Automated segmentation of tissue images for computerized IHC analysis
This paper presents two automated methods for the segmentation ofimmunohistochemical tissue images that overcome the limitations of themanual approach aswell as of the existing computerized techniques. The first independent method, based on unsupervised color clustering, recognizes automatically the target cancerous areas in the specimen and disregards the stroma; the second method, based on colors separation and morphological processing, exploits automated segmentation of the nuclear membranes of the cancerous cells. Extensive experimental results on real tissue images demonstrate the accuracy of our techniques compared to manual segmentations; additional experiments show that our techniques are more effective in immunohistochemical images than popular approaches based on supervised learning or active contours. The proposed procedure can be exploited for any applications that require tissues and cells exploration and to perform reliable and standardized measures of the activity of specific proteins involved in multi-factorial genetic pathologie
Progress towards an accurate determination of the Boltzmann constant by Doppler spectroscopy
In this paper, we present significant progress performed on an experiment
dedicated to the determination of the Boltzmann constant, k, by accurately
measuring the Doppler absorption profile of a line in a gas of ammonia at
thermal equilibrium. This optical method based on the first principles of
statistical mechanics is an alternative to the acoustical method which has led
to the unique determination of k published by the CODATA with a relative
accuracy of 1.7 ppm. We report on the first measurement of the Boltzmann
constant by laser spectroscopy with a statistical uncertainty below 10 ppm,
more specifically 6.4 ppm. This progress results from improvements in the
detection method and in the statistical treatment of the data. In addition, we
have recorded the hyperfine structure of the probed saQ(6,3) rovibrational line
of ammonia by saturation spectroscopy and thus determine very precisely the
induced 4.36 (2) ppm broadening of the absorption linewidth. We also show that,
in our well chosen experimental conditions, saturation effects have a
negligible impact on the linewidth. Finally, we draw the route to future
developments for an absolute determination of with an accuracy of a few ppm.Comment: 22 pages, 11 figure
Effect of mechanical consolidation on adhesion mechanism of hybrid non-woven Alfa fibers reinforced unsaturated polyester composite
Vibrational analysis based on FTIR measurements were performed on hybrid non-woven Alfa fibers reinforced unsaturated polyester composite (HNWAFRUP) and its constituents. The reinforcement was made up of Alfa and wool fibers in the relative volume fractions ratio 4:1. These non-woven fibers sheet were consolidated mechanically by means of the needle punching. This study revealed that the increase of passages numbers in this device allowed the separation of the Alfa fibers and the reduction of the lignins component in these fibers which decreased its hydrophilic character. Chemical reactions between wool and Alfa fibers were based mainly on the appearance of new vibrations originating from wool fibers in the reinforcement. Adhesion mechanism between the matrix and the reinforcement was established by chemical bonds formed by secondary bonding. Tensile testing performed on this composite revealed the enhancement of its mechanical properties attributed to a less fiber to fiber contact of wool fibers. Nevertheless, the decrease of its specific Youngs modulus revealed the mechanical consolidation effect on Alfa fibers/matrix adhesion
Comparative Study of Human and Automated Screening for Antinuclear Antibodies by Immunofluorescence on HEp-2 Cells
Background: Several automated systems had been developed in order to reduce inter-observer variability in
indirect immunofluorescence (IIF) interpretation. We aimed to evaluate the performance of a processing system in
antinuclear antibodies (ANA) screening on HEp-2 cells.
Patients and Methods: This study included 64 ANA-positive sera and 107 ANA-negative sera that underwent IIF on two
commercial kits of HEp-2 cells (BioSystems® and Euroimmun®). IIF results were compared with a novel automated
interpretation system, the “CyclopusCADImmuno®” (CAD).
Results: All ANA-positive sera images were recognized as positive by CAD (sensitivity = 100%), while 17 (15.9%) of the
ANA-negative sera images were interpreted as positive (specificity = 84.1%), =0.799 (SD=0.045). Comparison of IIF
pattern determination between human and CAD system revealed on HEp-2 (BioSystems®), a complete concordance in
6 (9.37%) sera, a partial concordance (sharing of at least 1 pattern) in 42 (65.6%) cases and in 16 (25%) sera the
pattern interpretation was discordant. Similarly, on HEp-2 (Euroimmun®) the concordance in pattern interpretation was
total in 5 (7.8%) sera, partial in 39 (60.9%) and absent in 20 (31.25%). For both tested HEp-2 cells kits agreement was
enhanced for the most common patterns, homogenous, fine speckled and coarse speckled. While there was an issue in
identification of nucleolar, dots and nuclear membranous patterns by CAD.
