    Membrane glucocorticoid receptors are localised in the extracellular matrix and signal through the MAPK pathway in mammalian skeletal muscle fibres

    A number of studies have previously proposed the existence of glucocorticoid receptors on the plasma membrane of many cell types including skeletal muscle fibres. However, their exact localisation and the cellular signalling pathway(s) they utilise to communicate with the rest of the cell are still poorly understood. In this study, we investigated the localisation and the mechanism(s) underlying the non-genomic physiological functions of these receptors in mouse skeletal muscle cells. The results show that the receptors were localised in the cytoplasm in myoblasts, in the nucleus in myotubes and in the extracellular matrix, in satellite cells and in the proximity of mitochondria in adult muscle fibres. Also, they bound laminin in a glucocorticoid-dependent manner. Treating small skeletal muscle fibre bundles with the synthetic glucocorticoid, beclomethasone dipropionate, increased the phosphorylation (=activation) of extracellular-signal regulated kinase 1&2, c-Jun N-terminal kinase and p38 mitogen-activated protein kinase. This occurred within 5min and depended on the fibre-type and the duration of the treatment. It was also abolished by the glucocorticoid receptor inhibitor, mifepristone, and a monoclonal antibody against the receptor. From these results we conclude that the non-genomic/non-canonical physiological functions of glucocorticoids, in adult skeletal muscle fibres are mediated by a glucocorticoid receptor localised in the extracellular matrix, in satellite cells and close to mitochondria and involve activation of the MAPK pathway

    Interest Rate Determination in the Interbank Market

    The purpose of this paper is to study the determinants of equilibrium in the market for daily funds. We use the EONIA panel database which includes daily information on the lending rates applied by contributing commercial banks. The data clearly shows an increase in both the time series volatility and the cross section dispersion of rates towards the end of the reserve maintenance period. These increases are highly correlated. With respect to quantities, we find that the volume of trade as well as the use of the standing facilities are also larger at the end of the maintenance period. Our theoretical model shows how the operational framework of monetary policy causes a reduction in the elasticity of the supply of funds by banks throughout the reserve maintenance period. This reduction in the elasticity together with market segmentation and heterogeneity are able to generate distributions for the interest rates and quantities traded with the same properties as in the data.Overnight interest rate; Monetary policy instruments; Eonia panel

    Carrier relaxation mechanisms in self-assembled (In,Ga)As/GaAs quantum dots: Efficient P -> S Auger relaxation of electrons

    We calculate the P-shell--to-S-shell decay lifetime \tau(P->S) of electrons in lens-shaped self-assembled (In,Ga)As/GaAs dots due to Auger electron-hole scattering within an atomistic pseudopotential-based approach. We find that this relaxation mechanism leads to fast decay of \tau(P->S)~1-7 ps for dots of different sizes. Our calculated Auger-type P-shell--to-S-shell decay lifetimes \tau(P->S) compare well to data in (In,Ga)As/GaAs dots, showing that as long as holes are present there is no need for an alternative polaron mechanism.Comment: Version published in Phys.     Equation of state in the fugacity format for the two-dimensional Coulomb gas

    We derive the exact general form of the equation of state, in the fugacity format, for the two-dimensional Coulomb gas. Our results are valid in the conducting phase of the Coulomb gas, for temperatures above the Kosterlitz-Thouless transition. The derivation of the equation of state is based on the knowledge of the general form of the short-distance expansion of the correlation functions of the Coulomb gas. We explicitly compute the expansion up to order O(ζ6)O(\zeta^6) in the activity ζ\zeta. Our results are in very good agreement with Monte Carlo simulations at very low density

    Prevalence of anginal symptoms and myocardial ischemia and their effect on clinical outcomes in outpatients with stable coronary artery disease: data from the international observational CLARIFY registry

