Cavity-enhanced photoionization of an ultracold rubidium beam for application in focused ion beams
A two-step photoionization strategy of an ultracold rubidium beam for
application in a focused ion beam instrument is analyzed and implemented. In
this strategy the atomic beam is partly selected with an aperture after which
the transmitted atoms are ionized in the overlap of a tightly cylindrically
focused excitation laser beam and an ionization laser beam whose power is
enhanced in a build-up cavity. The advantage of this strategy, as compared to
without the use of a build-up cavity, is that higher ionization degrees can be
reached at higher currents. Optical Bloch equations including the
photoionization process are used to calculate what ionization degree and
ionization position distribution can be reached. Furthermore, the ionization
strategy is tested on an ultracold beam of Rb atoms. The beam current is
measured as a function of the excitation and ionization laser beam intensity
and the selection aperture size. Although details are different, the global
trends of the measurements agree well with the calculation. With a selection
aperture diameter of 52 m, a current of pA is
measured, which according to calculations is 63% of the current equivalent of
the transmitted atomic flux. Taking into account the ionization degree the ion
beam peak reduced brightness is estimated at A/(msreV).Comment: 13 pages, 9 figure
Methylation Status of Imprinted Genes and Repetitive Elements in Sperm DNA from Infertile Males
Stochastic, environmentally and/or genetically induced disturbances in the genome-wide epigenetic reprogramming processes during male germ-cell development may contribute to male infertility. To test this hypothesis, we have studied the methylation levels of 2 paternally (H19 and GTL2) and 5 maternally methylated (LIT1, MEST, NESPAS, PEG3, and SNRPN) imprinted genes, as well as of ALU and LINE1 repetitive elements in 141 sperm samples, which were used for assisted reproductive technologies (ART), including 106 couples with strictly male-factor or combined male and female infertility and 28 couples with strictly female-factor infertility. Aberrant methylation imprints showed a significant association with abnormal semen parameters, but did not seem to influence ART outcome. Repeat methylation also differed significantly between sperm samples from infertile and presumably fertile males. However, in contrast to imprinted genes, ALU methylation had a significant impact on pregnancy and live-birth rate in couples with male-factor or combined infertility. ALU methylation was significantly high-er in sperm samples leading to pregnancy and live-birth than in those that did not. Design and experimental validation of a compact collimated Knudsen source
In this paper we discuss the design and performance of a collimated Knudsen
source which has the benefit of a simple design over recirculating sources.
Measurements of the flux, transverse velocity distribution and brightness at
different temperatures were conducted to evaluate the performance. The scaling
of the flux and brightness with the source temperature follow the theoretical
predictions. The transverse velocity distribution in the transparent operation
regime also agrees with the simulated data. The source was found able to
produce a flux of s at a temperature of 433 K. Furthermore the
transverse reduced brightness of an ion beam with equal properties as the
atomic beam reads A/(m sr eV) which is sufficient for
our goal: the creation of an ultra-cold ion beam by ionization of a
laser-cooled and compressed atomic rubidium beam

Direct magneto-optical compression of an effusive atomic beam for high-resolution focused ion beam application
An atomic rubidium beam formed in a 70 mm long two-dimensional
magneto-optical trap (2D MOT), directly loaded from a collimated Knudsen
source, is analyzed using laser-induced fluorescence. The longitudinal velocity
distribution, the transverse temperature and the flux of the atomic beam are
reported. The equivalent transverse reduced brightness of an ion beam with
similar properties as the atomic beam is calculated because the beam is
developed to be photoionized and applied in a focused ion beam. In a single
two-dimensional magneto-optical trapping step an equivalent transverse reduced
brightness of A/(m sr eV) was
achieved with a beam flux equivalent to nA. The
temperature of the beam is further reduced with an optical molasses after the
2D MOT. This increased the equivalent brightness to A/(m sr eV). For currents below 10 pA, for which disorder-induced
heating can be suppressed, this number is also a good estimate of the ion beam
brightness that can be expected. Such an ion beam brightness would be a six
times improvement over the liquid metal ion source and could improve the