Conclusion: Assessment of ANA by IIF on HEp-2 cells using the automated interpretation system, the
“CyclopusCADImmuno®” is a reliable method for positive/negative differentiation. Continuous integration of IIF images
would improve the pattern identification by the CAD
Mise à jour 2014 des recommandations du GEFPICS pour l’évaluation du statut HER2 dans les cancers du sein en France
De nouvelles recommandations internationales pour l’évaluation du statut HER2 dans les cancers du sein, basées sur plus de dix ans d’expérience et sur les résultats d’études cliniques et de concordance entre les différentes techniques de détection, viennent tout juste de voir le jour. Le présent article a pour objet de faire le point sur ces nouvelles recommandations, à la lumière de la publication récente du groupe de travail de l’American Society of Clinical Oncology (ASCO) et du Collège des pathologistes américains (CAP), adaptées à la pratique de la pathologie en France et revues par le groupe GEFPICS. À l’ère de la médecine personnalisée, la détermination du statut HER2 reste un élément phare dans le panel des biomarqueurs théranostiques des cancers du sein. Si l’interprétation du statut HER2 dans les cancers du sein est aisée dans la majorité des cas, un certain nombre de situations anatomocliniques est d’interprétation plus délicate, telles que la possibilité rare mais réelle de l’hétérogénéité intra-tumorale du statut de HER2, les formes à différenciation micropapillaire ou la ré-évaluation du statut des biomarqueurs lors de la rechute métastatique. Ces nouvelles recommandations abordent ces différentes questions, reprécisent les conditions pré-analytiques optimales et les critères d’interprétation (notamment des cas 2+), afin de réduire au maximum le risque de faux négatifs. Plus que jamais, la mobilisation de la spécialité d’anatomo-cytopathologie autour de la qualité des tests théranostiques témoigne de son implication dans la chaîne des soins en cancérologie., Summary International guidelines on HER2 determination in breast cancer have just been updated by the American Society of Clinical Oncology (ASCO) and College of American Pathologists (CAP), on the basis of more than ten-year practice, results of clinical trials and concordance studies. The GEFPICS group, composed of expert pathologists in breast cancer, herein presents these recommendations, adapted to the French routine practice. These guidelines highlight the possible diagnosis difficulties with regards to HER2 status determination, such as intra-tumor heterogeneity, special histological subtypes and biomarker re-evaluation during metastatic relapse. Pre-analytical issues and updated scoring criteria (especially for equivocal cases) are detailed, in order to decrease the occurrence of false negative cases. In the era of personalized medicine, pathologists are more than ever involved in the quality of oncotheranostic biomarker evaluation.
