Effect of oleic acid supplementation on prostaglandin production in maternal endometrial and fetal allantochorion cells isolated from late gestation ewes
Elevated circulating non-esterified fatty acids including oleic acid (OA) are associated with many pregnancy related complications. Prostaglandins (PGs) play crucial roles during parturition. We investigated the effect of OA supplementation on PG production using an in vitro model of ovine placenta
Proven invasive pulmonary mucormycosis successfully treated with amphotericin B and surgery in patient with acute myeloblastic leukemia: a case report
INTRODUCTION: Invasive mucormycosis (zygomycosis) is the third most frequent fungal infection in patients with hematologic malignancies. It often results in a fatal outcome mainly due to the difficulty of early diagnosis and its resistance to antimycotics. CASE PRESENTATION: A 52-year-old Caucasian man was diagnosed with acute myeloblastic leukemia. Following the induction chemotherapy he developed febrile neutropenia. Meropenem (3×1000mg/day) was introduced empirically. A chest computed tomography showed soft-tissue consolidation change in his right upper lobe. A bronchoscopy was performed and the histology indicated invasive pulmonary aspergillosis based on fungal hypha detection. Also, high risk patients are routinely screened for invasive fungal infections using commercially available serological enzyme-linked immunosorbent assay tests: galactomannan and mannan (Bio-Rad, France), as well as anti-Aspergillus immunoglobulin G and/or immunoglobulin M and anti-Candida immunoglobulin G and/or immunoglobulin M antibodies (Virion-Serion, Germany). Galactomannan showed low positivity and voriconazole therapy (2×400mg/first day; 2×300mg/following days) was implemented. The patient became afebrile and a partial remission of disease was established. After 2 months, the patient developed a fever and a chest multi-slice computed tomography showed soft-tissue mass compressing his upper right bronchus. Voriconazole (2×400mg/first day; 2×300mg/following days) was reintroduced and bronchoscopy was repeated. Histologic examination of the new specimen was done, as well as a revision of the earlier samples in the reference laboratory and the diagnosis was switched to invasive pulmonary mucormycosis. The treatment was changed to amphotericin B colloidal dispersion (1×400mg/day). The complete remission of acute myeloblastic leukemia was verified after 2 months. During his immunerestitution, a high positivity of the anti-Aspergillus immunoglobulin M antibodies was found in a single serum sample and pulmonary radiography was unchanged. A lobectomy of his right upper pulmonary lobe was done and the mycology culture of the lung tissue sample revealed Rhizopus oryzae. He remained in complete remission for more than 1 year. CONCLUSIONS: Invasive mucormycosis was successfully treated with amphotericin B, surgery and secondary itraconazole prophylaxis. As a rare disease invasive mucormycosis is not well understood by the medical community and therefore an improvement of education about prevention, diagnosis and treatment of invasive mucormycosis is necessary
<i>TP53</i> drives invasion through expression of its ∆133p53β variant
International audienceTP53 is conventionally thought to prevent cancer formation and progression to metastasis, while mutant TP53 has transforming activities. However, in the clinic, TP53 mutation status does not accurately predict cancer progression. Here we report, based on clinical analysis corroborated with experimental data, that the p53 isoform D133p53b promotes cancer cell invasion, regardless of TP53 mutation status. D133p53b increases risk of cancer recurrence and death in breast cancer patients. Furthermore D133p53b is critical to define invasiveness in a panel of breast and colon cell lines, expressing WT or mutant TP53. Endogenous mutant D133p53b depletion prevents invasiveness without affecting mutant full-length p53 protein expression. Mechanistically WT and mutant D133p53b induces EMT. Our findings provide explanations to 2 long-lasting and important clinical conundrums: how WT TP53 can promote cancer cell invasion and reciprocally why mutant TP53 gene does not systematically induce cancer progression
A rapid and reliable determination of doxycycline hyclate by HPLC with UV detection in pharmaceutical samples
An accurate, sensitive and reproducible high performance liquid chromatographic (HPLC) method for the quantification of doxycycline hyclate in pharmaceutical samples has been developed and validated. The drug and the standard were eluted from a Lichrosorb RP-8 (250 mm´4.6 mm, 10 mm particle size) at 20 °C with a mobile phase consisting of methanol, acetonitrile and 0.010 M aqueous solution of oxalic acid (2:3:5, v/v/v). The flow rate was 1.25 ml min-1. A UV detector set at 350 nm was used to monitor the effluent. Each analysis required no longer than 4 min. The limits of detection and quantification were 1.15 and 3.84 μg ml-1, respectively. Recoveries for different concentrations ranged from 99.58 to 101.93 %
Preserving Jewish Heritage : Conversas after the Establishment of the Barcelona Tribunal (1487-1505)
The study explores the world of Jewish women conversas and their devotion in the perpetuation of Jewish legacy after the reestablishment of the Barcelona Tribunal in 1487 until 1505. This investigation, based on the manuscript Liber descriptionis reconsiliationisque, purgationis et condemmnationis hereticorum ALIAS de Gestis Hereticorum written by the inquisitorial notary of the Barcelona Tribunal, Pere Miquel Carbonell, will furnish new insights into the lives and spiritual endeavour of these heroic women who faced the wrath of the Inquisition. Through a close and fine-grained analysis of the trial records and legal proceedings of the Barcelona Tribunal, this study will provide a better comprehension of a complex period and a fuller reconstruction of the conversas' religious practices as a system with its own structure
Relation Between Quality of Executive Functions, Attention and Religious Beliefs in Elderly Population
The presence of the religious beliefs in elderly may affect the self-control, socialization and the quality of life. We will try to determine the relation between the quality of executive functions, attention and the religious beliefs in elderly. This research included 50 subjects (27 female and 23 male). The following instruments were used: Questionnaire for the religiousness assessment; MMSE for cognitive condition assessment; WCST for the executive function assessment; TMT A B for the attention assessment. The higher degree of efficiency in achievement had the respondents who did not belong to the group of the religious estimating executive functions and attention, with statistical significance (p<.01). Statistically significant relation between the quality of executive function and attention and negative emotional experience was present in respondents who belonged to the group of the religious (p<.01). The respondents with the efficient executive functions and attention don`t belong to the group of the religious ones
What is the potential of p53 isoforms as a predictive biomarker in the treatment of cancer?
Introduction: For decades, p53 was researched as a single protein with alterations described as mutants. The discovery of 12 human p53 isoforms expressed from 9 transcripts changed this perception, eloquently explaining the numerous roles p53 plays, including apoptosis, senescence, and regeneration. Area covered: Here, we summarise the p53 isoforms and their relevance to cancer to establish an understanding and theorise on potential applications of the isoforms in clinical practice. Expert commentary: Pertaining to the different expression of isoforms in different tumors, it is concluded that the clinical use of isoforms as prognostic and predictive biomarkers will be different depending on the cell type, the tissue origin of the tumors, the position of the TP53 mutation and the driver-oncogene.</p
