Attention and the subject of depiction some remarks on Husserl’s approach to the function of attention in phantasy, image consciousness and pictorial experience
This study aims at exposing the phenomenological description of attention as presented by Husserl in his 1904-05 Göttingen-lecture Principal Parts of the Phenomenology and Theory of Knowledge, in its relevance for the study of so-called “intuitive re-presentations”, that is, phantasy and image-consciousness. Starting with the exposition of the fundamental traits of the intentional theory of attention, this study discusses the definition of attention in the terms of meaning [Meinen] and interest, which allows it to become an encompassing modification of all kinds of lived experiences that does not imply an alteration of their act-character (Husserl, 2004: 73). We refer to this character of attention as “plasticity”. In what follows, the study underlines these two definitions of attention and their importance for the understanding of phantasy and image-consciousness. Both kinds of re-presentations will be described stressing the role of attention in the “structuring” of the intentional act and in its affective basis. Finally, the study deals more specifically with the complex description of image consciousness from the viewpoint of the attentional meaning of the image subject
Preferential Paths of Air-water Two-phase Flow in Porous Structures with Special Consideration of Channel Thickness Effects.
Accurate understanding and predicting the flow paths of immiscible two-phase flow in rocky porous structures are of critical importance for the evaluation of oil or gas recovery and prediction of rock slides caused by gas-liquid flow. A 2D phase field model was established for compressible air-water two-phase flow in heterogenous porous structures. The dynamic characteristics of air-water two-phase interface and preferential paths in porous structures were simulated. The factors affecting the path selection of two-phase flow in porous structures were analyzed. Transparent physical models of complex porous structures were prepared using 3D printing technology. Tracer dye was used to visually observe the flow characteristics and path selection in air-water two-phase displacement experiments. The experimental observations agree with the numerical results used to validate the accuracy of phase field model. The effects of channel thickness on the air-water two-phase flow behavior and paths in porous structures were also analyzed. The results indicate that thick channels can induce secondary air flow paths due to the increase in flow resistance; consequently, the flow distribution is different from that in narrow channels. This study provides a new reference for quantitatively analyzing multi-phase flow and predicting the preferential paths of immiscible fluids in porous structures
New insights into the classification and nomenclature of cortical GABAergic interneurons.
A systematic classification and accepted nomenclature of neuron types is much needed but is currently lacking. This article describes a possible taxonomical solution for classifying GABAergic interneurons of the cerebral cortex based on a novel, web-based interactive system that allows experts to classify neurons with pre-determined criteria. Using Bayesian analysis and clustering algorithms on the resulting data, we investigated the suitability of several anatomical terms and neuron names for cortical GABAergic interneurons. Moreover, we show that supervised classification models could automatically categorize interneurons in agreement with experts' assignments. These results demonstrate a practical and objective approach to the naming, characterization and classification of neurons based on community consensus
TYPE AND EXPERIENCE. AN INQUIRY INTO THE ROLE AND FUNCTION OF TYPE AND TYPIFYING-APPERCEPTION IN EXPERIENCE AND COGNITION
The work concentrates on the early works and production of Edmund Husserl, and especially, on the time we consider fundamental for the development of Husserl's later phenomenology. That will also cover a period normally less taken into consideration by the critic. Ideality is in our understanding linked as label-term to the Husserlian \uabeidetics\ubb, which is for its part explicitly derived in the first Book of Ideas from eidos, a term introduced in reality years earlier by Husserl, in connection with essence [Wesen]. It is in fact only around the time of the works belonging to this fundamental introduction to phenomenology that the concept of eidos is assumed as an equivalent for \uabpure essence\ubb and phenomenology is established for its part as \u201ceidetics\u201d or \u201ceidetic science\u201d. Around the time of the Logical Investigations indeed, the term essence comprehends a large variety of concepts less distinguished in comparison with to later Husserlian works, for example, for what concern the concept of eidos. Among the different definitions and functions showed by essence and idea in the 1900 work and