    Importance: In the era of widespread revascularization and effective antianginals, the prevalence and prognostic effect of anginal symptoms and myocardial ischemia among patients with stable coronary artery disease (CAD) are unknown.<p></p> Objective: To describe the current clinical patterns among patients with stable CAD and the association of anginal symptoms or myocardial ischemia with clinical outcomes.<p></p> Design, Setting, and Participants: The Prospective Observational Longitudinal Registry of Patients With Stable Coronary Artery Disease (CLARIFY) registry enrolled outpatients in 45 countries with stable CAD in 2009 to 2010 with 2-year follow-up (median, 24.1 months; range, 1 day to 3 years). Enrollees included 32 105 outpatients with prior myocardial infarction, chest pain, and evidence of myocardial ischemia, evidence of CAD on angiography, or prior revascularization. Of these, 20 291 (63.2%) had undergone a noninvasive test for myocardial ischemia within 12 months of enrollment and were categorized into one of the following 4 groups: no angina or ischemia (n = 13 207 [65.1%]); evidence of myocardial ischemia without angina (silent ischemia) (n = 3028 [14.9%]); anginal symptoms alone (n = 1842 [9.1%]); and angina and ischemia (n = 2214 [10.9%]).<p></p> Exposures: Stable CAD.<p></p> Main Outcome and Measure: The composite of cardiovascular (CV)–related death or nonfatal myocardial infarction.<p></p> Results: Overall, 4056 patients (20.0%) had anginal symptoms and 5242 (25.8%) had evidence of myocardial ischemia on results of noninvasive testing. Of 469 CV-related deaths or myocardial infarctions, 58.2% occurred in patients without angina or ischemia, 12.4% in patients with ischemia alone, 12.2% in patients with angina alone, and 17.3% in patients with both. The hazard ratios for the primary outcome relative to patients without angina or ischemia and adjusted for age, sex, geographic region, smoking status, hypertension, diabetes mellitus, and dyslipidemia were 0.90 (95% CI, 0.68-1.20; P = .47) for ischemia alone, 1.45 (95% CI, 1.08-1.95; P = .01) for angina alone, and 1.75 (95% CI, 1.34-2.29; P <.001) for both. Similar findings were observed for CV-related death and for fatal or nonfatal myocardial infarction.<p></p> Conclusions and Relevance: In outpatients with stable CAD, anginal symptoms (with or without ischemia on noninvasive testing) but not silent ischemia appear to be associated with an increased risk for adverse CV outcomes. Most CV events occurred in patients without angina or ischemia

    Secondary user relations in emerging mobile computing environments

    Mobile technologies are enabling access to information in diverse environ.ments, and are exposing a wider group of individuals to said technology. Therefore, this paper proposes that a wider view of user relations than is usually considered in information systems research is required. Specifically, we examine the potential effects of emerging mobile technologies on end-­‐user relations with a focus on the ‘secondary user’, those who are not intended to interact directly with the technology but are intended consumers of the technology’s output. For illustration, we draw on a study of a U.K. regional Fire and Rescue Service and deconstruct mobile technology use at Fire Service incidents. Our findings provide insights, which suggest that, because of the nature of mobile technologies and their context of use, secondary user relations in such emerging mobile environments are important and need further exploration

    Guest charges in an electrolyte: renormalized charge, long- and short-distance behavior of the electric potential and density profile

    We complement a recent exact study by L. Samaj on the properties of a guest charge QQ immersed in a two-dimensional electrolyte with charges +1/1+1/-1. In particular, we are interested in the behavior of the density profiles and electric potential created by the charge and the electrolyte, and in the determination of the renormalized charge which is obtained from the long-distance asymptotics of the electric potential. In Samaj's previous work, exact results for arbitrary coulombic coupling β\beta were obtained for a system where all the charges are points, provided βQ<2\beta Q<2 and β<2\beta < 2. Here, we first focus on the mean field situation which we believe describes correctly the limit β0\beta\to 0 but βQ\beta Q large. In this limit we can study the case when the guest charge is a hard disk and its charge is above the collapse value βQ>2\beta Q>2. We compare our results for the renormalized charge with the exact predictions and we test on a solid ground some conjectures of the previous study. Our study shows that the exact formulas obtained by Samaj for the renormalized charge are not valid for βQ>2\beta Q>2, contrary to a hypothesis put forward by Samaj. We also determine the short-distance asymptotics of the density profiles of the coions and counterions near the guest charge, for arbitrary coulombic coupling. We show that the coion density profile exhibit a change of behavior if the guest charge becomes large enough (βQ2β\beta Q\geq 2-\beta). This is interpreted as a first step of the counterion condensation (for large coulombic coupling), the second step taking place at the usual Manning--Oosawa threshold βQ=2\beta Q=2
