Estudio experimental comparativo de la osteotomía a cielo abierto y percutánea en la elongación ósea

Se ha realizado un trabajo experimental sobre elongacione s femorales en corderos, planteado con los siguiente s objetivos: 1) Determina r el abordaj e (a "cielo abierto" o percutáneo ) y nive l de osteotomí a ideale s en las elongacione s ósea s y 2) Estudiar la integridad de las estructuras endomedulare s tras las osteotomías percutá- neas llamadas corticotomías. Se han utilizado 50 corderos macho s de 2 mese s de edad que se han agrupado según el tipo y nivel de osteotomía empleado para la elongación: percutánea metafisaría, percutánea diafisaria, abierta diafisaria y abierta metafisaría. Además, en otro grupo de 10 corderos se ha estudiado la integridad de los vasos endomedulares tras el intento de osteotomía cortical percutánea diafisaria. Las conclusiones han sido: 1) La osteotomía ideal para las elongaciones óseas es la realizada percutáneament e y de localízación metafisaría. 2) La integridad de las estructuras endomedulare s tras la osteotomía, no parec e ser la responsabl e de las satisfactorias reconstruccione s obtenidas con el uso de osteotomías percutáneas.An experimental study on femoral lengthening in lambs has been performed with the following objetives in mind: 1) To determine the ideal technique and level of osteotomy in bone lengthening: 2) To assess the integrity of the intramedullary vessels after the so-called corticotomies. Fifty 2-month-old male lambs have been included in this experiment. Animal grouping has been made according to the type and level of the lengthening osteotomy performed: percutaneous metaphyseal, percutaneous diaphyseal, open diaphyseal and open metaphyseal. Another group of ten lambs has been included in orde r to asse s the condition of the intramedullary vessels just after a percutaneous diaphyseal osteotomy. The main conclusions of this study are as follows: 1) The ideal tecnique and level of osteotomy for bone lengthening in lambs seem to be percutaneous and metaphyseal. 2) The satisfactory reconstructions of the lengthened segment observed using percutaneous osteotomie s cannot be explained by the maintenanc e of the integrity of the intramedullary vessels after the osteotomy

Ensayos de tapas troncocónicas de PRFV

This paper presents the tests carried out on truncated cone covers made of glass fibre reinforced plastic (GFRP) that are used as covering and closing devices for circulation areas used by pedestrians and vehicles. Physical and numerical tests were carried out on flat covers to characterise the material. Then, with the characterised material, physical and numerical tests were carried out on GFRP truncated cone covers, simulating real operating conditions. The results obtained allow us to infer that the truncated cone covers certify the A15, B125 and C250 classes established by the UNE-EN 124-1:2015 standard.  En este trabajo se muestran los ensayos realizados a tapas troncocónicas de plástico reforzado con fibras de vidrio (PRFV) que son empleadas como dispositivos de cubrimiento y de cierre para zonas de circulación utilizadas por peatones y vehículos. Se realizaron ensayos físicos y numéricos sobre tapas planas para caracterizar el material. Luego, con el material caracterizado, se realizaron ensayos físicos y numéricos sobre tapas troncocónicas de PRFV, simulando las condiciones de funcionamiento reales. Los resultados obtenidos permiten inferir que las tapas troncocónicas certifican las clases A15, B125 y C250 que establece la norma UNE-EN 124-1:2015

ERK inhibitor LY3214996-based treatment strategies for RAS-driven lung cancer

RAS gene mutations are the most frequent oncogenic event in lung cancer. They activate multiple RAS-centric signaling networks among them the MAPK, PI3K and RB pathways. Within the MAPK pathway ERK1/2 proteins exert a bottleneck function for transmitting mitogenic signals and activating cytoplasmic and nuclear targets. In view of disappointing anti-tumor activity and toxicity of continuously applied MEK inhibitors in patients with KRAS mutant lung cancer, research has recently focused on ERK1/2 proteins as therapeutic targets and on ERK inhibitors for their ability to prevent bypass and feedback pathway activation. Here we show that intermittent application of the novel and selective ATP-competitive ERK1/2 inhibitor LY3214996 exerts single-agent activity in patient-derived xenograft (PDX) models of RAS mutant lung cancer. Combination treatments were well tolerated and resulted in synergistic (ERKi plus PI3K/mTORi LY3023414) and additive (ERKi plus CDK4/6i abemaciclib) tumor growth inhibition in PDX models. Future clinical trials are required to investigate if intermittent ERK inhibitor-based treatment schedules can overcome toxicities observed with continuous MEK inhibition and - equally important - to identify biomarkers for patient stratification

Genomic and biological study of fusion genes as resistance mechanisms to EGFR inhibitors

The clinical significance of gene fusions detected by DNA-based next generation sequencing remains unclear as resistance mechanisms to EGFR tyrosine kinase inhibitors in EGFR mutant non-small cell lung cancer. By studying EGFR inhibitor-resistant patients treated with a combination of an EGFR inhibitor and a drug targeting the putative resistance-causing fusion oncogene, we identify patients who benefit and those who do not from this treatment approach. Through evaluation including RNA-seq of potential drug resistance-imparting fusion oncogenes in 504 patients with EGFR mutant lung cancer, we identify only a minority of them as functional, potentially capable of imparting EGFR inhibitor resistance. We further functionally validate fusion oncogenes in vitro using CRISPR-based editing of EGFR mutant cell lines and use these models to identify known and unknown drug resistance mechanisms to combination therapies. Collectively, our results partially reveal the complex nature of fusion oncogenes as potential drug resistance mechanisms and highlight approaches that can be undertaken to determine their functional significance.</p