in the course on logic and epistemology before the Ideas, we will try to stress one peculiar aspect of such overly complex thematic, which is the partial definition of essence in terms of conceptual universality. With respect to this characterization, we will therefore try to indicate the kind of definition given to it by Husserl, which partially recalls the traditional interpretation of the Universal in the sense of an ideal terms, or a common element, over against the multiplicity. This latter is therefore interpreted in the sense of a universal object. Our aim will be consequently, to analyze a series of manuscripts on the period prior to the Logical Investigations, where, according to our interpretation, all the features assigned to the same concept are present. That will be basically, show the \u201corigin\u201d of one important aspect of the future doctrine of essence, even besides the later interpretation via Lotze and Bolzano of Ideality
Determination of contact angles for three-phase flow in porous media using an energy balance
HYPOTHESIS: We define contact angles, θ, during displacement of three fluid phases in a porous medium using energy balance, extending previous work on two-phase flow. We test if this theory can be applied to quantify the three contact angles and wettability order in pore-scale images of three-phase displacement. THEORY: For three phases labelled 1, 2 and 3, and solid, s, using conservation of energy ignoring viscous dissipation (Δa1scosθ12-Δa12-ϕκ12ΔS1)σ12=(Δa3scosθ23+Δa23-ϕκ23ΔS3)σ23+Δa13σ13, where ϕ is the porosity, σ is the interfacial tension, a is the specific interfacial area, S is the saturation, and κ is the fluid-fluid interfacial curvature. Δ represents the change during a displacement. The third contact angle, θ13 can be found using the Bartell-Osterhof relationship. The energy balance is also extended to an arbitrary number of phases. FINDINGS: X-ray imaging of porous media and the fluids within them, at pore-scale resolution, allows the difference terms in the energy balance equation to be measured. This enables wettability, the contact angles, to be determined for complex displacements, to characterize the behaviour, and for input into pore-scale models. Two synchrotron imaging datasets are used to illustrate the approach, comparing the flow of oil, water and gas in a water-wet and an altered-wettability limestone rock sample. We show that in the water-wet case, as expected, water (phase 1) is the most wetting phase, oil (phase 2) is intermediate wet, while gas (phase 3) is most non-wetting with effective contact angles of θ12≈48° and θ13≈44°, while θ23=0 since oil is always present in spreading layers. In contrast, for the altered-wettability case, oil is most wetting, gas is intermediate-wet, while water is most non-wetting with contact angles of θ12=134°±~10°,θ13=119°±~10°, and θ23=66°±~10°
Pore-scale mechanisms of CO2 storage in oilfields
Rapid implementation of global scale carbon capture and storage is required to limit temperature rises to 1.5 °C this century. Depleted oilfields provide an immediate option for storage, since injection infrastructure is in place and there is an economic benefit from enhanced oil recovery. To design secure storage, we need to understand how the fluids are configured in the microscopic pore spaces of the reservoir rock. We use high-resolution X-ray imaging to study the flow of oil, water and CO2 in an oil-wet rock at subsurface conditions of high temperature and pressure. We show that contrary to conventional understanding, CO2 does not reside in the largest pores, which would facilitate its escape, but instead occupies smaller pores or is present in layers in the corners of the pore space. The CO2 flow is restricted by a factor of ten, compared to if it occupied the larger pores. This shows that CO2 injection in oilfields provides secure storage with limited recycling of gas; the injection of large amounts of water to capillary trap the CO2 is unnecessary
In situ characterization of immiscible three-phase flow at the pore scale for a water-wet carbonate rock
X-ray micro-tomography is used to image the pore-scale configurations of fluid in a rock saturated with three phases - brine, oil and gas - mimicking a subsurface reservoir, at high pressure and temperature. We determine pore occupancy during a displacement sequence that involves waterflooding, gas injection and water re-injection. In the water-wet sample considered, brine occupied the smallest pores, gas the biggest, while oil occupied pores of intermediate size and is displaced by both water and gas. Double displacement events have been observed, where gas displaces oil that displaces water or vice versa. The thickness of water and oil layers have been quantified, as have the contact angles between gas and oil, and oil and water. These results are used to explain the nature of trapping in three-phase flow, specifically how oil preferentially traps gas in the presence of water
Complement Activation Determines the Therapeutic Activity of Rituximab In Vivo
