    A decision-making approach for investigating the potential effects of near sourcing on supply chain

    Purpose - Near sourcing is starting to be regarded as a valid alternative to global sourcing in order to leverage supply chain (SC) responsiveness and economic efficiency. The present work proposes a decision-making approach developed in collaboration with a leading Italian retailer that was willing to turn the global store furniture procurement process into near sourcing. Design/methodology/approach - Action research is employed. The limitations of the traditional SC organisation and purchasing process of the company are first identified. On such basis, an inventory management model is applied to run spreadsheet estimates where different purchasing and SC management strategies are adopted to determine the solution providing the lowest cost performance. Finally, a risk analysis of the selected best SC arrangement is conducted and results are discussed. Findings - Switching from East Asian suppliers to continental vendors enables a SC reengineering that increases flexibility and responsiveness to demand uncertainty which, together with decreased transportation costs, assures economic viability, thus proving the benefits of near sourcing. Research limitations/implications - The decision-making framework provides a methodological roadmap to address the comparison between near and global sourcing policies and to calculate the savings of the former against the latter. The approach could include additional organisational aspects and cost categories impacting on near sourcing and could be adapted to investigate different products, services, and business sectors. Originality/value - The work provides SC researchers and practitioners with a structured approach for understanding what drives companies to adopt near sourcing and for quantitatively assessing its advantage

    Administration of the antioxidant n-acetyl-cysteine in pregnant mice has long-term positive effects on metabolic and behavioral endpoints of male and female offspring prenatally exposed to a high-fat diet

    A growing body of evidence suggests the consumption of high-fat diet (HFD) during pregnancy to model maternal obesity and the associated increase in oxidative stress (OS), might act as powerful prenatal stressors, leading to adult stress-related metabolic or behavioral disorders. We hypothesized that administration of antioxidants throughout gestation might counteract the negative effects of prenatal exposure to metabolic challenges (maternal HFD feeding during pregnancy) on the developing fetus. In this study, female C57BL/6J mice were fed HFD for 13 weeks (from 5-weeks of age until delivery) and were exposed to the N-acetyl-cysteine (NAC) antioxidant from 10-weeks of age until right before delivery. Body weight of the offspring was assessed following birth, up to weaning and at adulthood. The metabolic, neuroendocrine and emotional profile of the adult offspring was tested at 3-months of age. Prenatal HFD increased mother’s body weight and offspring’s weight at the time of weaning, when administered in conjunction with NAC. In females, NAC administration reduced high levels of leptin resulting from prenatal HFD. Prenatal NAC administration also resulted in greater glucose tolerance and insulin sensitivity while increasing adiponectin levels, as well as increasing exploratory behavior, an effect accompanied by reduced plasma corticosterone levels in response to restraint stress. Analysis of glutathione levels in the hypothalamus and in brown adipose tissue indicates that, while HFD administration to pregnant dams led to reduced levels of glutathione in the offspring, as in the male hypothalamus, NAC was able to revert this effect and to increase glutathione levels both in the periphery (Brown Adipose Tissue, both males and females) and in the central nervous system (males). Overall, results from this study indicate that the body redox milieu should be tightly regulated during fetal life and that buffering OS during pregnancy can have important long-term consequences on metabolic and behavioral endpoints

    Climate control on sulphate and nitrate concentrations in alpine streams of Northern Italy along a nitrogen saturation gradient

    International audienceThe role of meteorology, hydrology and atmospheric deposition on the temporal pattern of SO4 and NO3 concentrations was investigated for three streams draining alpine catchments in Northern Italy. The study sites lie on a gradient of atmospheric fluxes of SO4 and NO3 (from about 50 to 80 meq m?2 y?1, and from 40 to 90 meq m?2 y?1, respectively). As a consequence of the increasing N input, the three catchments are also representative of aggrading levels of N saturation. Different methods of statistical analysis were applied to monthly data for the period 1997?2005 to identify which variables (temperature, precipitation, hydrology, SO4 and NO3 deposition) were the main predictors of water chemistry and its change in time. Hydrological changes and snow cover proved to be the main confounding factors in the response to atmospheric deposition in the River Masino catchment. Its particular characteristics (small catchment area, rapid flushing during runoff and thin soil cover) meant that this site responded without a significant delay to SO4 deposition decrease. It also showed a clear seasonal pattern of NO3 concentration, in response to hydrology and biological uptake in the growing season. The selected driving variables failed to model the water chemistry at the other study sites. Nevertheless, temperature, especially extreme values, turned out to be important in both SO4 and NO3 export from the catchments. This result might be largely explained by the effect of warm periods on temperature-dependent processes such as mineralization, nitrification and S desorption. Our findings suggest that surface waters in the alpine area will be extremely sensitive to a climate warming scenario: higher temperatures and increasing frequency of drought could exacerbate the effects of high chronic N deposition

