    Immunotherapy: A promising novel endometriosis therapy

    Endometriosis is a common disease of the female reproductive system and has malignant features. Although endometriosis by itself is a benign disease, its erosive growth characteristics lead to severe pelvic pain and female infertility. Unfortunately, several aspects of the pathogenesis of endometriosis are still unclear. Furthermore, the clinical therapeutic methods are unsatisfactory. The recurrence rate of endometriosis is high. Accumulating evidence suggests that the onset and development of endometriosis are closely related to the abnormal function of the female autoimmune system, especially the function of some immune cells such as the aggregation of neutrophils, abnormal differentiation of macrophages, decreased cytotoxicity of NK cells, and abnormal function of T- and B-cell lines. Therefore, immunotherapy is probably a novel therapeutic strategy for endometriosis besides surgery and hormone therapy. However, information regarding the clinical application of immunotherapy in the treatment of endometriosis is very limited. This article aimed to review the effects of existing immunomodulators on the development of endometriosis, including immune cell regulators and immune factor regulators. These immunomodulators clinically or experimentally inhibit the pathogenesis and development of endometriosis lesions by acting on the immune cells, immune factors, or immune-related signaling pathways. Thus, immunotherapy is probably a novel and effective clinical treatment choice for endometriosis. Experimental studies of the detailed mechanism of immunotherapy and large-scale clinical studies about the effectiveness and safety of this promising therapeutic method are required in the future

    The radiosensitizing effect of doranidazole on human colorectal cancer cells exposed to high doses of irradiation

    <p>Abstract</p> <p>Background</p> <p>This paper investigates the effects of a new radiosensitizer, doranidazole, and enhancing irradiation on colorectal cancer cells.</p> <p>Methods</p> <p>The radiosensitizing effect of doranidazole was determined using colony formation and propidium iodide (PI) assays to measure cell growth inhibition and the cell killing effect of human colorectal cancer cell lines exposed to high doses of γ-ray irradiation under hypoxic conditions <it>in vitro</it>. Fluorescence staining and cell migration assays were also used to assess the radiosensitizing effect.</p> <p>Results</p> <p>Cell proliferation evaluated by clonogenic survival curves was significantly inhibited by 5 mmol/L doranidazole, particularly at doses ranging from 10 to 30 Gy of irradiation. The radiosensitizing effect of doranidazole on colorectal cancer cells occurs in a time- and dose-dependent manner. Doranidazole also inhibited the mobility of cell invasion and migration.</p> <p>Conclusion</p> <p>Doranidazole can enhance the killing effect and the cell growth inhibition of colorectal cancer after high-dose irradiation in a time and dose-dependent manner.</p

    A population-based study of interactions between high-risk human papillomavirus infection and vaginal local cytokines CD4 CD8 IL-10 with cervical intraepithelial neoplasia

    BackgroundCluster of Differentiation-4(CD4),Cluster of Differentiation-8(CD8), and interleukin-10 (IL-10) have long been considered to be related to cervical cancer, but the exact relationship remains unclear. Few studies investigated the relationship between CD4,CD8,IL-10, and high-risk human papillomavirus (HPV) with risk of cervical intraepithelial neoplasia (CIN).ObjectiveOur aim is to evaluate the relationship between CD4, CD8, IL-10, and high-risk HPV infection with the risk of CIN, as well as their interactions on CIN.DesignIn 2014-2015, a cross-sectional study of screening data was conducted among 2285 women aged 19–65 years who participated in an ongoing community-based cohort of 40,000 women in Shanxi, China. Using categorical and spline analyses to evaluate the relationship between local vaginal fluids of CD4,CD8,CD4/CD8,IL-10, and CIN risk. A total of 1,503 controls were followed up until January 31, 2019. A nested case-control study was used to assess the relationship between vaginal lavage CD4, CD8, CD4/CD8, and IL-10 levels and the risk of CIN progression.ResultsAfter adjusting for possible confounding factors,CD4 and CD8 levels were positively related to CIN risk (the 1st versus 4th quartile CD4,CD8 OR = 0.45[0.34, 0.60] and 0.34[0.26, 0.45] for CIN1, 0.32 [0.21, 0.48] and 0.24 [0.16, 0.38] for CIN2/3). Increased CD4 and CD8 levels were positively related to the occurrence of CIN(P-overall&lt;0.01).CD4/CD8 levels and the risk of CIN1 followed a nonlinear “U-shape” (P-nonlinear &lt;0.01). IL-10 levels and the risk of CIN1 followed a nonlinear “n-shape”(P-nonlinear &lt;0.01).IL-10 levels were inversely related to the occurrence of CIN2/3(OR = 3.87, [2.49, 6.00],P-overall&lt;0.01). The highest risk of CIN was observed in women with high-risk HPV, whose CD4 and CD8 levels were the highest(P-interaction &lt; 0.01).Patients with the lowest IL-10 levels(IL-10 ≤ 53.17pg/ml) who are positive for high-risk HPV infection have the highest risk of CIN2/3(OR = 18.46,[9.33-36.51]). Nested case-control analysis observed a positive relationship between CD4,CD8 levels, and risk of CIN progression (CD4 OR = 0.34,[0.13, 0.94];CD8 OR = 0.27, [0.09, 0.79]),and an opposite relationship between IL-10 levels and risk of CIN progression (OR = 2.92, [1.09, 7.84]).ConclusionsLocal vaginal CD4 and CD8 levels were positively correlated with CIN risk, and IL-10 levels were inversely correlated with CIN2/3, whether or not with high-risk HPV infection in Chinese women

    Characteristics of Oil-Water Relative Permeability and Influence Mechanism in Fractured-Vuggy Medium

    AbstractThe fracture system of fractured-vuggy reservoir is dominated by orthogonal fracture, and the vug system develops along the fracture. In view of this characteristic, two different kinds of models were made in this paper: the fracture medium model and the fractured-vuggy medium model. A series of two-phase fluid flow experiments were conducted to obtain the relative permeability curves. The results show that the oil and water relative permeability curves have a regular alternative relation of growing and declining, while the percolation resistance changes little, which indicates that there is no Jamin action that caused by the complexity of pore structure. Fracture aperture, fracture network structure, vug density and vug porosity have different influence on the relative permeability characteristic of fractured-vuggy medium, and the influence shows a obvious regularity

To solve these problems, a more efficient variable stiffness analysis model has been propose, which can obtain quantitative stiffness value of the machining surface. Applying simulate cutting force in sampling points using finite element analysis software ABAQUS, the single direction variable stiffness rule can be obtained. The variable stiffness matrix has been propose by analyzing multi-directions coupling variable stiffness rule. Combining with the three direction cutting force value, the reasonability of existing processing parameters can be verified and the optimized cutting parameters can be designed

The framework of intelligent manufacturing execution process control technology proposed an effective way to realize the manufacturing process intelligent control based on existing MES; the technology of logistics identification and control based on RFID realized the effectiveness control of logistics; the intelligent monitoring technology of mobile terminal solve the mobile terminal positioning problem, MES pushing information to mobile terminal problem and other problems. Finally, the author developed a set of intelligent manufacturing execution process control system which includes RFID reader, MES server, mobile terminal, and verified the effectiveness of the above mentioned technologies
