The CCHamide 1 receptor modulates sensory perception and olfactory behavior in starved Drosophila
The olfactory response of the vinegar fly Drosophila melanogaster to food odor is modulated by starvation. Here we show that this modulation is not restricted to food odors and their detecting sensory neurons but rather increases the behavioral response to odors as different as food odors, repellents and pheromones. The increased behavioral responsiveness is paralleled by an increased physiological sensitivity of sensory neurons regardless whether they express olfactory or ionotropic receptors and regardless whether they are housed in basiconic, coeloconic, or trichoid sensilla. Silencing several genes that become up-regulated under starvation confirmed the involvement of the short neuropeptide f receptor in the starvation effect. In addition it revealed that the CCHamide-1 receptor is another important factor governing starvation-induced olfactory modifications
Host Plant Species Differentiation in a Polyphagous Moth: Olfaction is Enough
International audiencePolyphagous herbivorous insects need to discriminate suitable from unsuitable host plants in complex plant communities. While studies on the olfactory system of monophagous herbivores have revealed close adaptations to their host plant's characteristic volatiles, such adaptive fine-tuning is not possible when a large diversity of plants is suitable. Instead, the available literature on polyphagous herbivore preferences suggests a higher level of plasticity, and a bias towards previously experienced plant species. It is therefore necessary to take into account the diversity of plant odors that polyphagous herbivores encounter in the wild in order to unravel the olfactory basis of their host plant choice behaviour. In this study we show that a polyphagous moth, Spodoptera littoralis, has the sensory ability to distinguish five host plant species using olfaction alone, this being a prerequisite to the ability to make a choice. We have used gas chromatography mass spectrometry (GC-MS) and gas chromatography electroantennographic detection (GC-EAD) in order to describe host plant odor profiles as perceived by S. littoralis. We find that each plant emits specific combinations and proportions of GC-EAD active volatiles, leading to statistically distinct profiles. In addition, at least four of these plants show GC-EAD active compound proportions that are conserved across individual plants, a characteristic that enables insects to act upon previous olfactory experiences during host plant choice. By identifying the volatiles involved in olfactory differentiation of alternative host plants by Spodoptera littoralis, we set the groundwork for deeper investigations of how olfactory perceptions translate into behaviour in polyphagous herbivores
Brief sensory experience differentially affects the volume of olfactory brain centres in a moth
International audienceExperience modifies behaviour in animals so that they adapt to their environment. In male noctuid moths, Spodoptera littoralis, brief pre-exposure to various behaviourally relevant sensory signals modifies subsequent behaviour towards the same or different sensory modalities. Correlated with a behavioural increase in responses of male moths to the female-emitted sex pheromone after pre-exposure to olfactory, acoustic or gustatory stimuli, an increase in sensitivity of olfactory neurons within the primary olfactory centre, the antennal lobe, is found for olfactory and acoustic stimuli, but not for gustatory stimuli. Here, we investigated whether anatomical changes occurring in the antennal lobes and in the mushroom bodies (the secondary olfactory centres) possibly correlated with the changes observed in behaviour and in olfactory neuron physiology. Our results showed that significant volume changes occurred in glomeruli (olfactory units) responsive to sex pheromone following exposure to both pheromone and predator sounds. The volume of the mushroom body input region (calyx) also increased significantly after pheromone and predator sound treatment. However, we found no changes in the volume of antennal lobe glomeruli or of the mushroom body calyx after pre-exposure to sucrose. These findings show a relationship of antennal lobe sensitivity changes to the pheromone with changes in the volume of the related glomeruli and the output area of antennal lobe projection neurons elicited by sensory cues causing a behavioural change. Behavioural changes observed after sucrose pre-exposure must originate from changes in higher integration centres in the brain.</p
The Scent of the Fly
(Z)-4-undecenal (Z4-11Al) is the volatile pheromone produced by females of the vinegar fly Drosophila melanogaster. Female flies emit Z4-11Al for species-specific communication and mate-finding. A sensory panel finds that synthetic Z4-11Al has a characteristic flavour, which can be perceived even at the small amounts produced by a single female fly. Since only females produce Z4-11Al, and not males, we can reliably distinguish between single D. melanogaster males and females, according to their scent. Females release Z4-11Al at 2.4 ng/h and we readily sense 1 ng synthetic Z4-11Al in a glass of wine (0.03 nmol/L), while a tenfold concentration is perceived as a loud off-flavour. This corroborates the observation that a glass of wine is spoilt by a single D. melanogaster fly falling into it, which we here show is caused by Z4-11Al. The biological role of Z4-11Al or structurally related aldehydes in humans and the basis for this semiochemical convergence remains yet unclear.
