    Modelling and Interpreting The Effects of Spatial Resolution on Solar Magnetic Field Maps

    Different methods for simulating the effects of spatial resolution on magnetic field maps are compared, including those commonly used for inter-instrument comparisons. The investigation first uses synthetic data, and the results are confirmed with {\it Hinode}/SpectroPolarimeter data. Four methods are examined, one which manipulates the Stokes spectra to simulate spatial-resolution degradation, and three "post-facto" methods where the magnetic field maps are manipulated directly. Throughout, statistical comparisons of the degraded maps with the originals serve to quantify the outcomes. Overall, we find that areas with inferred magnetic fill fractions close to unity may be insensitive to optical spatial resolution; areas of sub-unity fill fractions are very sensitive. Trends with worsening spatial resolution can include increased average field strength, lower total flux, and a field vector oriented closer to the line of sight. Further-derived quantities such as vertical current density show variations even in areas of high average magnetic fill-fraction. In short, unresolved maps fail to represent the distribution of the underlying unresolved fields, and the "post-facto" methods generally do not reproduce the effects of a smaller telescope aperture. It is argued that selecting a method in order to reconcile disparate spatial resolution effects should depend on the goal, as one method may better preserve the field distribution, while another can reproduce spatial resolution degradation.     O relatório resulta do trabalho de avaliação ao curso de Engenharia de Ordenamento dos Recursos Naturais levado a cabo pela equipa de Auto-Avaliação da Escola Superior Agrária. Esta equipa, designada pelo Conselho Científico com a finalidade de proceder a todas as solicitações de avaliação tanto dos cursos como da instituição, é basicamente composta por um coordenador e um elemento de cada unidade departamental. Decorrente do tipo de curso que se está a avaliar juntam-se à equipa os respectivos coordenadores do curso e um relator. Além deste elementos participaram também neste trabalho um representante dos alunos (designado pela associação de estudantes) e dois funcionários representantes do pessoal administrativo e do pessoal não docente. Em termos metodológicos optou-se pelo seguimento do guião proposto pelo CNAVES, tendo-se recolhido informação de diversos modos: pesquisa documental e bases de dados nos serviços administrativos, inquéritos auto administrados a alunos, docentes e funcionários, inquérito postal aos diplomados e entrevistas directas às entidades empregadoras. Uma vez recolhida e tratada a informação procedeu-se à sua análise crítica tendo como referência os trabalhos de avaliação que até então decorreram relativamente à Escola e ao curso de Ordenamento dos Recursos Naturais

    Estudo da Variação Intradiária das Concentrações de Pólen de Gramíneas na Atmosfera de Portugal Continental

    Introdução: O pólen da família das Poaceae (gramíneas) é uma das principais fontes de aeroalergénios no mundo, particularmente na Europa Mediterrânica. Representa, por isso, um importante factor de risco de asma, rinite e/ou conjuntivite alérgica e constitui a principal causa de polinose em Portugal. Objectivo: Analisar a variação intradiária das concentrações de pólen de gramíneas na atmosfera das 5 estações de monitorização continentais da Rede Portuguesa de Aerobiologia (RPA): Porto, Coimbra, Lisboa, Évora e Portimão. Métodos: Neste estudo utilizaram -se os dados diários e horários das monitorizações de pólen de gramíneas das cinco estações de monitorização continentais da RPA, ao longo de 7 anos (2002 -2008). Resultados: Entre as localidades encontraram -se diferenças significativas, em termos de Estudo da variação intradiária das concentrações de pólen de gramíneas na atmosfera de Portugal Continental O pólen atmosférico de gramíneas constitui a principal causa de rinite, asma, conjuntivite e eczema nos indivíduos alérgicos ao pólen na área Mediterrânica1,2, nomeadamente em Portugal3,4. Dado que o pólen, particularmente o pólen de gramíneas, é um factor de risco para as doenças alérgicas respiratórias exercendo um impacto negativo sobre a qualidade de vida dos indivíduos sensibilizados, é de particular interesse, quer dos profissionais de saúde, quer do doente conhecer a sua distribuição intradiária, ou seja a variação horária das concentrações de pólen de gramíneas ao longo do dia, de modo a adequar de forma mais eficaz medidas de evicção e de intervenção terapêutica. curvas horárias. O pólen encontrou -se presente na atmosfera durante 24 horas em todas as localidades, e os valores das concentrações horárias variaram ao longo do dia e de ano para ano. As concentrações mais baixas registaram -se entre as 22 e as 6 horas e as mais elevadas, entre as 7 e as 21 horas, as quais em Évora ultrapassaram os 30 grãos de pólen/m3/hora. Em geral, registaram -se 2 picos de concentrações máximas, um de manhã (9 -10 horas) ou à tarde (12 -13 horas) e outro no final da tarde / início da noite (19 -20 horas). Conclusões: O ritmo diurno difere muito de local para local. Cada localidade tem o seu próprio padrão de variação das concentrações horárias do pólen atmosférico de gramíneas que se pode dever, quer às diferentes espécies presentes, quer às diferentes condições ambientais. O risco de exposição variou de localidade para localidade e ao longo do dia, sendo o Porto a localidade onde este é menor, enquanto Évora apresenta o maior risco

