    分割錯視に関する実験的研究(II)―図形の大きさ及び配置を変化させた場合―

    1970-03本報告では、まず描画図形によって分割錯視図形を呈示し、線分数、線分の長さ、照明の明るさ等を変化させて、それらが錯視量に及ぼす影響を調べた。また、タキストコープによって光点図形として分割錯視図形を呈示し、点の数の変化及びそれらの配置の効果について、実験方法をこれまでの調整法から極限法に変えて測定してみた。さらに、分割錯視配置から三角形配置への移行を3光点を用いて行ない、それが2点間の距離の知覚にどのような影響を与えるかを検討した。結果は以下のごとくである。(1)線分の長さ、照明の明るさ等を変化させても、線分数の変化に伴う錯視量の変化の傾向に大きな違いは見られず、9本線分附近で錯視量の極大値を持つ山型の変化を示した。(2)両端2点間の距離がこれまでの半分にされたことにより、最大の錯視量を持つ点の数が減少した。しかし、描画図形と光図形との違いや実験方法の違いによる影響はほとんど認められなかった。(3)4光点図形を不均等分割に配置していくにつれて、両端2点間の距離の過大視量が一定の割合で減少した。(4)3光点から成る分割錯視図形の分割点を垂直上方向に移動するにつれて、図形は三角形として見られるようになり、それに従って両端2点間の距離の過大視量が減少し、正三角形配置では、過大視はほとんど消失した。departmental bulletin pape

    Crystal Structures of SecYEG in Lipidic Cubic Phase Elucidate a Precise Resting and a Peptide-Bound State

    The bacterial SecYEG translocon functions as a conserved protein-conducting channel. Conformational transitions of SecYEG allow protein translocation across the membrane without perturbation of membrane permeability. Here, we report the crystal structures of intact SecYEG at 2.7-A resolution and of peptide-bound SecYEG at 3.6-A resolution. The higher-resolution structure revealed that the cytoplasmic loop of SecG covers the hourglass-shaped channel, which was confirmed to also occur in the membrane by disulfide bond formation analysis and molecular dynamics simulation. The cytoplasmic loop may be involved in protein translocation. In addition, the previously unknown peptide-bound crystal structure of SecYEG implies that interactions between the cytoplasmic side of SecY and signal peptides are related to lateral gate opening at the first step of protein translocation. These SecYEG structures therefore provide a number of structural insights into the Sec machinery for further study.journal articl

    This paper is concerned with the reduction of the power loss of the thrust bearing. A thrust bearing with a disk floated by externally pressurized oil is proposed, and the static characteristics of the thrust bearing are investigated by experiments and simplified analysis in the laminar regime. As a result, the following conclusions are obtained: The power loss of the thrust bearing decreases by approximately one-half of the same-shaped hydrostatic bearing without any disk. The disk is floating nearly in the middle of the oil film. The rotational speed of the disk is about one-half of the shaft rotational speed.     Tuning of in vivo cognate B-T cell interactions by Intersectin 2 is required for effective anti-viral B cell immunity

    Wiskott-Aldrich syndrome (WAS) is an immune pathology associated with mutations in WAS protein (WASp) or in WASp interacting protein (WIP). Together with the small GTPase Cdc42 and other effectors, these proteins participate in the remodelling of the actin network downstream of BCR engagement. Here we show that mice lacking the adaptor protein ITSN2, a G-nucleotide exchange factor (GEF) for Cdc42 that also interacts with WASp and WIP, exhibited increased mortality during primary infection, incomplete protection after Flu vaccination, reduced germinal centre formation and impaired antibody responses to vaccination. These defects were found, at least in part, to be intrinsic to the B cell compartment. In vivo, ITSN2 deficient B cells show a reduction in the expression of SLAM, CD84 or ICOSL that correlates with a diminished ability to form long term conjugates with T cells, to proliferate in vivo, and to differentiate into germinal centre cells. In conclusion, our study not only revealed a key role for ITSN2 as an important regulator of adaptive immune-response during vaccination and viral infection but it is also likely to contribute to a better understanding of human immune pathologies

    A switch from canonical to noncanonical autophagy shapes B cell responses

    Autophagy is important in a variety of cellular and pathophysiological situations; however, its role in immune responses remains elusive. Here, we show that among B cells, germinal center (GC) cells exhibited the highest rate of autophagy during viral infection. In contrast to mechanistic target of rapamycin complex 1-dependent canonical autophagy, GC B cell autophagy occurred predominantly through a noncanonical pathway. B cell stimulation was sufficient to down-regulate canonical autophagy transiently while triggering noncanonical autophagy. Genetic ablation of WD repeat domain, phosphoinositide-interacting protein 2 in B cells alone enhanced this noncanonical autophagy, resulting in changes of mitochondrial homeostasis and alterations in GC and antibody-secreting cells.
