    Anthropogenic modification of the natural fire landscape and its consequences for vegetation patterns on the Cape Peninsula

    Understanding the spatial probability of fire and how urban development may alter natural patterns is particularly important in areas where alternate ecosystem states occur at fine spatial scales. The Cape Peninsula, South Africa, is a one such region where fire-sensitive forest patches occur interspersed in a sea of fire-dependent fynbos. Fire is believed to be an important determinate of forest distribution, with absence or occurrence of fires potentially allowing patch contraction and expansion. In this thesis I use a series of computer models to determine the extent to which anthropogenic development and land transformation have altered the spatial variation in fire likelihood, or the 'burn probability', and its consequence for the distribution of forest on the Cape Peninsula. The two multi-model, fire behaviour simulation systems I use are FlamMap and FARSITE. FARSITE is a deterministic simulation package used globally for discrete event simulation. In an effort to assess the viability of using the FARSITE model for fire prediction in fynbos and the determinants of model accuracy, I predicted fire area for a historical fire on the Cape Peninsula using a variety of fuel models and wind conditions. Following this validation, FlamMap was used to simulate the burn probability of the Cape Peninsula under natural conditions – no urban development present – and transformed conditions – where urban areas mapped as non-burnable fuel models. I then determined changes in forest distribution documented over the last 50 years relative to changes in burn probability as a result of urbanisation. My results show that an increase in urbanisation on the Cape Flats has produced a significant urban shadow effect due to the interruption of natural fire catchments. This urban shadow effect has resulted in an overall increase in area of fire refuges on the Peninsula and expansion of forest, particularly on the more mesic eastern slopes at Kirstenbosch and Newlands. The results strongly support that urban-mediated changes to fire patterns are drivers of forest expansion in this region, and adds further evidence to support the significance of fire in determining biome boundaries in the fynbos

Physical activity intervention studies with older people often have low recruitment, yet little is known about non-participants. Methods: Patients aged 60–74 years from three UK general practices were invited to participate in a nurse-supported pedometer-based walking intervention. Demographic characteristics of 298 participants and 690 non-participants were compared. Health status and physical activity of 298 participants and 183 non-participants who completed a survey were compared using age, sex adjusted odds ratios (OR) (95% confidence intervals). 15 non-participants were interviewed to explore perceived barriers to participation. Results: Recruitment was 30% (298/988). Participants were more likely than non-participants to be female (54% v 47%; p = 0.04) and to live in affluent postcodes (73% v 62% in top quintile; p < 0.001). Participants were more likely than non-participants who completed the survey to have an occupational pension OR 2.06 (1.35-3.13), a limiting longstanding illness OR 1.72 (1.05-2.79) and less likely to report being active OR 0.55 (0.33-0.93) or walking fast OR 0.56 (0.37-0.84). Interviewees supported general practice-based physical activity studies, particularly walking, but barriers to participation included: already sufficiently active, reluctance to walk alone or at night, physical symptoms, depression, time constraints, trial equipment and duration. Conclusion: Gender and deprivation differences suggest some selection bias. However, trial participants reported more health problems and lower activity than non-participants who completed the survey, suggesting appropriate trial selection in a general practice population.     BACKGROUND: Physical activity is essential for older peoples' physical and mental health and for maintaining independence. Guidelines recommend at least 150 minutes weekly, of at least moderate intensity physical activity, with activity on most days. Older people's most common physical activity is walking, light intensity if strolling, moderate if brisker. Less than 20% of United Kingdom 65-74 year olds report achieving the guidelines, despite most being able to. Effective behaviour change techniques include strategies such as goal setting, self-monitoring, building self-efficacy and relapse prevention. Primary care physical activity consultations allow individual tailoring of advice. Pedometers measure step-counts and accelerometers measure physical activity intensity. This protocol describes an innovative intervention to increase walking in older people, incorporating pedometer and accelerometer feedback within a primary care nurse physical activity consultation, using behaviour change techniques. DESIGN: Randomised controlled trial with intervention and control (usual care) arms plus process and qualitative evaluations. PARTICIPANTS: 300 people aged 60-74 years registered with 3 general practices within Oxfordshire and Berkshire West primary care trusts, able to walk outside and with no restrictions to increasing their physical activity. INTERVENTION: 3 month pedometer and accelerometer based intervention supported by practice nurse physical activity consultations. Four consultations based on behaviour change techniques, physical activity diary, pedometer average daily steps and accelerometer feedback on physical activity intensity. Individual physical activity plans based on increasing walking and other existing physical activity will be produced. OUTCOMES: Change in average daily steps (primary outcome) and average time spent in at least moderate intensity physical activity weekly (secondary outcome) at 3 months and 12 months, assessed by accelerometry. Other outcomes include quality of life, mood, exercise self-efficacy, injuries. Qualitative evaluations will explore reasons for trial non-participation, the intervention's acceptability to patients and nurses and factors enhancing or acting as barriers for older people in increasing their physical activity levels. DISCUSSION: The PACE-Lift trial will determine the feasibility and efficacy of an intervention for increasing physical activity among older primary care patients. Steps taken to minimise bias and the challenges anticipated will be discussed. Word count 341.     Parkinson Phenotype in Aged PINK1-Deficient Mice Is Accompanied by Progressive Mitochondrial Dysfunction in Absence of Neurodegeneration

