New in vivo approach to broaden the thioredoxin family interactome in chloroplasts

Post-translational redox modifications provide an important mechanism for the control of major cellular processes. Thioredoxins (Trxs), which are key actors in this regulatory mechanism, are ubiquitous proteins that catalyse thiol-disulfide exchange reactions. In chloroplasts, Trx f, Trx m and NADPH-dependent Trx reductase C (NTRC) have been identified as transmitters of the redox signal by transferring electrons to downstream target enzymes. The number of characterised Trx targets has greatly increased in the last few years, but most of them were determined using in vitro procedures lacking isoform specificity. With this background, we have developed a new in vivo approach based on the overexpression of His-tagged single-cysteine mutants of Trx f, Trx m or NTRC into Nicotiana benthamiana plants. The over-expressed mutated Trxs, capable of forming a stable mixed disulfide bond with target proteins in plants, were immobilised on affinity columns packed with Ni-NTA agarose, and the covalently linked targets were eluted with dithiothreitol and identified by mass spectrometry-based proteomics. The in vivo approach allowed identification of 6, 9 and 42 new potential targets for Trx f, Trx m and NTRC, respectively, and an apparent specificity between NTRC and Trxs was achieved. Ways to Instruct Physical Education for Students with Special Care Needs: An Interview Survey with af Volleyball Instructor

本研究は，バレーボール指導者へのインタビューより，特別な配慮が必要な児童生徒への体育指導のあり方を検討した。 　2019(平成31)年２月中旬に，体育指導の専門家１名に対してインタビューを行った。まず，「各学年に合わせてどのような体育指導を行おうと思っているか」，そして，「特別な配慮を必要とする児童生徒に対してどのような体育指導を行おうと思っているか」など，自由に語ってもらうこととした。もし可能であれば，個人情報に抵触しない範囲で経験にもとづいて教えてほしいことも伝えた。インタビュー内容から，協力者の思いを抽出してまとめ，インタビュー協力者が「特別な配慮が必要な児童生徒に対する体育指導」をどのように考えているのかを検討していった。インタビューにもとづいてまとめた内容については，執筆者３名で検討を重ねるとともに，インタビュー協力者に一読を賜り，協力者の指導観であることを確認してもらった。インタビューの趣旨について口頭で説明し，了解を得た後にインタビューを開始した。内容の集約ができたのちに，インタビュー協力者に内容を確認してもらった。原稿が仕上がったのちに一読してもらい，公表の了承を得た。 　検討の結果，「聴覚障がいのある児童生徒への指導の場合」，「発達障がいのある児童生徒への指導の場合」，また，「低学年の児童生徒への指導の場合」，「児童生徒へのバレーボール指導を通しての指導観」について見出すことができた。これらのことから，特別な配慮が必要な児童生徒への体育指導には，触れてみる，動作を見て学ぶという，触覚や視覚という感覚を重視した指導が必要であることが把握できた。departmental bulletin pape

Killing of <i>G. duodenalis</i> trophozoites by orlistat.

<p>Giardiacidal effects of orlistat or MTZ were determined for drug concentrations indicated using <i>G. duodenalis</i> strains WB-C6 and 14-03/F7. WB-C6 trophozoites (5×10⁴) and 14-03/F7 trophozoites (1×10⁵) were treated with orlistat and MTZ, respectively, for 24 h and, after drug removal, cultured for another 48 h. Trophozoite numbers were determined every 24 h. (A) Orlistat was used at 5×(21.5 µM), 10×(43 µM), and 15×(64.5 µM) of the IC50_24h value for WB-C6. (B) Similarly, MTZ was used at 5×(55 µM), 10×(110 µM), and 15×(165 µM) of the IC50_24h concentration for WB-C6. (C) Orlistat was used at 5×(14 µM), 10×(28 µM), and 15×(42 µM) of the IC50_24h value for 14-03/F7. (D) Similarly, MTZ was used at 5×(31 µM), 10×(62 µM), and 15×(93 µM) of the IC50_24h concentration for 14-03/F7. Data are expressed as mean ± SD of parasite numbers determined in triplicate cultures.
