    Influence of canopy fruit location on morphological, histochemical and biochemical changes in two oil olive cultivars

    The influence of different irradiance conditions was evaluated under natural solar radiation by comparing well-exposed (in) and shaded fruit (out) in canopies of olive trees (Olea europaea L). Over a 2-year period, from 50 days after full bloom up to harvest time, “in” and “out” olive samples of two genotypes (“Frantoio Millennio” and “Coratina 5/19”) were periodically collected. Morphological, histochemical, and biochemical analysis were performed to study the changes on fruit morphometric traits, oil body accumulation, and b-glucosidase enzyme activity. Some parameters were modified by shading inside the canopy in which the proportion of incident photosynthetically active radiation intercepted by the crop was 47%. Shaded fruits developed at slow rate and were characterized by late darkgoing time, reduced size, with a tendency toward oblong shape. The rapid histochemical procedure proposed to estimate the oil body accumulation during fruit ripening showed that a reduced irradiance caused a decrease in oil body density. The canopy position influenced, in a different way, the b-glucosidase activity in relation to the fruit-ripening stage in both genotypes. These findings indicate that providing an adequate and uniform lighting of the olive canopy by careful choices of orchard management practices can be a key factor for several yield components

    Photoperiodic Modulation of Circadian Clock and Reproductive Axis Gene Expression in the Pre-Pubertal European Sea Bass Brain

    The acquisition of reproductive competence requires the activation of the brain-pituitary-gonad (BPG) axis, which in most vertebrates, including fishes, is initiated by changes in photoperiod. In the European sea bass long-term exposure to continuous light (LL) alters the rhythm of reproductive hormones, delays spermatogenesis and reduces the incidence of precocious males. In contrast, an early shift from long to short photoperiod (AP) accelerates spermatogenesis. However, how photoperiod affects key genes in the brain to trigger the onset of puberty is still largely unknown. Here, we investigated if the integration of the light stimulus by clock proteins is sufficient to activate key genes that trigger the BPG axis in the European sea bass. We found that the clock genes clock, npas2, bmal1 and the BPG genes gnrh, kiss and kissr share conserved transcription factor frameworks in their promoters, suggesting co-regulation. Other gene promoters of the BGP axis were also predicted to be co-regulated by the same frameworks. Co-regulation was confirmed through gene expression analysis of brains from males exposed to LL or AP photoperiod compared to natural conditions: LL fish had suppressed gnrh1, kiss2, galr1b and esr1, while AP fish had stimulated npas2, gnrh1, gnrh2, kiss2, kiss1rb and galr1b compared to NP.     Effects of in vitro digestion on the antioxidant activity of three phenolic extracts from olive mill wastewaters

    The aim of this study was to assess the impact of in vitro digestion on the antioxidant activity of three extracts rich in phenols (two purified organic extracts (A20, A21) and one powdered extract stabilized with maltodextrins (SP)) obtained from olive mill wastewaters (OMWW). The content and composition of phenols and antioxidant activity was determined before and after in vitro digestion. The phenol content of the A20 and A21 samples were higher (>75%) than that of the SP sample before in vitro digestion. After the entire in vitro digestion, 89.3, 76.9, and 50% loss of phenols was found in A20, A21 and SP, respectively. ABTS•+ and ORAC values decreased during in vitro digestion of A20 and A21 samples, while they remained almost constant in SP. IC50 increased during digestion of A20 and A21, evidencing a loss of antioxidant capacity after the intestinal phase; an opposite IC50 trend was noted in SP, confirming the protective role of maltodextrins. For these reasons, SP represents a promising formulation to be used in the food field

    Growth, yield and oil quality of adult pedestrian olive orchards grown at four different planting systems

    This study evaluated growth, yield and olive oil quality of mature pedestrian olive orchards. Trees of three Sicilian cultivars Calatina, Nocellara del Belice and Abunara were planted at four combinations of planting densities and training forms. Trees at 2 x 5 m were trained to central leader (CLx2), those at 3 x 5 m to free palmette (FPx3), those at 4 x 5 to globe vase (GVx4), and those at 5 x 5 to poly-conic vase (PVx5). 'Calatina' had the smallest trees in terms of trunk size in all growing systems, while canopy size of trees at higher densities was similar for all three cultivars. 'Calatina' was also the most growth efficient (m3 of canopy per cm2 of TCSA) and produced the least amount of pruning wood in the hedgerow systems (CLx2 and FPx3). Fruit yield per tree tended to be higher in more vigorous cultivars (Abunara and Nocellara) grown to 3D systems (GVx4 and PVx5), while 'Calatina' was the most yield efficient (kg of fruit per cm2 of TCSA) especially in the hedgerow growing systems. Fruit and oil yield per ha and average production value tended to be highest in CLx2 trees and lowest in GVx4 trees, with 'Calatina' showing the sharpest changes and 'Nocellara' the smallest changes among growing systems. According to the Jaen index, CLx2 tended to induce earlier fruit maturation, followed by PVx5, GVx4, and FPx3. The growing system did not affect oil fatty acid composition, while 'Calatina' had the highest amount of mono-unsaturated fatty acids and the lowest amount of saturated fatty acids. 'Abunara' oils exhibited the highest amount of total phenols in CLx2, while 'Calatina' and 'Nocellara' oils exhibited the highest amount in FPx3 and PVx5. Both, trans-2-hexenal ("cut grass" sensory note) and hexenyl acetate ("floral" sensory note) tended to be lowest in oils from trees grown at CLx2 and highest in those from trees grown at GVx4, showing a somewhat inverse relationship with fruit ripening degree. The outcome of the present study on mature pedestrian orchards shows that proper combinations of cultivars, planting densities, and training forms (canopy shape) may result in efficient intensive systems for growing olive in areas where super-high density systems cannot be profitable due to agronomic and environmental limitations (water shortage, steep sloping sites, small farm size, etc.). Pedestrian growing systems can also be used to exploit olive biodiversity by allowing the use of available local genotypes. For this reason, they may represent an effective and sustainable solution against unexpected climate changes and associated emerging diseases

