    The tyrosinase gene from medakafish,Oryzias latipes

    名古屋大学Nagoya University博士(理学)I have determined the 9.8kb genomic nucleotide sequence of the tyrosinase gene and its 5’ upstream region from a teleost, medakafish (Oryzias latipes), and shown that the coding region is composed of five exons and four introns, spanning 4.7kb. While the number and sizes of the exons were found to be similar to those of mammalian tyrosinase genes, however, the total size of the coding region (4.7kb) was demonstrated to be less than one tenth those of mouse (ca. 70kb) and human (>70kb) genes. Primer extension analysis revealed that the transcription initiation site starts with a long untranslated leader sequence (340 nucleotide long) from the AUG start codon. A characteristic CATGTG sequence known as a putative regulatory motif in melanocyte-specific genes was present in the 131th base upstream from the initiation site, while other typical regulatory elements such as the TATA-box or M-box common to terrestrial vertebrates were lacking. Transgenic experiments were carried out by microinjecting two kinds of plasmid clones into fertilized eggs of the albino i^1 mutant; one consisting of the genomic tyrosinase gene with the 10kb 5’ upstream region, and the other the tyrosinase cDNA with the 3kb 5’ upstream region. The results showed that 53 and 45 of 114 and 118 transgenic eggs, respectively, developed normally beyond hatching and 15 and 10 exhibited a mosaic pattern of pigmentation. Despite the absence of typical regulatory elements like a TATA-box in both cases correct melanin pigmentation was obtained without ectopic expression.     A Method of Estimating Earthquake Ground Motion Using Nearby K-net Records

    A method of estimating earthquake ground motion using microtremor horizontal to vertical(H/V) Fourier spectral ratio is presented. In this formulation, an earthquake ground motion is estimated from the ground motion recorded in the nearby observation site by using the correction factors which consist of the microtremor H/V spectral ratio of the both sites (the reference site and estimation site), the path and local site effect factor of waves β, and the site factor of vertical ground motions γ. By using the observed microtremor records and earthquake ground motions in the Miyazaki Prefecture, the presented method is examined. It is found from this examination that the path and local site effect factor of waves β and the site factor of vertical ground motions γ can be assumed approximately to be 1.0. Also the error of estimation of Fourier spectral amplitude of earthquake horizontal ground motion is discussed

    Elaborate Simulation And Prediction Of Seismic Behavior Of A Truss Bridge

    In this study, the time-history analysis procedure is elaborately carried out for a truss bridge by identifying the actual structural vibrating characteristics measured by on-site tests and employing the input ground motions at different sites converted from the motions recorded at other observation spot. This procedure is first validated by simulating the seismic behavior of a deck-type truss bridge subjected to a moderate earthquake (M5.7) happened near the bridge in 2002. Then an extreme earthquake (M7.5) is assumed on the basis of ground motions recorded in the moderate earthquake, and the corresponding seismic responses of the bridge are predicted, by consistently estimating the ground motions at hypocenter with assumption of the ground faults and formulating ground motions at the site with consideration of spreading procedure from the hypocenter

    本論文では、常時微動H/Vスペクトル比と近傍の強震観測点の地震動記録のみを用いて未観測点の地震動を推定する方法を提案する。本提案式では、「上下動の増幅特性を表す係数」と「常時微動と地震動H/Vスペクトル比の違いを表す補正係数」の2つの係数が、地点毎に計測される常時微動H/Vスベクトル比の特性から推定できることを示した。2地点問距離が3~6kmと短い宮崎県内のK-NET観測点とFDMA(消防庁)観測点並びに、福岡市内の地震動記録と常時微動記録を用いて地震動推定法の推定精度を検証し、これら2つの係数に地点毎の地盤振動特牲を考慮することにより、短周期地震動(周期0.1秒から2秒)の推定精度が向上することを示した。A method of estimating earthquake ground motion using microtremor horizontal to vertical (H/V)Fourier spectral ratio is presented.     Seismic Response Analysis of BFG Holder Considering Nonlinear Soil Foundation Interaction

    Because currently existing important industrial and civil structures were designed and constructed on the basis of the past earthquake resistant design regulations, it is important examine the seismic response behaviors of these existing important structures by using the current knowledge on earthquake loadings and the advanced method of 3-dimensional nonlinear seismic response analysis. In this paper, the 3-dimensional nonlinear seismic response behaviors of a BFG holder have been examined using a 3-dimensional fiber element method that can make it possible to model digitally the substructures and superstructures based on the nonlinear stress-strain behaviors of the materials of the structures. the computer code of 3-D filber element method used in this analysis can take into account for the 3-dimensional nonlinear dynarmic soil foundation interaction, and also the material and geometricl nonlinear behaviors of the beam and shell elements of the BFG holder. It is found from numerical analysis that the seismic behaviors of BFG holder strongly change depending on the height (position) of the piston of the BFG holder and also the piston moves laterally and rotationally during earthquake loadings
