Modelling of the effect of ELMs on fuel retention at the bulk W divertor of JET
Effect of ELMs on fuel retention at the bulk W target of JET ITER-Like Wall was studied with multi-scale calculations. Plasma input parameters were taken from ELMy H-mode plasma experiment. The energetic intra-ELM fuel particles get implanted and create near-surface defects up to depths of few tens of nm, which act as the main fuel trapping sites during ELMs. Clustering of implantation-induced vacancies were found to take place. The incoming flux of inter-ELM plasma particles increases the different filling levels of trapped fuel in defects. The temperature increase of the W target during the pulse increases the fuel detrapping rate. The inter-ELM fuel particle flux refills the partially emptied trapping sites and fills new sites. This leads to a competing effect on the retention and release rates of the implanted particles. At high temperatures the main retention appeared in larger vacancy clusters due to increased clustering rate
Why Are Outcomes Different for Registry Patients Enrolled Prospectively and Retrospectively? Insights from the Global Anticoagulant Registry in the FIELD-Atrial Fibrillation (GARFIELD-AF).
Background: Retrospective and prospective observational studies are designed to reflect real-world evidence on clinical practice, but can yield conflicting results. The GARFIELD-AF Registry includes both methods of enrolment and allows analysis of differences in patient characteristics and outcomes that may result. Methods and Results: Patients with atrial fibrillation (AF) and ≥1 risk factor for stroke at diagnosis of AF were recruited either retrospectively (n = 5069) or prospectively (n = 5501) from 19 countries and then followed prospectively. The retrospectively enrolled cohort comprised patients with established AF (for a least 6, and up to 24 months before enrolment), who were identified retrospectively (and baseline and partial follow-up data were collected from the emedical records) and then followed prospectively between 0-18 months (such that the total time of follow-up was 24 months; data collection Dec-2009 and Oct-2010). In the prospectively enrolled cohort, patients with newly diagnosed AF (≤6 weeks after diagnosis) were recruited between Mar-2010 and Oct-2011 and were followed for 24 months after enrolment. Differences between the cohorts were observed in clinical characteristics, including type of AF, stroke prevention strategies, and event rates. More patients in the retrospectively identified cohort received vitamin K antagonists (62.1% vs. 53.2%) and fewer received non-vitamin K oral anticoagulants (1.8% vs . 4.2%). All-cause mortality rates per 100 person-years during the prospective follow-up (starting the first study visit up to 1 year) were significantly lower in the retrospective than prospectively identified cohort (3.04 [95% CI 2.51 to 3.67] vs . 4.05 [95% CI 3.53 to 4.63]; p = 0.016). Conclusions: Interpretations of data from registries that aim to evaluate the characteristics and outcomes of patients with AF must take account of differences in registry design and the impact of recall bias and survivorship bias that is incurred with retrospective enrolment. Velocity-space sensitivity of the time-of-flight neutron spectrometer at JET
The velocity-space sensitivities of fast-ion diagnostics are often described by so-called weight functions. Recently, we formulated weight functions showing the velocity-space sensitivity of the often dominant beam-target part of neutron energy spectra. These weight functions for neutron emission spectrometry (NES) are independent of the particular NES diagnostic. Here we apply these NES weight functions to the time-of-flight spectrometer TOFOR at JET. By taking the instrumental response function of TOFOR into account, we calculate time-of-flight NES weight functions that enable us to directly determine the velocity-space sensitivity of a given part of a measured time-of-flight spectrum from TOFOR
On the mechanisms governing gas penetration into a tokamak plasma during a massive gas injection