Rituximab is an anti-CD20 chimeric mAb effective for the treatment of B-NHL. It can lyse lymphoma cells in vitro through both C- and Ab-dependent cellular cytotoxicity. The mechanism of action of rituximab in vivo is however still unclear. We have set up a new in vivo model in nonimmunodeficient mice by stable transduction of the human CD20 cDNA in the murine lymphoma line EL4. Animals injected i.v. with the EL4-CD20+ lymphoma cells died within 30 days with evident liver, spleen, and bone marrow involvement, confirmed by immunohistochemistry and PCR analysis. A single injection of rituximab or the murine anti-CD20 Ab 1F5, given i.p. 1 day after the tumor, cured 100% of the animals. Indeed, at week 4 after tumor cell inoculation, CD20+ cells were undetectable in all organs analyzed in rituximab-treated animals, as determined by immunohistochemistry and PCR. Rituximab had no direct effect on tumor growth in vitro. Depletion of either NK cells or neutrophils or both in tumor-injected animals did not affect the therapeutic activity of the drug. Similarly, rituximab was able to eradicate tumor cells in athymic nude mice, suggesting that its activity is T cell independent. In contrast, the protective activity of rituximab or the 1F5 Ab was completely abolished in syngeneic knockout animals lacking C1q, the first component of the classical pathway of C (C1qa−/−). These data demonstrate that C activation is fundamental for rituximab therapeutic activity in vivo
Different vimentin expression in two clones derived from a human colocarcinoma cell line (LoVo) showing different sensitivity to doxorubicin.
We selected two clones, isolated from the human colocarcinoma cell line LoVo, showing a sensitivity to doxorubicin similar to (LoVo clone 5) or three times lower than (LoVo clone 7) the parental cell line. Since vimentin was atypically expressed in a human breast carcinoma cell line made resistant to doxorubicin, we looked at vimentin expression in these two clones with spontaneously different sensitivity to the drug. For comparison we used the parental cell line LoVo WT and LoVo/DX made resistant pharmacologically. mRNA for vimentin was undetectable by Northern blot analysis in LoVo WT and in LoVo clone 5, while expression of this gene was high in LoVo clone 7 and in LoVo/DX. This increase in mRNA levels was not related to an amplification of DNA, as suggested by Southern blot analysis. Immunofluorescence and immunocytochemistry findings confirmed, at protein level, the mRNA data. In LoVo clones 5 and 7, there were respectively 8.6% and 71% vimentin-positive cells, although the two clones showed similar expression of multidrug resistance gene 1 (mdr-1) and accumulated intracellular doxorubicin at similar levels. Similarly, drug efflux was the same for both clones. Our results show for the first time that cells resistant to doxorubicin express vimentin independently of the mdr glycoprotein. However when cells from clone 5 were transfected with human vimentin cDNA, they did not become resistant, indicating that vimentin can be considered as a marker of resistance in these cells but does not give rise to a resistant phenotype by itself
Multiplex staining depicts the immune infiltrate in colitis-induced colon cancer model
Assessment of the host immune response pattern is of increasing importance as highly prognostic and diagnostic, in immune-related diseases and in some types of cancer. Chronic inflammation is a major hallmark in colon cancer formation, but, despite the extent of local inflammatory infiltrate has been demonstrated to be extremely informative, its evaluation is not routinely assessed due to the complexity and limitations of classical immunohistochemistry (IHC). In the last years, technological advance helped in bypassing technical limits, setting up multiplex IHC (mIHC) based on tyramide signal amplification (TSA) method and designing software suited to aid pathologists in cell scoring analysis. Several studies verified the efficacy of this method, but they were restricted to the analysis of human samples. In the era of translational medicine the use of animal models to depict human pathologies, in a more complete and complex approach, is really crucial. Nevertheless, the optimization and validation of this method to species other than human is still poor. We took advantage of Multispectral Imaging System to identify the immunoprofile of Dextran Sulphate Sodium (DSS)-treated mouse colon. We optimized a protocol to sequentially stain formalin fixed paraffin embedded murine colon samples for CD3, CD8a, CD4, and CD4R5B0 antigens. With this approach we obtained a detailed lymphocyte profile, while preserving the morphological tissue context, generally lost with techniques like gene expression profiling or flow cytometry. This study, comparing the results obtained by mIHC with immunophenotyping performed with cytofluorimetric and standard IHC methods validates the potentiality and the applicability of this innovative approach
