structures formed by the extended flanking segments are shown to contribute to a stabilization of 2−3 °C in Tm for the c-Myc promoter G-quadruplex. Understanding the intrinsic thermodynamic stability and kinetic properties of the c-Myc G-quadruplex loop isomers can aid in our understanding of their biological roles and drug targeting