A new 1D radial fluid code, IMAGINE, is used to simulate the penetration of gas into a tokamak plasma during a massive gas injection (MGI). The main result is that the gas is in general strongly braked as it reaches the plasma, due to mechanisms related to charge exchange and (to a smaller extent) recombination. As a result, only a fraction of the gas penetrates into the plasma. Also, a shock wave is created in the gas which propagates away from the plasma, braking and compressing the incoming gas. Simulation results are quantitatively consistent, at least in terms of orders of magnitude, with experimental data for a D 2 MGI into a JET Ohmic plasma. Simulations of MGI into the background plasma surrounding a runaway electron beam show that if the background electron density is too high, the gas may not penetrate, suggesting a possible explanation for the recent results of Reux et al in JET (2015 Nucl. Cross-cultural adaptation of the City Birth Trauma Scale for the Brazilian context
Background: Posttraumatic stress disorder consists of a set of symptoms that occurs in response to one or more traumatic events and can occur in postpartum, from traumatic situations related to the birth or to the baby’s health in the first days of life. It is important tracking the presence of birth trauma, but there is not available instruments in the Brazilian context for this purpose. Objectives: To present the cross-cultural adaptation of City Birth Trauma Scale (BiTS) into Brazilian portuguese. Methods: Cross-cultural adaptation involved independent translations, synthesis,back-translation, and submission to the original author’s appreciation. After the scale was subjected to face validity, followed by a pilot study with postpartum mothers. Results: All steps were performed for the cross-cultural adaptation. Regarding face validity, items evaluated concerning different types of equivalence, presented satisfactory agreement values (≥4.20). Most of the expert’s suggestions were followed, being the main ones related to adjustments in prepositions, pronouns and verbal subjects. Pilot study showed that the mothers had been able to understand and respond to the instrument without adjustments. Discussion: BiTS’s Brazilian version proved to be cross-culturally adapted, ensuring the possibility of intercultural data comparison from the semantic, idiomatic, cultural, and conceptual perspectives. Caprine lentivirus in sheep milk and semen.
Abstract: With the objective of detecting the presence of caprine lentivirus (CLV) in ewe milk and in ram semen, ten matrixes and four reproducers experimentally infected with CLV were used. Samples of ewe milk were collected during the four months of lactation, five collections per animal, totaling 50 samples. Regarding the rams, eight semen collections were made per animal, during one year of experimentation, totaling 32 samples. The milk and semen samples were submitted to DNA extraction and the nested polymerase chain reaction test (nPCR) to detect CLV proviral DNA. Eight (16%) of the milk samples were positive in nPCR originating from two ewes. Only one (3.12%) semen sample was positive. The amplification products were sequenced, and were confirmed to be a CLV genomic sequence. Thus, the presence of CLV proviral DNA in sheep milk and semen was demonstrated, confirming the feasibility of infection between species, and alerting to the risk of spreading infections. [Lentivírus caprino em leite e sêmen de ovinos]. Resumo: Com o objetivo de detectar a presença do lentivírus caprino (LVC) no leite de ovelhas e no sêmen de carneiros, utilizaram-se 10 matrizes e quatro reprodutores infectados experimentalmente com o LVC. Foram coletadas amostras de leite das ovelhas durante os quatro meses de lactação, ocorrendo cinco coletas por animal, totalizando 50 amostras. Quanto aos carneiros, realizaram-se oito coletas de sêmen por animal, durante um ano de experimentação, totalizando 32 amostras. As amostras de leite e de sêmen foram submetidas à extração de DNA e à prova de reação em cadeia da polimerase do tipo nested (nPCR) visando à detecção de DNA proviral do LVC. Oito (16%) amostras de leite foram positivas na nPCR oriundas de duas ovelhas. Apenas uma (3,12%) amostra de sêmen apresentou positividade. Produtos da amplificação foram sequenciados, confirmando-se tratar de sequência genômica do LVC. Dessa forma, demonstrou-se a presença do DNA proviral do LVC em leite e sêmen de ovinos, confirmando a viabilidade da infecção entre espécies e, assim, alertando sobre o risco de que a infecção seja disseminada.[ALICE ANDRIOLI PINHEIRO]
Avaliação terapêutica do praziquantel (Embay 8440) na infecção humana pelo S. mansoni
