    The Influence of Mixing Conditions of Reaction Medium on Protease-Catalyzed Peptide Synthesis: The Use of Sonication

    Sonication was demonstrated to be a useful method mixing reactoin medium. A model reaction, Boc-Gly-OH + H-Phe-N₂H₂Ph → Boc-Gly-phe-N₂H₂Ph, was adopted to compare two mixing methods of medium, sonication and magnetic-stirring. Under biphasic system including 25% of organic solvent, sonication was preferable to magnetic-stirring for several cosolvents and showed being able to have some potentiality as mixing method. On the other hand another model reaction, Boc-Tyr (Bzl)-OH + H-Gly-N₂H₂Ph→Boc-Tyr (Bzl)-Gly-N₂H₂Ph, which had not been achieved by other investigators, was progressed by the new mixing method.     α-1,3-フコース転移酵素cDNAの導入と強制発現により、L細胞のインテグリン依存性の接着が促進された。ヒト骨髄球系細胞に於て、ガングリオシドの合成を制御する酵素としてGM3を合成するシアル酸転移酵素とLc3Cerを合成するN-アセチルグルコサミン転移酵素を同定した。ヒト結腸癌組織においてシアリルLe^aの抗原を持つ糖鎖の構造を明かにした。ルウィス血液型陰性の個体に於て、FuT-III遺伝子の変異をヌクレオチドレベルで解明した。胃体部と胃前庭部に於て、消化管ムチンの硫酸化糖鎖の構造パターンが異なっていた。Gsl-5糖鎖を形成するβ-1、6-N-アセチルグルコサミン転移酵素を精製し、アミノ酸部分配列を明かにした。肝細胞膜の表層糖鎖末端β-ガラクトシドが肝細胞接着に関与し、この機構にC-型レクチンは関与しないと示唆された。成人型異染性脳白質変性症の原因となるアリルスルファタ-ゼAの新しい構造異常を見出した。ウシ精巣性ヒアルロニダーゼの糖転移反応を利用して、新しいハイブリッド型プロチオグリカンを作成した。ニワトリCa^依存性α-マンノシダ-ゼの特異性を解明した。マクロファ-ジレクチンのチロシン残基(Tyr5)がインタ-ナリゼイションに必須なことを明かにした。また、C-型レクチンの1つであるコンアグルチニンのcDNAおよびゲノムシークエンスを明かにした。Sophora japonica樹皮中のレクチンが損傷の修復に関与する可能性を指摘した。アルコール依存症患者血清に糖鎖欠損トランスフェリンを見出した。コンドロイチン硫酸のペプチドとの結合部位を合成した。ヒトマクロファージのレクチンcDNAを単離し、構造決定した。科学研究費補助金 研究種目:総合研究(A) 課題番号:03304050 研究代表者:村松 喬 研究期間:1991-1993年度research repor

    Evaluation for Optical and Thermal Properties of Si-nanopillars(NPs)/SiGe Composite Film Intentionally Varied NPs Spacing

    Thermoelectric (TE) devices have been attracted attention because of their importance in converting waste heat into electrical energy. Si-based materials are expected to use as low-cost TE devices. By introducing nanostructure into the TE materials, both low thermal (κ) and high electrical conductivities are required. We have succeeded to fabricate a highly periodic Si-nanopillars (NPs) structure embedded in Si0.7Ge0.3. Bio-template and Neutral-Beam Etching techniques enable us to fabricate the NPs of 10 nm diameter. In this study, we have prepared two samples with different NP spacing of 40 and 60 nm. Our preliminary 3 measurements showed that κ of 40 and 60 nm spacing samples were 2.6 and 2.8 W/mK, respectively. Then, these were 35% lower than that of SiGe bulk material. However, this method requires an electrode as thin as the composite film. This is very difficult to prepare and we should use an electrode of 10 m width. Therefore, another evaluation technique is necessary to estimate κ more accurately. Piezoelectric photothermal (PPT) measurements were carried out to study the effect of NPs spacing on the optical and thermal properties from the non-radiative recombination point of view. The observed PPT signal of the 40 nm spacing sample was 10 times smaller than that of the 60 nm spacing sample. Since the PPT signal depends on κ as well as optical absorption coefficient ( ), we carried out a theoretical calculation based on a one-dimensional heat propagation equation to estimate κ and . However, the calculated results gave us unexpectedly lower κ and higher. A detailed 3-dimensional heat propagation model including carrier diffusion of photo excited carriers should be considered.departmental bulletin pape

    Evaluation of thermal properties of semiconductors by using a laser heterodyne photothermal displacement measurement (2) Measurement and calculation of Si-NP/SiGe composite film

    Laser heterodyne photothermal displacement measurements were carried out to clarify the thermal conductivity (????) of Si-nanopillars/SiGe composite films. To investigate how the nanopillar spacing reduces ????, three samples with different nanopillar spacing were prepared. Comparing the theoretical calculations based on the single-layer thermal diffusion equation with the experimental results, surface displacement and its time variation could not be explained by change in value of ????????. It was found that differences in the Si-NP spacing may have caused a change in optical properties such as the optical absorption coefficient. Furthermore, theoretical calculations based on the thermal diffusion equation of the three-layer model were used to reproduce the experimental results. The ???????? of the Si-NP/SiGe composite film was estimated by fitting the time variation of displacement. Obtained best-fitted value of ???????? were too small and were the order of 0.1 W/mK. Although quantitative analysis for ???????? needs improvement, the present LH-PD measurement and three-layer model analysis are useful to investigate the thermal properties of semiconductors.departmental bulletin pape