    Exploring contextual adaptations in caregiver interventions for families raising children with developmental disabilities

    There are increasing efforts to scale up services globally for families raising children with developmental disabilities (DDs). Existing interventions, often developed in high income, Western settings, need substantial adaptation before they can be implemented in different contexts.The aim of this study was to explore perspectives on the role that context plays in the adaptation and implementation of interventions targeting caregivers of children with DDs across settings. The study question was applied to the Caregiver Skills Training (CST) programme of the World Health Organization specifically, as well as to stakeholder experiences with caregiver interventions more broadly. Two focus group discussions (FGDs; n=15 participants) and 25 individual semi-structured interviews were conducted. Participants were caregivers of children with DDs and professionals involved in adapting or implementing the CST across five continents and different income settings. Data were analysed thematically.Four main themes were developed: 1) Setting the scene for adaptations; 2) Integrating an intervention into local public services; 3) Understanding the reality of caregivers; 4) Challenges of sustaining an intervention. Informants thought that contextual adaptations were key for the intervention to fit in locally, even more so than cultural factors. The socio-economic context of caregivers, including poverty, was highlighted as heavily affecting service access and engagement with the intervention. Competing health priorities other than DDs, financial constraints, and management of long-term collaborations were identified as barriers. This study validates the notion that attention to contextual factors is an essential part of the adaptation of caregiver interventions for children with DDs, by providing perspectives from different geographical regions. We recommend a stronger policy and research focus on contextual adaptations of interventions and addressing unmet socio-economic needs of caregivers.

    Effect of different irrigation volumes during fruit development on quality of virgin olive oil of cv. Frantoio

    An experiment was carried out in a high-density olive (Olea europaea L. cv. Frantoio) orchard to determine the effect of different irrigation regimes (full, deficit, complementary) on virgin olive oil (VOO) quality over three consecutive years. Irrigation had negligible effects on free acidity, peroxide value, and fatty acid composition of VOO, but strongly influenced its phenolic concentration. Trees with high water status yielded oils with lower concentrations of total phenols and O-diphenols with respect to oils from severely stressed trees. The concentrations of secoiridoids, like the dialdehydic form of decarboxymethyl elenolic acid linked to (3,4-dihydroxyphenyl)ethanol (3,4-DHPEA-EDA), the isomer of the oleuropein aglycon (3,4-DHPEA-EA) and the dialdehydic form of decarboxymethyl elenolic acid linked to (p-hydroxyphenyl) ethanol (p-HPEA-EDA), were lower in fully-irrigated trees than in trees under deficit irrigation or those that received complementary irrigation only. The concentrations of lignans (+)-1-acetoxipinoresinol and (+)-1-pinoresinol were unaffected by the irrigation regime. Volatile compounds, evaluated on the VOO head space, seemed to be more consistently influenced by the year rather than soil water availability

    Design and in-house validation of a portable system for the determination of free acidity in virgin olive oil

    Nutritional and healthy values are well known properties of virgin olive oil (VOO). The product quality, in terms of belonging to a specific quality grade (extra virgin, virgin, lampante), is defined by a set of chemical-physical and sensory measurements. According to the official regulation of the European Union (EU Reg. 1348/2013)the free acidity is the first parameter that has to be determined by analysts; it gives information about the quality of the olives used to produce the VOO as well as the hydrolytic state of VOO just produced and stored. The official procedure is based on an acid-base titration that needs to be carried out in a chemical laboratory. In this paper a portable battery-operated electronic system to measure olive oil free acidity is presented: the system can be used for quick \u201cin situ\u201d tests in a production environment (olive oil mills or packaging centers)by people without particular training. The working principle of the system is based on the creation of an emulsion between oil and a hydroalcoholic solution: the free acidity is estimated on the value of the emulsion electrical conductance. The proposed system has been calibrated and in-house validated showing good results in terms of limit of detection and quantification, precision and accuracy. Moreover, a good correlation (R 2adj = 0.97)with free acidity data obtained applying the official method on 30 olive oil samples belonging to different commercial categories (extra virgin, virgin and lampante olive oil)has been evidenced

    A regulated deficit irrigation strategy for hedgerow olive orchards with high plant density

    Background & Aims There is not a consensus on the best irrigation approach for super-high density (SHD) olive orchards. Our aim was to design and test a regulated deficit irrigation (RDI) strategy for a sustainable balance between water saving, tree vigour and oil production. Methods We tested our RDI strategy for 3 years in an ‘Arbequina’ orchard with 1,667 trees ha−1. Two levels of irrigation reduction were applied, 60RDI and 30RDI, scaled to replacing 60 % and 30 %, respectively, of the of irrigation needs (IN). We also had a full irrigation (FI) treatment as control, with IN totalling 4,701 m3 ha−1 Results The 30RDI treatment showed the best balance between water saving, tree vigour and oil production. With a yearly irrigation amount (IA) of 1,366 m3 ha−1, which meant 72 % water saving as compared to FI, the reduction in oil yield was 26 % only. Conclusions Our results, together with recent knowledge on the effect of water stress on fruit development, allowed us to suggest a potentially improved RDI strategy for which a total IA of ca. 2,100 m3 ha−1 was calculated. Both some management details and the benefits of this suggested RDI strategy are still to be tested
