Recombinant human neuroblastoma sh-sy5y cells overexpressing α-synuclein form amyloid aggregates in seed-dependent and seed-independent manners
Alpha-synuclein (α-Syn) has prion-like properties and is one of the causative proteins of Parkinson’s disease (PD) and dementia with Lewy bodies (DLB). Neuronal loss is thought to result from amyloid aggregation caused by misfolding of protein and propagation of misfolded protein aggregate as seeds through the neural network. This may contribute to the progression of PD and DLB. Prevention of accumulation and propagation of amyloid and neuronal loss is therefore important in the prevention and slowing of the progression of PD and DLB. In this study, we aimed to establish a neuronal cell line stably overexpressing α-Syn and to clarify its ellular characteristics for the study of amyloid aggregation and aggregation inhibition. Through gene transfer and selection with the antibiotic G418, we obtained subclones derived from SH-SY5Y, the human euroblastoma cell line that overexpress α-Syn. The subclones were found to form amyloid aggregates in a markedly short period of time upon addition of α-Syn amyloid fibril-derived seeds. They also expressed βIII tubulin, a mature neuron marker, in long-term culture with differentiation induction by retinoic acid or brain-derived neurotrophic factor stimulation. The subclones are thus suggested to retain the ability to differentiate into mature neurons as well as the wild type. Furthermore, the subclones in long-term culture showed seed-independent aggregate formation, nuclear condensation, and fragmentation. This suggests that the subclones spontaneously form ggregates and undergo cell death under long-term culture due to high α-Syn expression levels. The α-Syn-overexpressing SH-SY5Y cell line established in this study is thought to be useful for research on α-Syn amyloid aggregate formation, inhibition of aggregate formation, propagation, and aggregate-associated cell death. Moreover, the study suggests the potential for use in research on neuronal dysfunction by inducing differentiation into mature neurons, and is therefore expected to be useful in preventing and slowing the progression of PD and DLB.ORIGINAL ARTICLEarticl
Phase-space tomography in magnetically confined plasmas
0000-0001-5669-1937In this paper, a tomography approach aiming at reconstructing a phase-space structure is proposed. For the phase-space resolved diagnostic system, a signal must be decomposed in real-space, velocity-space, and time; therefore, it is challenging to obtain a sufficiently high signal intensity in a single detector bin. To overcome this difficulty, three different sets of data having different integration directions in real-space, velocity-space, and time are simultaneously used, and a reconstruction of the original structure in the phase-space is attempted by a tomographic manner. The proposed method is demonstrated using a synthetic dataset in the actual diagnostic setup in the Large Helical Device. Time evolution of a phase-space perturbation induced by the Landau damping, which is caused by energetic particle-driven magnetohydrodynamic bursts, is successfully reconstructed by this method. Robustness against realistic diagnostic noise is also presented.journal articl
Extended investigation of superdeformed bands in Tb nuclei
A detailed study of known and new SD bands in Tb isotopes has been performed with the use of the EUROBALL IV -ray array. The high-statistics data set has allowed for the extension of known SD bands at low and high spins by new -ray transitions. These transitions, as it turns out, correspond to the rotational frequencies where the principal superdeformed gaps (Z=66,N=86) close giving rise to up- or down-bending mechanisms. This enables to attribute the underlying theoretical configurations with much higher confidence as compared to the previous identifications. Five new SD bands have been discovered, three of them assigned to the 152Tb and the two others to the 151Tb nuclei. Nuclear mean-field calculations have been used to interpret the structure of known SD bands as well as of the new ones in terms of nucleonic configurations

In order to waste concrete is consist of mortar and coarse aggregate, it is impossible to separate them completely. So waste concrete crush up, and are using as base coarse material. But road construction is reducing now. From the point of view, when waste concrete using as coarse aggregate, these chemical and mechanical properties are unknown.
