Deep carbon storage potential of buried floodplain soils.
Soils account for the largest terrestrial pool of carbon and have the potential for even greater quantities of carbon sequestration. Typical soil carbon (C) stocks used in global carbon models only account for the upper 1 meter of soil. Previously unaccounted for deep carbon pools (>1 m) were generally considered to provide a negligible input to total C contents and represent less dynamic C pools. Here we assess deep soil C pools associated with an alluvial floodplain ecosystem transitioning from agricultural production to restoration of native vegetation. We analyzed the soil organic carbon (SOC) concentrations of 87 surface soil samples (0-15 cm) and 23 subsurface boreholes (0-3 m). We evaluated the quantitative importance of the burial process in the sequestration of subsurface C and found our subsurface soils (0-3 m) contained considerably more C than typical C stocks of 0-1 m. This deep unaccounted soil C could have considerable implications for global C accounting. We compared differences in surface soil C related to vegetation and land use history and determined that flooding restoration could promote greater C accumulation in surface soils. We conclude deep floodplain soils may store substantial quantities of C and floodplain restoration should promote active C sequestration

Implications of the Internet of Things for the Small and Medium Sized Business
The Internet of Things (IoT) is the network of physical objects containing the embedded capability to interact, communicate, and otherwise exchange data with one another and the external environment over a network without human intervention. As the Internet of Things begins to grow into almost all aspects of business, the small to midsize business (SMB) must not get behind. Some unanswered questions include: How will small and medium-sized businesses (SMB) participate in IoT? Have they already begun participating and what challenges have they encountered along the way? Do smaller organizations have a disadvantage compared to larger organizations in the IoT landscape? A literature review and a survey of owners, executives and employees of SMBs were performed in order to gain a better understanding of the current state of awareness and use of IoT technologies. The thesis concludes with a theoretical recommended implementation plan that combines the literature on the strengths and weaknesses of the SMB with the results of the survey as well as the literature examining the challenges and opportunities of IoT into a series of recommendations for implementation. These recommendations are given in the form of a “Maturity Model” that will cover the steps from infancy to a fully mature implementation of IoT solutions