    Nonylphenol and octylphenol differently affect cell redox balance by modulating the nitric oxide signaling

    Nonylphenol (NP) and octylphenol (OP) are pervasive environmental contaminants belonging to the broader class of compounds known as alkylphenols, with potential human toxic effects. Classified as "xenoestrogens," NP and OP are able to interfere with the cell endocrine physiology via a direct interaction with the estrogen receptors. Here, using HepG2 cells in culture, the changes of the cell redox balance and mitochondrial activity induced by OP and NP have been investigated at mu M concentrations, largely below those provoking acute toxicity, as those typical of environmental contaminants. Following 24 h cell exposure to both OP and NP, ROS production appeared significantly increased (p <= 0.01), together with the production of higher NO oxides (p = 0.003) and peroxynitrated protein-derivatives (NP versus CTR, p = 0.003). The mitochondrial proton electrochemical potential gradient instead was decreased (p <= 0.05), as the oxygen consumption by complex IV, particularly following incubation with NP (NP versus CTR, p = 0.017). Consistently, the RT-PCR and Western blot analyses proved that the OP and NP can modulate to a different extent the expression of the inducible NOS (NP versus CTR, p <= 0.01) and the endothelial NOS (OP versus CTR, p <= 0.05), with a significant variation of the coupling efficiency of the latter (NP versus CTR, p <= 0.05), a finding that may provide a novel clue to understand the specific xenoestrogenic properties of OP and NP

    Evidence for detrimental cross interactions between reactive oxygen and nitrogen species in Leber's hereditary optic neuropathy cells

    Here we have collected evidence suggesting that chronic changes in the NO homeostasis and the rise of reactive oxygen species bioavailability can contribute to cell dysfunction in Leber’s hereditary optic neuropathy (LHON) patients.We report that peripheral blood mononuclear cells (PBMCs), derived froma female LHON patient with bilateral reduced vision and carrying the pathogenic mutation 11778/ND4, display increased levels of reactive oxygen species (ROS) and reactive nitrogen species (RNS), as revealed by flow cytometry, fluorometric measurements of nitrite/nitrate, and 3-nitrotyrosine immunodetection. Moreover, viability assays with the tetrazolium dye MTT showed that lymphoblasts from the same patient are more sensitive to prolonged NO exposure, leading to cell death. Taken together these findings suggest that oxidative and nitrosative stress cooperatively play an important role in driving LHON pathology when excess NO remains available over time in the cell environment

    Two-fluid and magnetohydrodynamic modelling of magnetic reconnection in the MAST spherical tokamak and the solar corona

    Twisted magnetic flux ropes are ubiquitous in space and laboratory plasmas, and the merging of such flux ropes through magnetic reconnection is an important mechanism for restructuring magnetic fields and releasing free magnetic energy. The merging-compression scenario is one possible start up scheme for spherical tokamaks, which has been used on the Mega Amp Spherical Tokamak MAST. Two current-carrying plasma rings, or flux ropes, approach each other through the mutual attraction of their like currents, and merge, through magnetic reconnection, into a single plasma torus, with substantial plasma heating. 2D resistive MHD and Hall MHD simulations of this process are reported, and new results for the temperature distribution of ions and electrons are presented. A model of the based on relaxation theory is also described, which is now extended to tight aspect ratio geometry. This model allows prediction of the final merged state and the heating. The implications of the relaxation model for heating of the solar corona are also discussed, and a model of the merger of two or more twisted coronal flux ropes is presented, allowing for different senses of twist

    Cardiovascular mitochondrial dysfunction induced by cocaine: biomarkers and possible beneficial effects of modulators of oxidative stress

    Cocaine abuse has long been known to cause morbidity and mortality due to its cardiovascular toxic effects. The pathogenesis of the cardiovascular toxicity of cocaine use has been largely reviewed, and the most recent data indicate a fundamental role of oxidative stress in cocaine-induced cardiovascular toxicity, indicating that mitochondrial dysfunction is involved in the mechanisms of oxidative stress. The comprehension of the mechanisms involving mitochondrial dysfunction could help in selecting the most appropriate mitochondria injury biological marker, such as superoxide dismutase-2 activity and glutathionylated hemoglobin. The potential use of modulators of oxidative stress (mitoubiquinone, the short-chain quinone idebenone, and allopurinol) in the treatment of cocaine cardiotoxic effects is also suggested to promote further investigations on these potential mitochondria-targeted antioxidant strategies