Mise à jour des recommandations du GEFPICS pour l’évaluation du statut HER2 dans les cancers du sein en France
En Europe, les patientes atteintes d’un cancer du sein invasif susceptibles de recevoir un traitement ciblé anti-HER2 sont actuellement sélectionnées sur la base d’un test immunohistochimique (IHC). Les techniques d’hybridation in situ (HIS) doivent être utilisées pour l’évaluation des cas IHC ambigus (2+) et pour l’étalonnage de la technique IHC. Les patientes éligibles au traitement ciblant HER2 présentent un statut HER2 positif défini par un test IHC 3+ ou un test 2+ amplifié. Une détection correcte du statut HER2 est indispensable à une utilisation optimale des thérapeutiques ciblées puisque leur efficacité est limitée aux patientes surexprimant HER2. Il est capital que l’évaluation du statut HER2 soit optimisée et fiable. Ces recommandations du groupe d’étude des facteurs pronostiques IHC dans le cancer du sein (GEFPICS) détaillent et commentent les différentes étapes des techniques IHC et HIS, les contrôles utilisables et les règles générales de l’apprentissage de la lecture. Une fois acquis, ce savoir-faire doit être pérennisé par l’observation de règles de bonnes pratiques techniques (utilisation rigoureuse de témoins internes et externes et participation régulière à des programmes d’Assurance qualité [AQ])., Summary In Europe, patients who may benefit from an HER2 targeted drug are currently selected by immunohistochemistry (IHC). In situ hybridization (ISH) techniques should be used for complementary assessment of ambiguous 2+ IHC cases and for the calibration of the IHC technique. Eligibility to an HER2 target treatment is defined by an HER2 positive status being IHC test 3+ or 2+ amplified. Reliable detection of HER2 status is essential to the appropriate usage of HER2 targeted drugs because its specificity is limited to tumors overexpressing HER2. It is essential that the IHC evaluation of the HER2 status of a mammary carcinoma is optimized and reliable. This GEFPICS’ guidelines look over the different steps of the IHC technique, the controls and, the rules for interpretation. Once acquired, this knowledge must be perpetuated by the observation of rules of good technical practice (internal and external controls, quality assurance programs)

Currently the climate computer offers many benefits and solves problems related to the regulation, monitoring and controls. Greenhouse growers remain vigilant and attentive, facing this technological development. they ensure competitiveness and optimize their investments / production cost which continues to grow. The application of artificial intelligence in the industry known for considerable growth, which is not the case in the field of agricultural greenhouses, where enforcement remains timid. it is from this fact, we undertake research work in this area and conduct a simulation based on meteorological data through MATLAB Simulink to finally analyze the thermal behavior -greenhouse microclimate energy . In this paper we present comparison of modeling and simulation management of the greenhouse microclimate by fuzzy logic between a wetland (Dar El Beida Algeria) and the other arid (Biskra Algeria)
Sterol composition of caper (Capparis spinosa) seeds
Caper is a perennial shrub of the Mediterranean Basin. The most important economical species is Capparis spinosa. Sterols of C. spinosa seed oil isolated from seven Tunisian stands were identified andquantified. C. spinosa contained  high levels of phytosterols (2240.4 mg/kg of total extracted lipids), of which -sitosterol, with 1390 mg/kg, was the most abundant (57.53%). Campesterol and stigmasterol accounted for 382 and 265 mg/kg, respectively (17.05 and 11.85% of the total sterols, respectively). C. spinosa seed oil also contained a high level of 5-avenasterol (6%). We detect also brassicasterol (3.39 mg/kg). Cholesterol and campestanol are detected in much lower levels. These results bring attention to the richness of C.spinosa seed oil with sterols which are the most important class of the minor components
Prevalence and risk factors of low back pain among undergraduate students of a sports and physical education institute in Tunisia
Introduction: For obvious reasons, athletes are at greater risk of sustaining a lumber (lower) spine injury due to physical activity. To our knowledge, no previous studies have examined the prevalence of low back pain (LBP) in a Tunisian sports and physical education institute.Aim: To assess the prevalence of LBP in different sports among students studying in a sports and physical education institute in Tunisia, to determine the causes of the injuries, and to propose solutions.Methods: A total of 3,379 boys and 2,579 girls were studied. A retrospective cross-sectional survey was conducted on a group of students aged 18.524.5 years at the Higher Institute of Sport and Physical Education of Sfax to estimate the prevalence of LBP and its relation to the type of sports. Data on age, weight, height, smoking, and the sport in which the student was injured in the low back were collected from the institute health service records from 2005 until 2013.Results: LBP was reported by 879 of the 5,958 study participants (14.8%). The prevalence of LBP was significantly higher (pB0.001) in females (17.6%) than in males (12.5%). LBP prevalence did not differ by body mass index or smoking habit (p0.05). The sports associated with the higher rates of LBP were gymnastics, judo, handball, and volleyball, followed by basketball and athletics.Conclusion: LBP is frequent among undergraduate students of a sports and physical education institute in Tunisia. It is strongly associated with fatigue after the long periods of training in different sports. Gymnastics, judo, handball, and volleyball were identified as high-risk sports for causing LBP.Keywords: low back pain; sports students; sports training; risk factor