Optimization of Gutzwiller Wavefunctions in Quantum Monte Carlo
Gutzwiller functions are popular variational wavefunctions for correlated
electrons in Hubbard models. Following the variational principle, we are
interested in the Gutzwiller parameters that minimize e.g. the expectation
value of the energy. Rewriting the expectation value as a rational function in
the Gutzwiller parameters, we find a very efficient way for performing that
minimization. The method can be used to optimize general Gutzwiller-type
wavefunctions both, in variational and in fixed-node diffusion Monte Carlo.Comment: 9 pages RevTeX with 10 eps figure
Association of GUCY2C expression in lymph nodes with time to recurrence and disease-free survival in pN0 colorectal cancer.
CONTEXT: The established relationship between lymph node metastasis and prognosis in colorectal cancer suggests that recurrence in 25% of patients with lymph nodes free of tumor cells by histopathology (pN0) reflects the presence of occult metastases. Guanylyl cyclase 2C (GUCY2C) is a marker expressed by colorectal tumors that could reveal occult metastases in lymph nodes and better estimate recurrence risk.
OBJECTIVE: To examine the association of occult lymph node metastases detected by quantifying GUCY2C messenger RNA, using the reverse transcriptase-polymerase chain reaction, with recurrence and survival in patients with colorectal cancer.
DESIGN, SETTING, AND PARTICIPANTS: Prospective study of 257 patients with pN0 colorectal cancer enrolled between March 2002 and June 2007 at 9 US and Canadian centers (7 academic medical centers and 2 community hospitals) provided 2570 fresh lymph nodes measuring 5 mm or larger for histopathology and GUCY2C messenger RNA analysis. Patients were followed up for a median of 24 months (range, 2-63 months) for disease recurrence or death.
MAIN OUTCOME MEASURES: Time to recurrence (primary outcome) and disease-free survival (secondary outcome) relative to expression of GUCY2C in lymph nodes.
RESULTS: Thirty-two patients (12.5%) had lymph nodes negative for GUCY2C (pN0 [mol-]), and all but 2 remained free of disease during follow-up (recurrence rate, 6.3%; 95% confidence interval [CI], 0.8%-20.8%). Conversely, 225 patients (87.5%) had lymph nodes positive for GUCY2C (pN0 [mol+]), and 47 developed recurrent disease (20.9%; 95% CI, 15.8%-26.8%) (P = .006). Multivariate analyses revealed that GUCY2C in lymph nodes was an independent marker of prognosis. Patients who were pN0 (mol+) exhibited earlier time to recurrence (adjusted hazard ratio, 4.66; 95% CI, 1.11-19.57; P = .04) and reduced disease-free survival (adjusted hazard ratio, 3.27; 95% CI, 1.15-9.29; P = .03).
CONCLUSION: Expression of GUCY2C in histologically negative lymph nodes appears to be independently associated with time to recurrence and disease-free survival in patients with pN0 colorectal cancer
Stripes and spin-incommensurabilities are favored by lattice anisotropies
Structural distortions in cuprate materials give a natural origin for
anisotropies in electron properties. We study a modified one-band t-J model in
which we allow for different hoppings and antiferromagnetic couplings in the
two spatial directions ( and ). Incommensurate peaks
in the spin structure factor show up only in the presence of a lattice
anisotropy, whereas charge correlations, indicating enhanced fluctuations at
incommensurate wave vectors, are almost unaffected with respect to the

A family case of fertile human 45,X,psu dic(15;Y) males
We report on a familial case including four male probands from three generations with a 45,X,psu dic(15;Y)(p11.2;q12) karyotype. 45,X is usually associated with a female phenotype and only rarely with maleness, due to translocation of small Y chromosomal fragments to autosomes. These male patients are commonly infertile because of missing azoospermia factor regions from the Y long arm. In our familial case we found a pseudodicentric translocation chromosome, that contains almost the entire chromosomes 15 and Y. The translocation took place in an unknown male ancestor of our probands and has no apparent effect on fertility and phenotype of the carrier. FISH analysis demonstrated the deletion of the pseudoautosomal region 2 (PAR2) from the Y chromosome and the loss of the nucleolus organizing region (NOR) from chromosome 15. The formation of the psu dic(15;Y) chromosome is a reciprocal event to the formation of the satellited Y chromosome (Yqs). Statistically, the formation of 45,X,psu dic(15;Y) (p11.2;q12) is as likely as the formation of Yqs. Nevertheless, it has not been described yet. This can be explained by the dicentricity of this translocation chromosome that usually leads to mitotic instability and meiotic imbalances. A second event, a stable inactivation of one of the two centromeres is obligatory to enable the transmission of the translocation chromosome and thus a stably reduced chromosome number from father to every son in this family
