In vitro characterization of missense mutations associated with quantitative protein Sdeficiency
名古屋大学NAGOYA University博士(医療技術学)Objective: To elucidate the molecular consequences of hereditary protein S (PS) deficiency, we investigated the in vitro synthesis of the PS missense mutants in COS-1 cells and their activated protein C (APC) cofactor activities. Patients: Four patients with quantitative PS deficiency suffering from venous thrombosis were examined. Results: We identified three distinct novel missense mutations, R275C, P375Q and D455Y, and two previously reported missense mutations, C80Y and R314H. The P375Q and D455Y mutations were found in one patient and observed to be in linkage on the same allele. The R314H mutant showed the lowest level of expression (32.7%), and the C80Y, P375Q + D455Y, and R275C mutants exhibited a moderate impairment of expression, that is, 43.8%, 49.5%, and 72.3% of the wild type, respectively. Furthermore, pulse-chase experiments demonstrated that all mutants showed impaired secretion and longer half-lives in the cells than the wild type PS. In the APC cofactor assays, the C80Y mutant showed no cofactor activity, and the R275C mutant showed reduced activity, 62.3% of the wild type PS, whereas the R314H and P375Q + D455Y mutants exhibited normal cofactor activity. Stepped wedge cluster randomized controlled trial designs: a review of reporting quality and design features
Background
The stepped wedge (SW) cluster randomized controlled trial (CRCT) design is being used with increasing frequency. However, there is limited published research on the quality of reporting of SW-CRCTs. We address this issue by conducting a literature review.
Methods
Medline, Ovid, Web of Knowledge, the Cochrane Library, PsycINFO, the ISRCTN registry, and ClinicalTrials.gov were searched to identify investigations employing the SW-CRCT design up to February 2015. For each included completed study, information was extracted on a selection of criteria, based on the CONSORT extension to CRCTs, to assess the quality of reporting.
Results
A total of 123 studies were included in our review, of which 39 were completed trial reports. The standard of reporting of SW-CRCTs varied in quality. The percentage of trials reporting each criterion varied to as low as 15.4%, with a median of 66.7%.
Conclusions
There is much room for improvement in the quality of reporting of SW-CRCTs. This is consistent with recent findings for CRCTs. Centrifuge modelling of building response to tunnel excavation
Understanding the building response to tunnelling-induced settlements is an important aspect of urban tunnelling in soft ground. Previous centrifuge modelling research demonstrated significant potential to study this tunnel–soil–structure interaction problem. However, these recent studies were limited by simplified building models, which might result in uncertainties when interpreting the building performance to tunnelling subsidence. This paper presents an experimental modelling procedure and the results of a series of centrifuge tests, involving relatively complex surface structures subjected to tunnelling in sand. Powder-based three-dimensional (3D) printing was adopted to fabricate building models with realistic layouts, facade openings and foundations. The 3D printed material had a Young's modulus and a brittle response similar to historic masonry. Modelling effects and boundary conditions are quantified. The good agreement between the experimentally obtained results and previous research demonstrates that the soil–structure interaction during tunnel excavation is well replicated. The experimental procedure provides a framework to quantify how building features affect the response of buildings to tunnelling subsidence.The authors are grateful to EPSRC grant EP/K018221/1 and Crossrail for the financial support

Autonomous Sensory Meridian Response (ASMR) is a type of tingling, static-like sensation that is triggered by special audiovisual stimulation. The sensation passes through the scalp and the back of the neck, sometimes even spreading to the ends of the extremities. In recent years, research on ASMR has been gradually increasing. However, few collections of ASMR video material have been evaluated so far. In the present study, 807 Chinese participants (ASMR participants = 435, non-ASMR participants = 372) were asked to evaluate two types of ASMR videos and one control video, assessing the intensity and duration of tingling sensations triggered by these videos. A total of 60 ASMR videos were screened. The subjective assessment of the experimental group on ASMR intensity and duration, as triggered by the ASMR video material, demonstrated that the library contains 60 ASMR videos that can effectively trigger ASMR in participants who are able to experience ASMR. This video library was then subjected to a test which revealed that Cronbach’s α = 0.933. This proves that the library has good reliability, that it can effectively trigger ASMR in participants who are able to experience ASMR, and that it can be used as experimental material in future ASMR research.</p
