Downregulation of Augmenter of Liver Regeneration Impairs the Therapeutic Efficacy of Liver Epithelial Progenitor Cells Against Acute Liver Injury by Enhancing Mitochondrial Fission
Abstract
Cell-based therapeutic approaches have been proven to be effective strategies for the treatment of acute liver injury (ALI). However, widespread application of these procedures is limited by several key issues, including rapid loss of stemness in vitro, aberrant differentiation into undesirable cell types, and low engraftment in vivo. In this study, liver epithelial progenitor cells (LEPCs) were characterized and transfected with augmenter of liver regeneration (ALR). The results revealed that in ALI mice with CCl4, the transplantation of ALR-bearing LEPCs into the liver markedly protected mice against ALI by decreasing the levels of alanine aminotransferase (ALT) and aspartate aminotransferase (AST), thus relieving hepatic tissue injury and attenuating inflammatory infiltration. Mechanistically, the knockdown of ALR in LEPCs activated the phosphorylation of dynamin-related protein 1 (Drp1) at the S616 site and thereby enhanced mitochondrial fission. In contrast, the transfection of ALR into LEPCs significantly inhibited Drp1 phosphorylation, thereby favoring the maintenance of mitochondrial integrity and the preservation of adenosine triphosphate contents in LEPCs. Consequently, the ALR-bearing LEPCs transplanted into ALI mice exhibited substantially greater homing ability to the injured liver via the SDF-1/CXCR4 axis than that of LEPCs-lacking ALR. In conclusion, we demonstrated that the transplantation of ALR-transfected LEPCs protected mice against CCl4-induced ALI, thus offering immense curative potential in the clinic.</jats:p

LPIN: A Lightweight Progressive Inpainting Network for Improving the Robustness of Remote Sensing Images Scene Classification
At present, the classification accuracy of high-resolution Remote Sensing Image Scene Classification (RSISC) has reached a quite high level on standard datasets. However, when coming to practical application, the intrinsic noise of satellite sensors and the disturbance of atmospheric environment often degrade real Remote Sensing (RS) images. It introduces defects to them, which affects the performance and reduces the robustness of RSISC methods. Moreover, due to the restriction of memory and power consumption, the methods also need a small number of parameters and fast computing speed to be implemented on small portable systems such as unmanned aerial vehicles. In this paper, a Lightweight Progressive Inpainting Network (LPIN) and a novel combined approach of LPIN and the existing RSISC methods are proposed to improve the robustness of RSISC tasks and satisfy the requirement of methods on portable systems. The defects in real RS images are inpainted by LPIN to provide a purified input for classification. With the combined approach, the classification accuracy on RS images with defects can be improved to the original level of those without defects. The LPIN is designed on the consideration of lightweight model. Measures are adopted to ensure a high gradient transmission efficiency while reducing the number of network parameters. Multiple loss functions are used to get reasonable and realistic inpainting results. Extensive tests of image inpainting of LPIN and classification tests with the combined approach on NWPU-RESISC45, UC Merced Land-Use and AID datasets are carried out which indicate that the LPIN achieves a state-of-the-art inpainting quality with less parameters and a faster inpainting speed. Furthermore, the combined approach keeps the comparable classification accuracy level on RS images with defects as that without defects, which will improve the robustness of high-resolution RSISC tasks

Axial compressive behaviour of square through-beam joints between CFST columns and RC beams with multi-layers of steel meshes
The axial compressive behaviour of an innovative type of square concrete filled steel tube (CFST) column to reinforced concrete (RC) beam joint was experimentally investigated in this paper. The innovative joint was designed such that (i) the steel tubes of the CFST columns were completely interrupted in the joint region, (ii) the longitudinal reinforcements from the RC beams could easily pass through the joint area and (iii) a reinforcement cage, including a series of reinforcement meshes and radial stirrups, was arranged in the joint area to strengthen the mechanical performance of the joint. A twostage experimental study was conducted to investigate the behaviour of the innovative joint under axial compression loads, where the first stage of the tests included three fullscale innovative joint specimens subjected to axial compression to assess the feasibility of the joint detailing and propose measures to further improve its axial compressive behaviour, and the second stage of the tests involved 14 innovative joint specimens with the improved detailing to study the effect of the geometric size of the joint, concrete strength and volume ratio of the steel meshes on the bearing strengths of the joints. It was generally found from the experiments that (i) the innovative joint is capable of achieving the design criterion of the 'strong jointweak member' with appropriate designs, and (ii) by decreasing the height factor and increasing the volume ratio of the steel meshes, the axial compressive strengths of the joints significantly increased, while the increase of the length factor is advantageous but limited to the resistances of the joint specimens. Because of the lack of existing design methods for the innovative joints, new design expressions were proposed to calculate the axial compression resistances of the innovative joints subjected to bearing loads, with the local compression effect, the confinement effect provided by the multilayers of steel meshes and the height effect of concrete considered. It was found that the proposed design methods were capable of providing accurate and safe resistance predictions for the innovative joints.Published versio