    Semiclassical quantization of the hydrogen atom in crossed electric and magnetic fields

    The S-matrix theory formulation of closed-orbit theory recently proposed by Granger and Greene is extended to atoms in crossed electric and magnetic fields. We then present a semiclassical quantization of the hydrogen atom in crossed fields, which succeeds in resolving individual lines in the spectrum, but is restricted to the strongest lines of each n-manifold. By means of a detailed semiclassical analysis of the quantum spectrum, we demonstrate that it is the abundance of bifurcations of closed orbits that precludes the resolution of finer details. They necessitate the inclusion of uniform semiclassical approximations into the quantization process. Uniform approximations for the generic types of closed-orbit bifurcation are derived, and a general method for including them in a high-resolution semiclassical quantization is devised

    Tomando como pano de fundo as recomendações da International Network of Agencies for Health Technology Assessment (INAHTA) e as propostas de trabalho do Danish Centre of Health Technology Assessment - National Board of Health, os autores colocam em ênfase que a avaliação em tecnologias da saúde (ATS) é um campo de estudos multidisciplinares, cujos resultados comportam diversas implicações clínicas, sociais, culturais, políticas, éticas e económicas no domínio do desenvolvimento, difusão e utilização das tecnologias na prestação dos cuidados de saúde. Refutando a mera ideia de que a ATS seja um veículo instrumentalizado para fornecer aos decisores políticos informações sobre os possíveis impactos e as consequências de uma nova tecnologia nos cuidados de saúde, os autores argumentam que é necessário refletir e debater sobre os critérios de ATS, sugerindo para o efeito a adoção dum modelo plural e multidisciplinar do tipo do modelo dinamarquês, tendo o cidadão-utente no centro do sistema de avaliação em tecnologias da saúde