    Acute haemolysis in childhood falciparum malaria

    Acute haemolysis associated with clinical episodes of high-level Plasmodium falciparum parasitaemia was studied in 20 children from an holoendemic area (coastal Tanzania). The change in blood haemoglobin (Hb) concentration ranged from -46 to +5 g/L during the 72-h observation period and was linearly related to maximum parasitaemia. Balance studies between loss of blood Hb, increase in plasma Hb and appearance of Hb in the urine indicated that extravascular clearance of red cells was the predominant mode of erythrocyte clearance. Most subjects, however, showed minor signs of intravascular haemolysis. The plasma Hb was ≪1% of blood Hb and haemoglobinuria was detected in 14/20 children but the excretion of Hb in urine was \u3c0.5% of total Hb loss. Haemoglobinuria was, however, a marker of severe haemolysis, since the maximum blood Hb loss in children without haemoglobinuria was 10 g/L. Erythrocyte-bound opsonins known to induce erythrophagocytosis, i.e., complement C3c fragments and autologous IgG, were increased in all patients. In the patients with major haemolysis, the changes correlated to the haemolysis over time. Hence, a similar mechanism for predominantly extravascular erythrocyte clearance may be operative in acute malarial anaemia, normal erythrocyte senescence and other forms of acute haemolysis

    Re-thinking Photographic Portraiture of 21st Century Italian Migrants in the Arc Lémanique area

    Contemporary history has identified three big Italian migration movements to Switzerland: the big diaspora of the end of the 19th century, the period between the First and the Second World Wars and the diaspora movement from the end of the Second World War until the 1970s. After the Schwarzenbach initiatives and the oil crisis of the 1970s, Italian migration to Switzerland has decreased. However, with the beginning of the 2007 – 2008 global economic crisis, Italy has witnessed a large new diaspora movement to the traditional countries of migration. Academic studies have found that the most diffused narrative on the brain drain phenomenon is oversimplistic and needs to be re-examined and updated; notably that this diaspora in the age of globalization has new and complex layers. The aim of this research is to re-think photographic portrayals of the cultural identity of Italian migrants of the 21st century in the area between Lausanne and Geneva in Switzerland (arc lémanique). The work reflects on issues related to the formation of cultural identity formation of the migrant as a continuing experience and to the sense of belonging to a space which becomes a known place after years of residency. How can an image capture a complex, ineffable, and unrepresentable notion of modern Italian cultural identity? Furthermore, how can an image represent the cultural identity of a group of people whose lives have been changed by the experience of physically migrating to a terra incognita? Research conducted in the archives of Swiss-French illustrated magazines (L’Illustré and L’Hebdo) and other online publications has highlighted distinctive photographic representations of Italian migrants in Switzerland from the 1960s-1970s to the period between 2006 and 2019. The analysis of pictures of Italian migrants made it possible to understand on a larger scale how the public imagination is shaped, how it impacts on the politics of the ‘other’ and how society reacts to the phenomena of migration. This process has supported and influenced the development of the photographic practice of this research in its attempt to render visible the complex notion of this new migratory identity

    Doing Justice to the Past: Memory and criticism in Herbert Marcuse

    In his inaugural lecture as director of the Institute for Social Research at the University of Frankfurt (1933), Horkheimer points out the need for a new understanding of history that avoids the contemporary versions of the Hegelian Verklärung. He synthesizes this challenge with an imperative: to do justice to past suffering. The result of this appeal can be found in the works of the members of the Frankfurt School in the form of multiple, even divergent, trains of thought that reach with unlike intensities the current debates on memory and its link with history. This paper focuses on three of these trains, which can be traced back to different periods of the work of Herbert Marcuse. It intends to systematize and present what can be considered alternative?although not necessarily contradictory?approaches aroused from the same concern over the critical power of nonreconciliatory memory: first, a genealogy inquiry that deconstructs the reified character of the given; second, a recollection of past images of happiness; and finally, a memory of the limits of all attainable freedom. Exploring these three moments, their shortcomings and tensions, may shed light on the complexity and present importance of the challenge they intend to face.Fil: Arese, Laura.