    BackgroundParkinson's disease (PD) is an adult-onset movement disorder of largely unknown etiology. We have previously shown that loss-of-function mutations of the mitochondrial protein kinase PINK1 (PTEN induced putative kinase 1) cause the recessive PARK6 variant of PD.Methodology/principal findingsNow we generated a PINK1 deficient mouse and observed several novel phenotypes: A progressive reduction of weight and of locomotor activity selectively for spontaneous movements occurred at old age. As in PD, abnormal dopamine levels in the aged nigrostriatal projection accompanied the reduced movements. Possibly in line with the PARK6 syndrome but in contrast to sporadic PD, a reduced lifespan, dysfunction of brainstem and sympathetic nerves, visible aggregates of alpha-synuclein within Lewy bodies or nigrostriatal neurodegeneration were not present in aged PINK1-deficient mice. However, we demonstrate PINK1 mutant mice to exhibit a progressive reduction in mitochondrial preprotein import correlating with defects of core mitochondrial functions like ATP-generation and respiration. In contrast to the strong effect of PINK1 on mitochondrial dynamics in Drosophila melanogaster and in spite of reduced expression of fission factor Mtp18, we show reduced fission and increased aggregation of mitochondria only under stress in PINK1-deficient mouse neurons.ConclusionThus, aging Pink1(-/-) mice show increasing mitochondrial dysfunction resulting in impaired neural activity similar to PD, in absence of overt neuronal death

    Global Spatial Risk Assessment of Sharks Under the Footprint of Fisheries

    Effective ocean management and conservation of highly migratory species depends on resolving overlap between animal movements and distributions and fishing effort. Yet, this information is lacking at a global scale. Here we show, using a big-data approach combining satellite-tracked movements of pelagic sharks and global fishing fleets, that 24% of the mean monthly space used by sharks falls under the footprint of pelagic longline fisheries. Space use hotspots of commercially valuable sharks and of internationally protected species had the highest overlap with longlines (up to 76% and 64%, respectively) and were also associated with significant increases in fishing effort. We conclude that pelagic sharks have limited spatial refuge from current levels of high-seas fishing effort. Results demonstrate an urgent need for conservation and management measures at high-seas shark hotspots and highlight the potential of simultaneous satellite surveillance of megafauna and fishers as a tool for near-real time, dynamic management

    Global tracking of marine megafauna space use reveals how to achieve conservation targets

    The recent Kunming-Montreal Global Biodiversity Framework (GBF) sets ambitious goals but no clear pathway for how zero loss of important biodiversity areas and halting human-induced extinction of threatened species will be achieved. We assembled a multi-taxa tracking dataset (11 million geopositions from 15,845 tracked individuals across 121 species) to provide a global assessment of space use of highly mobile marine megafauna, showing that 63% of the area that they cover is used 80% of the time as important migratory corridors or residence areas. The GBF 30% threshold (Target 3) will be insufficient for marine megafauna’s effective conservation, leaving important areas exposed to major anthropogenic threats. Coupling area protection with mitigation strategies (e.g., fishing regulation, wildlife-traffic separation) will be essential to reach international goals and conserve biodiversity

    Occult moisture inputs in the fynbos : Foliar moisure uptake in Ericaceae, Restionaceae and Proteaceae / Annabelle J. Rogers

    'Occult' precipitation may be an important moisture source that can impact specific plant responses to water stress in certain ecosystems. Species in many ecosystems have been shown to use this moisture source via foliar moisture uptake. The Cape Peninsula and the surrounding southern coast experience high altitude mountain cloud cover in the summer dry period. Previous work on species that exist exclusively in the cloud belt found that species from the Erica and Restionaceae family were able to absorb water through their leaf surfaces, while members of the Proteaceae were unable to do so (Gibson, 2012). Adding to the body of research, this study explores whether foliar uptake occurs in species that do not exist in the cloud belt of the Cape Peninsula and the relative importance of this method of uptake in positively impacting a plants water balance during a cloud event. Direct foliar uptake was assessed at the leaf level using two methods; submersion and mist exposure. The importance of foliar uptake was measured at the whole plant level by exposing whole plants to cloud in a mist chamber. By covering the soil in a subset of plants, we were able to isolate the importance of foliar uptake relative to uptake via drip. Results found that the restios and ericas showed a stronger ability to take up moisture via their leaves than protea species, which showed little ability to do so. In contrast to this, the mist exposure method showed no significant uptake in any species except E. quadrangularis. This disparity is possibly due to specific leaf morphology. All species showed significant hydration in response to a whole plant cloud event in both the covered and uncovered treatments. The significant hydration in response to an occult event suggests that certain low land species are capable of utilizing small occult inputs despite the lack of regular summer cloud events as seen in the cloud belt species. As family patterns of foliar moisture uptake seen in cloud belt species (Gibson 2012) are conserved in the surveyed low land species, it can be concluded that moisture uptake is not a trait specific to cloud belt species, and represents a family level pattern of ability