[email protected] Abstract Although a number of studies on the phenotypic changes that occur after T-cell activation have already been published, the specific immunophenotypic features of T-lymphocytes and the frequency at which TCR-variable region (TCR-V) restricted T-cell expansions occur "in vivo" during acute viral infection still remains to be established. We report on the immunophenotype and TCR-V repertoire of peripheral blood T-cells from 28 patients with acute infectious mononucleosis. Immunophenotypic studies were performed by flow cytometry using direct immunofluorescence techniques and stain-and-then-lyse sample preparation protocols with three- and four-colour combinations of monoclonal antibodies directed against a large panel of T- and NK-cell associated markers, activation- and adhesion-related molecules and TCR-Vbeta, -Vgamma and -Vdelta families. Nearly all patients (27/28) showed a massive expansion of CD8(+)/TCRalphabeta(+) T cells, the majority (>90%) of which displayed an immunophenotype compatible with T-cell activation: CD2(+high), CD7(+low), CD11a(+high), CD38(+high), HLA-DR(+high), CD28(+/-low), CD45RO(+high), CD45RA(-/+low), CD11b(-/+low), CD11c(+/-low), CD16(-), CD56(-), CD57(-), CD62L(-), CD94(-), CD158a(-), CD161(-), NKB1(-). Additionally, the levels of both CD3 and CD5 were slightly decreased compared to those found in normal individuals. Late-activation antigens, such as CD57, were found in small proportions of CD8(+)/TCRalphabeta(+) T-cells. Increased numbers of CD4(+)/TCRalphabeta(+) T-cells, TCRgammadelta(+) T-cells and NK-cells were also noticed in 17, 16 and 13 of the 28 cases studied, respectively. Evidence for activation of CD4(+)/TCRalphabeta(+) and TCRgammadelta(+) T-cells relied on changes similar to those described for CD8(+)/TCRalphabeta(+) although less pronounced, except for higher levels of both CD5 and CD28 in the absence of reactivity for CD11c on CD4(+)/TCRalphabeta(+) T-cells and higher levels of CD161 and CD94 on TCRgammadelta(+) T-cells. Small expansions of one or more TCR-Vbeta families accounting for 12 +/- 7% of either the CD8(+)/TCRalphabeta(+) or the CD4(+)/TCRalphabeta(+) T-cell compartment were found in 12 of 14 patients studied, whereas the distribution of the TCR-Vgamma and -Vdelta repertoires tested in 2 of the individuals with expanded TCRgammadelta(+) T-cells was similar to that observed in control individuals. The results presented here provide evidence for an extensive T-cell activation during acute viral infection and establish the immunophenotype patterns associated with this condition. Abstract Indolent natural killer (NK) cell lymphoproliferative disorders include a heterogeneous group of patients in whom persistent expansions of mature, typically CD56(+), NK cells in the absence of any clonal marker are present in the peripheral blood. In the present study we report on the clinical, hematological, immunophenotypic, serological, and molecular features of a series of 26 patients with chronic large granular NK cell lymphocytosis, whose NK cells were either CD56(-) or expressed very low levels of CD56 (CD56(-/+dim) NK cells), in the context of an aberrant activation-related mature phenotype and proved to be monoclonal using the human androgen receptor gene polymerase chain reaction-based assay. As normal CD56(+) NK cells, CD56(-/+dim) NK cells were granzyme B(+), CD3(-), TCRalphabeta/gammadelta(-), CD5(-), CD28(-), CD11a(+bright), CD45RA(+bright), CD122(+), and CD25(-) and they showed variable and heterogeneous expression of both CD8 and CD57. Nevertheless, they displayed several unusual immunophenotypic features. Accordingly, besides being CD56(-/+dim), they were CD11b(-/+dim) (heterogeneous), CD7(-/+dim) (heterogeneous), CD2(+) (homogeneous), CD11c(+bright) (homogeneous), and CD38(-/+dim) (heterogeneous). Moreover, CD56(-/+dim) NK cells heterogeneously expressed HLA-DR. In that concerning the expression of killer receptors, CD56(-/+dim) NK cells showed bright and homogeneous CD94 expression, and dim and heterogeneous reactivity for CD161, whereas CD158a and NKB1 expression was variable. From the functional point of view, CD56(-/+dim) showed a typical Th1 pattern of cytokine production (interferon-gamma(+), tumor necrosis factor-alpha(+)). From the clinical point of view, these patients usually had an indolent clinical course, progression into a massive lymphocytosis with lung infiltration leading to death being observed in only one case. Despite this, they frequently had associated cytopenias as well as neoplastic diseases and/or viral infections. In summary, we describe a unique and homogeneous group of monoclonal chronic large granular NK cell lymphocytosis with an aberrant activation-related CD56(-/+dim)/CD11b(-/+dim) phenotype and an indolent clinical course, whose main clinical features are related to concomitant diseases.
