    Experimental study on the dynamic properties of rigid polyurethane foam in stress-controlled cyclic uniaxial tests

    application/pdfFrequent major earthquakes in Japan necessitate earthquake-resistant infrastructure. The dynamic deformation characteristics of rigid polyurethane foam, used as a lightweight embankment material, is examined in this study. The effect of the initial static stress on the shear modulus of single-layer samples was small, while the shear modulus of two-layer samples increased with increasing initial static stress. The damping ratio of single-layer samples converged to 4–5%, but decreased with increasing initial static stress up to 30 kPa in two-layer samples before increasing above 40 kPa. Results from cyclic uniaxial and triaxial tests were compared. The dynamic properties depended on restriction conditions and the deformation direction

    高温・加圧型熱重量分析装置の開発研究

    エネルギ-、環境さらには材料開発などの研究のブレ-クスル-が高温・高圧下での反応工学、反応プロセスの導入により達成される可能性が高い。このため、高温かつ高圧という厳しい制約条件を満たす精度の高い信頼性のある反応速度測定手法を確立することが急務となっている。そこで、本研究では高温・加圧型熱天秤装置を試作し、その測定法を確立することを最終目的とし、平成7年度は試作装置の制作、ならびにその信頼性を検証するためにゼオライト/CO_2,活性炭/CO_2を中心とした固体吸着剤による吸・脱着、CaO/CO_2/CaCO_3などの吸・発熱固・気化学反応を例とし、常温〜1200℃、常圧〜 10気圧程度の条件下で測定を行い、既往の常圧熱天秤、カロリメ-タ-式吸着装置などの結果と比較、本装置の信頼性を評価した。平成8年度は試作装置の測定デ-タの一般性、普遍性を確かめ実用機器としての位置づけを明確にするため、測定上の問題となるCaO/H_2O/Ca(OH)_2、CaO/SO_2 /CaSO_4、CaO/HCl/CaCl_2などの凝縮性、腐食性ガスが関与する固・気高温化学反応の熱重量分析を先と同様の条件で測定し、その測定限界条件の検討を行うと共に引き続き装置の信頼性の評価を行った結果、本装置の信頼性画検証された。さらに上記の結果を総合的に検討した結果、広範囲測定を可能とするアドバンスト型熱重量分析装置の設計基準が確立された。科学研究費補助金 研究種目:基盤研究(A)(1) 課題番号:07555545 研究代表者:松田 仁樹 研究期間:1995-1996年度research repor

    Functional Characterization of IPSC-Derived Brain Cells as a Model for X-Linked Adrenoleukodystrophy

    <div><p>X-ALD is an inherited neurodegenerative disorder where mutations in the <i>ABCD1</i> gene result in clinically diverse phenotypes: the fatal disorder of cerebral childhood ALD (cALD) or a milder disorder of adrenomyeloneuropathy (AMN). The various models used to study the pathobiology of X-ALD disease lack the appropriate presentation for different phenotypes of cALD vs AMN. This study demonstrates that induced pluripotent stem cells (IPSC) derived brain cells astrocytes (Ast), neurons and oligodendrocytes (OLs) express morphological and functional activities of the respective brain cell types. The excessive accumulation of saturated VLCFA, a “hallmark” of X-ALD, was observed in both AMN OLs and cALD OLs with higher levels observed in cALD OLs than AMN OLs. The levels of <i>ELOVL1</i> (<i>ELOVL</i> Fatty Acid Elongase 1) mRNA parallel the VLCFA load in AMN and cALD OLs. Furthermore, cALD Ast expressed higher levels of proinflammatory cytokines than AMN Ast and control Ast with or without stimulation with lipopolysaccharide. These results document that IPSC-derived Ast and OLs from cALD and AMN fibroblasts mimic the respective biochemical disease phenotypes and thus provide an ideal platform to investigate the mechanism of VLCFA load in cALD OLs and VLCFA-induced inflammatory disease mechanisms of cALD Ast and thus for testing of new therapeutics for AMN and cALD disease of X-ALD.</p></div

    Essed for EMSA as described in 'Material and Methods' (A). bEND.3 cells were transiently transfected with p65, p50 expression vectors along with NF-κB luciferase reporter construct (0.5 μg/well) and pCMV-β-galactosidase (0.5 μg/well) followed by treatment with 15d-PGJ2 (5–20 μM) for 4 h and processed for luciferase and β-galactosidase activities. Luciferase activity was normalized with respect to β-gal activity (B). Results were calculated as mean ± SD for 3 independent experiments. Samples were run in triplicates. &&& p < 0.001 compared with control and !!! p < 0.001 compared with TNFα treatment (50 ng/ml). Cells were treated with 15d-PGJ2(10 μM) for 30 min followed by TNFα for 20 min and stained with anti-p65 antibody as described in 'Material and Methods' (C).