Foram tratados com praziquantel, dose oral única de 40 ou 50 mg/kg, 200 indivíduos portadores de esquistossomose mansoni, matriculados na Clínica de Doenças Infecciosas e Parasitárias do Hospital das Clínicas da Faculdade de Medicina da Universidade de São Paulo. As idades dos pacientes variavam de seis a 63 anos, sendo que 33 (16,5%) eram menores de 15 anos. Os principais efeitos colaterais consistiram em tontura 27,5%; sonolência 21,0%; eólica 17,5%; náusea 16,0%; diarréia 9,0%; cefaléia 7,5%; vômito 4,0%; febre 2,0%; disenteria 1,0%; tremor 1,0%; exantema 1,0% e urticaria 0,5%. A toxicidade do medicamento foi investigada mediante a realização, pré e pós-tratamento, de exames hematimétricos, bem como de função renal (uréia e creatinina), hepática (enzimas de liberação hépato-canalicular e bilirrubinas), cardíaca (ECG) e neuropsiquiátrica (EEG). Não foram encontradas nesses controles alterações relevantes que repercutissem clinicamente. O controle de cura verificou-se em 115 indivíduos, através de oito coproscopias, no período de seis meses, subseqüente ao tratamento, utilizando-se duas técnicas (HOFFMAN e KATO/KATZ) para cada amostra de fezes. Dos 82 indivíduos que tiveram as oito coproscopias negativadas, 62 realizaram biópsia retal. Essa mostrou-se positiva em duas oportunidades, indicando um percentual de 3,2% de achados falsos negativos com relação às coproscopias. Os índices de cura variaram de 65,2%, nos pacientes menores de 15 anos, a 75% nos acima dessa idade. Para todas as faixas etárias a eficácia foi de 71,3%. Os resultados obtidos demonstram ter o praziquantel, nas doses empregadas, relativa eficácia no tratamento da esquistossomose mansoni, bem como ser determinante de baixos efeitos tóxico-colaterais
CONTAMINATION POTENTIAL OF SPECIFIC IONS IN SOIL TREATED WITH REJECT BRINE FROM DESALINATION PLANTS
Abyssal DNA: Eukaryotic Diversity in Atlantic Equatorial Deep-Sea Sediments Assessed Through DNA Metabarcoding
Background/Objectives: We evaluated eukaryotic diversity in two cores obtained from abyssal sediments collected at depths of 4280 m and 4444 m in the equatorial Atlantic, between the Fernando de Noronha and São Pedro and São Paulo archipelagos, using a DNA metabarcoding approach applied to environmental DNA (eDNA) samples. Results: In total, we detected 248,905 DNA reads that were assigned to 65 amplicon sequence variants (ASVs) in the two core sediments (176,073 DNA reads and 59 ASVs were detected in sediment obtained at 4280 m depth, and 72,832 DNA reads and 14 ASVs were detected in the core at 4444 m). These represented three Kingdoms and five phyla: Fungi (Ascomycota and Basidiomycota), Viridiplantae (Chlorophyta and Streptophyta) and Chromista (Ciliophora), in rank abundance order. Ascomycota was the dominant phylum, followed by Basidiomycota. Didymella sp., Cladosporium sp., Scopulariopsis sp., Alternaria eichhorniae, Curvularia sp., Hortaea werneckii, Penicillium sp. (Ascomycota) and Malassezia globosa (Basidiomycota) were the most abundant taxa. Pseudochlorella pyrenoidosa (Chlorophyta) was the most abundant representative of Viridiplantae detected, and Spirotrachelostyla tani (Ciliophora) was the only Chromista detected, both present as minor components of the assigned eukaryotic diversity and only in the 4280 m core. The eukaryotic assemblages displayed moderate diversity indices, and those from the deeper core (4444 m depth) displayed the highest diversity values. Few assigned taxa were present in both samples. The two cores differed in their geological characteristics, consistent with their location in different depositional basins. The core obtained at 4280 m depth, located further north and more isolated from the adjacent continent by two fracture zones, appears to receive less terrigenous sediment input. In contrast, the core obtained at 4444 m depth is under greater continental influence and receives more terrigenous input from the continent. These geological and geographic differences may contribute to the varying eukaryotic eDNA diversities found. Results: Our metabarcoding study revealed the presence of a sediment eukaryotic community dominated by fungi. This included assigned ASVs representing groups with different ecological roles, such as cosmopolitan and phytopathogenic members and extremophiles, some of which may be able to survive and function in the polyextreme deep-sea abyssal conditions. Abyssal sediments present a potential habitat for studying organisms at the edge of viable conditions for life on Earth. eDNA metabarcoding provides a promising technique for detecting cryptic and uncultured biodiversity compared to traditional approaches, opening avenues for further ecological, evolutionary and biotechnological studies
