    2017-08Yokukansankachimpihange (YKSCH), a traditional Japanese medicine, is widely used for the amelioration of the behavioral and psychological symptoms of dementia with digestive dysfunction. Regardless of its successful use for digestive dysfunction, the effect of YKSCH on body weight was unknown. Furthermore, if YKSCH increased body weight, it might increase motivation according to Kampo medicine theory. Therefore, we investigated whether YKSCH had the potential to increase body weight and enhance motivation in mice. To address this, C57BL/6J mice were used to evaluate the long-term effect of YKSCH on body weight and food-incentive motivation. As part of the evaluation, we optimized an operant test for use over the long-term. We found that feeding mice YKSCH-containing chow increased body weight, but did not increase their motivation to food reward. We propose that YKSCH may be a good treatment option for preventing decrease in body weight in patients with dementia.departmental bulletin pape

    鶏における大腸菌性蜂窩織炎とブドウ球菌症に対する Macrophage Activating Chinese Mixed Herbs (MACH) の防御効果

    インターフェロン誘起作用を有し、マクロファージ遊走能を促進する効果を持つことが知られているMacrophage Activating Chinese Mixed Herbs(MACH)の、ブロイラー鶏の大腸菌性蜂窩織炎およびひなのブドウ球菌症に対する防御効果を検証した。鶏をMACH添加飼料給餌群と普通の市販飼料給餌群に分け、実験1では蜂窩織炎野外例より分離された大腸菌を43日齢のブロイラー鶏の大腿部皮膚に掻傷を作出し、その上に塗布した。実験2ではブドウ球菌症野外例より分離された黄色ブドウ球菌(SA)を14日齢の雛の大腿部と胸部筋肉内に接種した。実験1では大腸菌感染による死亡率、蜂窩織炎発現羽数はMACH添加飼料給餌鶏で抑制され、掻傷の治癒機転についてもMACHが効果的に作用していることが示された。実験2では、ブドウ球菌症によるひなの死亡率、病変発現羽数はMACH添加飼料群と市販飼料群との間で差が認められず、浮腫性皮膚炎はMACH添加飼料群において重度であった。組織学的にはMACH添加群では接種部位におけるマクロファージ数が増加しており、マクロファージの遊走能促進効果が示唆された。MACHは掻傷からの大腸菌の侵入といった局所感染の防御において有用であり、また、軽度病変の発現を抑える効果を示すことが示唆された。しかし、重度病変の発現を抑えるまでには至らず、その効能は日齢や攻撃菌の病原性により差が出るのではないかと考えられた。これらのことから、 植物多糖体であるMACHは、抗菌剤を用いることのできない出荷時や無薬飼育法における感染防御に対して、あるいは抗菌剤と併用することで、日常の健康管理に有用な添加物質になる可能性が考えられた。The protective effect of Macrophage Activating Chinese Mixed Herbs (MACH),known to have interferon-inducing activity and to promote migration of macrophages,was investigated pathologically for Escherichia coli (E. coli) cellulitis and staphylococcosis in chickens. Chickens used for this study were fed diets with or without MACH. In experiment 1,three E. coli strains isolated from the field case of cellulitis were applied to scratches made on the thighs of 52-day-0ld broiler chickens. In experiment 2,Staphylococcus aureus (SA) isolated from the field case of chicken staphylococcosis was inoculated intramuscularly in thighs and thoraxes of 14-day-0ld chicks. In experiment 1,chickens infected with E. coli had lower mortality and morbidity when fed MACH. MACH also enhanced wound healing. In experiment 2,chickens infected with SA exhibited no difference between groups fed diets with or without MACH. Dermatitis lesions were more severe in MACH groups. Histologically, macrophage counts increased at the inoculation site,which suggests that macrophage migration may be promoted. This suggested that MACH was useful for cutaneous protection from invasion of E.coli, and effective against development of mild lesions. However,it was,not potent enough to suppress the development severe lesions,and its effect may vary with age and pathogen. MACH may thus be useful for routine health management as an additive substance with or without antibiotics.journal articl
