Slow troponin C gene expression in chicken heart and liver is regulated by similar enhancers
AbstractTwo isoforms of troponin C (TnC) are encoded by distinct single copy genes. Expression of fast TnC is restricted to the skeletal muscle, whereas the slow isoform is expressed in both skeletal and cardiac muscle. Chicken slow TnC (cTnC) gene is also expressed in some non-muscle tissues like the liver and the brain. Expression of cTnC gene is regulated by two distinct enhancers in cardiac and skeletal muscles. The cardiac specific enhancer is located in the immediate 5′ flanking region (bp −124 to −79) of the murine cTnC gene whereas the skeletal enhancer is located within the first intron (bp 997 to 1141). In the present study we have examined how cTnC gene expression is regulated in the chicken liver. Transient transfection of liver cells with cTnC-CAT reporter constructs containing various regions of the murine cTnC gene showed that its expression in chicken liver is regulated by the cardiac specific enhancer. Furthermore, electrophoretic mobility shift assays using synthetic oligonucleotides corresponding to both CEF-1 and CEF-2 regions of the murine cardiac enhancer revealed formation of specific DNA-protein complexes. Ultraviolet light induced covalent linking of nuclear proteins to CEF-1 and CEF-2 oligomers were used to examine the nature of the cardiac enhancer binding polypeptides; one polypeptide of 48 kDa appeared to bind to both CEF-1 and CEF-2 sequences

Platelet refractoriness in acquired hemophagocytic syndrome
BACKGROUND: Acquired hemophagocytic syndrome (AHPS) is a severe inflammatory disorder often caused by Epstein-Barr virus (EBV). Proliferation of activated macrophages produces uncontrolled cytokine production. Thrombocytopenia is common in AHPS, previously attributed to inadequate or ineffective marrow platelet (PLT) production. PLT transfusion response is not well reported. Two patients with fatal AHPS developed unexplained PLT transfusion refractoriness before definitive diagnosis.
CASE REPORTS: PLT refractoriness was noted during the care of two patients. The refractoriness was determined to be nonimmune and both demonstrated various clinical signs and laboratory findings consistent with AHPS. The first patient\u27s AHPS was attributable to EBV infection. In the other patient, no underlying cause could be found. Both patients had an aggressive clinical course and succumbed to this relatively rare syndrome. The PLT refractoriness was evident before the AHPS diagnosis was made.
DISCUSSION: AHPS is not generally a consideration in the evaluation of nonimmune PLT refractoriness. However, these illustrative cases make an argument for its consideration in the differential diagnosis of PLT refractoriness in severely ill patients. Once present, it is unclear if the refractoriness can be reversed by AHPS-targeted therapy
Cybersecurity, Bureaucratic Vitalism and European Emergency
Securing the internet has arguably become paradigmatic for modern security practice, not only because modern life is considered to be impossible or valueless if disconnected, but also because emergent cyber-relations and their complex interconnections are refashioning traditional security logics. This paper analyses European modes of governing geared toward securing vital, emergent cyber-systems in the face of the interconnected emergency. It develops the concept of ‘bureaucratic vitalism’ to get at the tension between the hierarchical organization and reductive knowledge frames of security apparatuses on the one hand, and the increasing desire for building ‘resilient’, dispersed, and flexible security assemblages on the other. The bureaucratic/vital juxtaposition seeks to capture the way in which cybersecurity governance takes emergent, complex systems as object and model without fully replicating this ideal in practice. Thus, we are concerned with the question of what happens when security apparatuses appropriate and translate vitalist concepts into practice. Our case renders visible the banal bureaucratic manoeuvres that seek to operate upon security emergencies by fostering connectivities, producing agencies, and staging exercises