Rescue of the MERTK phagocytic defect in a human iPSC disease model using translational read-through inducing drugs.
Inherited retinal dystrophies are an important cause of blindness, for which currently there are no effective treatments. In order to study this heterogeneous group of diseases, adequate disease models are required in order to better understand pathology and to test potential therapies. Induced pluripotent stem cells offer a new way to recapitulate patient specific diseases in vitro, providing an almost limitless amount of material to study. We used fibroblast-derived induced pluripotent stem cells to generate retinal pigment epithelium (RPE) from an individual suffering from retinitis pigmentosa associated with biallelic variants in MERTK. MERTK has an essential role in phagocytosis, one of the major functions of the RPE. The MERTK deficiency in this individual results from a nonsense variant and so the MERTK-RPE cells were subsequently treated with two translational readthrough inducing drugs (G418 & PTC124) to investigate potential restoration of expression of the affected gene and production of a full-length protein. The data show that PTC124 was able to reinstate phagocytosis of labeled photoreceptor outer segments at a reduced, but significant level. These findings represent a confirmation of the usefulness of iPSC derived disease specific models in investigating the pathogenesis and screening potential treatments for these rare blinding disorders
Leveraging DNA-Methylation Quantitative-Trait Loci to Characterize the Relationship between Methylomic Variation, Gene Expression, and Complex Traits
Characterizing the complex relationship between genetic, epigenetic, and transcriptomic variation has the potential to increase understanding about the mechanisms underpinning health and disease phenotypes. We undertook a comprehensive analysis of common genetic variation on DNA methylation (DNAm) by using the Illumina EPIC array to profile samples from the UK Household Longitudinal study. We identified 12,689,548 significant DNA methylation quantitative trait loci (mQTL) associations (p 60 human traits by using summary-data-based Mendelian randomization (SMR) to identify 1,662 pleiotropic associations between 36 complex traits and 1,246 DNAm sites. We also use SMR to characterize the relationship between DNAm and gene expression and thereby identify 6,798 pleiotropic associations between 5,420 DNAm sites and the transcription of 1,702 genes. Our mQTL database and SMR results are available via a searchable online database as a resource to the research community
Post traumatic internal iliac arteriovenous fistula presenting with persistent gross hematuria.
Traumatic pelvic arteriovenous fistulas (AVFs) are exceedingly rare and can lead to significant morbidity. Herein, we describe the case of a 31 year-old male who presented with two months of recurrent gross hematuria following a gluteal gun-shot wound and was found to have a right internal iliac AVF. To our knowledge, this is the first reported case of a post traumatic pelvic AVF presenting with gross hematuria
Near-UV and optical observations of the transiting exoplanet TrES-3b
We observed nine primary transits of the hot Jupiter TrES-3b in several
optical and near-UV photometric bands from 2009 June to 2012 April in an
attempt to detect its magnetic field. Vidotto, Jardine and Helling suggest that
the magnetic field of TrES-3b can be constrained if its near-UV light curve
shows an early ingress compared to its optical light curve, while its egress
remains unaffected. Predicted magnetic field strengths of Jupiter-like planets
should range between 8 G and 30 G. Using these magnetic field values and an
assumed B_star of 100 G, the Vidotto et al. method predicts a timing difference
of 5-11 min. We did not detect an early ingress in our three nights of near-UV
observations, despite an average cadence of 68 s and an average photometric
precision of 3.7 mmag. However, we determined an upper limit of TrES-3b's
magnetic field strength to range between 0.013 and 1.3 G (for a 1-100 G
magnetic field strength range for the host star, TrES-3) using a timing
difference of 138 s derived from the Nyquist-Shannon sampling theorem. To
verify our results of an abnormally small magnetic field strength for TrES-3b
and to further constrain the techniques of Vidotto et al., we propose future
observations of TrES-3b with other platforms capable of achieving a shorter
near-UV cadence. We also present a refinement of the physical parameters of
TrES-3b, an updated ephemeris and its first published near-UV light curve. We
find that the near-UV planetary radius of Rp = 1.386+0.248-0.144 RJup is

Socioeconomic Position and DNA Methylation Age Acceleration across the Lifecourse.
Accelerated DNA methylation age is linked to all-cause mortality and environmental factors, but studies of associations with socioeconomic position are limited. Studies generally use small selected samples, and it is unclear how findings with two commonly used methylation age calculations (Horvath and Hannum) translate to general population samples including younger and older adults. In 1099 UK adults aged 28-98 y in 2011-12, we assessed the relationship of Horvath and Hannum DNA methylation age acceleration with a range of social position measures: current income and employment, education, income and unemployment across a 12-year period, and childhood social class. Accounting for confounders, participants less advantaged in childhood were epigenetically 'older' as adults: compared to participants with professional/managerial parents, Hannum age was 1.07 years higher (95% confidence interval (CI):0.20-1.94) for those with parents in semi-skilled/unskilled occupations, and 1.85 years higher (95%CI:0.67-3.02) for participants without a working parent at age 14. No other robust associations were seen. Results accord with research implicating early life circumstances as critical for DNA methylation age in adulthood. Since methylation age acceleration as measured by the Horvath and Hannum estimators appears strongly linked to chronological age, research examining associations with the social environment must take steps to avoid age-related confounding
Effects of antiplatelet therapy on stroke risk by brain imaging features of intracerebral haemorrhage and cerebral small vessel diseases: subgroup analyses of the RESTART randomised, open-label trial
Background
Findings from the RESTART trial suggest that starting antiplatelet therapy might reduce the risk of recurrent symptomatic intracerebral haemorrhage compared with avoiding antiplatelet therapy. Brain imaging features of intracerebral haemorrhage and cerebral small vessel diseases (such as cerebral microbleeds) are associated with greater risks of recurrent intracerebral haemorrhage. We did subgroup analyses of the RESTART trial to explore whether these brain imaging features modify the effects of antiplatelet therapy
