A phase I randomized therapeutic MVA-B vaccination improves the magnitude and quality of the T cell immune responses in HIV-1-infected subjects on HAART
Trial Design
Previous studies suggested that poxvirus-based vaccines might be instrumental in the therapeutic
HIV field. A phase I clinical trial was conducted in HIV-1-infected patients on highly
active antiretroviral therapy (HAART), with CD4 T cell counts above 450 cells/mm3 and
undetectable viremia. Thirty participants were randomized (2:1) to receive either 3 intramuscular
injections of MVA-B vaccine (coding for clade B HIV-1 Env, Gag, Pol and Nef antigens)
or placebo, followed by interruption of HAART.
Methods
The magnitude, breadth, quality and phenotype of the HIV-1-specific T cell response were
assayed by intracellular cytokine staining (ICS) in 22 volunteers pre- and post-vaccination.
Results
MVA-B vaccine induced newly detected HIV-1-specific CD4 T cell responses and expanded
pre-existing responses (mostly against Gag, Pol and Nef antigens) that were high in magnitude,
broadly directed and showed an enhanced polyfunctionality with a T effector memory
(TEM) phenotype, while maintaining the magnitude and quality of the pre-existing HIV-1-
specific CD8 T cell responses. In addition, vaccination also triggered preferential CD8+ T
cell polyfunctional responses to the MVA vector antigens that increase in magnitude after
two and three booster doses
New perspectives on evolutionary medicine: the relevance of microevolution for human health and disease
Evolutionary medicine (EM) is a growing field focusing on the evolutionary basis of human diseases and their changes through time. To date, the majority of EM studies have used pure theories of hominin macroevolution to explain the present-day state of human health. Here, we propose a different approach by addressing more empirical and health-oriented research concerning past, current and future microevolutionary changes of human structure, functions and pathologies. Studying generation-to-generation changes of human morphology that occurred in historical times, and still occur in present-day populations under the forces of evolution, helps to explain medical conditions and warns clinicians that their current practices may influence future humans. Also, analyzing historic tissue specimens such as mummies is crucial in order to address the molecular evolution of pathogens, of the human genome, and their coadaptations.Frank Jakobus Rühli and Maciej Henneber
Archaeometric analysis of ceramic production in Tiwanaku state (c.500-1000ce) : An exploratory study
The chemical and mineralogical characterization of seven ceramic fragments produced within Tiwanaku state (c.500-1000 ce) is reported. The instrumental techniques used included X-ray elemental and mineralogical chemical analysis, Raman spectroscopy, and scanning and light microscopy. The results indicate there are several clay types, although they show similarities, such as the use of a plant-based temper. The red colour of the decoration is hematite, and manganese oxides such as jacobsite are present in the black. The white colour is a mixture of gypsum and clay, and the orange is a mixture of hematite and clay. The use of colours, the quality of the clays and the temperatures reached during pottery firing point to expertise in ceramic production and to complex decision-making processes. The multi-elemental archaeometric approach documented here could become an important tool to shed a light on ancient ceramic technology and the internal variance of Tiwanaku pottery.Peer reviewe
Violence in hunters, fishermen, and gatherers of the Chinchorro culture: Archaic societies of the Atacama Desert (10,000–4,000 cal yr BP)
Objectives: This article addresses evidence of violence imbedded in both soft and hard tissues from early populations of hunters, fishermen, and gatherers, known as the Chinchorro culture, who lived between 10,000 and 4,000 cal yr BP, along the coast of the Atacama Desert, one of the driest environments on Earth. Our study is aimed to test two hypotheses (a) that interactions and violent behaviors increased through time as population density and social complexity augmented; and (b) that violence was more prevalent between local Chinchorro groups and groups from other inland locations. Material and Methods: Two lines of data were analyzed: (1) bioarchaeology, through the quantification of physical traces of interpersonal violence in skeletons and mummies from a sample of 136 adult individuals and, (2) isotopic chemical analysis (strontium) of individuals with traces of trauma in order to determine their local or foreign origin. Results: Violence among Chinchorro populations was ubiquitous and remained invariant over time, with a remarkable skew to male (about 25% above female across the complete sample). Moreover, the chemical signature of individuals with traces of violence was not of foreign origin. Discussion: The violence exerted by the Chinchorro groups was not related to increased population size, nor social complexity and was mostly restricted to individuals coming from the same coastal habitat. That is, our data suggest that violence was constant across the Archaic period among the Chinchorro, implying that violent behavior was part of the sociocultural repertory of these populations, likely associated to mechanisms to resolve conflicts and social tensions
Population dynamics of an RNA virus and its defective interfering particles in passage cultures
<p>Abstract</p> <p>Background</p> <p>Viruses can fall prey to their defective interfering (DI) particles. When viruses are cultured by serial passage on susceptible host cells, the presence of virus-like DI particles can cause virus populations to rise and fall, reflecting predator-prey interactions between DI and virus particles. The levels of virus and DI particles in each population passage can be determined experimentally by plaque and yield-reduction assays, respectively.</p> <p>Results</p> <p>To better understand DI and virus particle interactions we measured vesicular stomatitis virus and DI particle production during serial-passage culture on BHK cells. When the multiplicity of infection (MOI, or ratio of infectious virus particles to cells) was fixed, virus yields followed a pattern of progressive decline, with higher MOI driving earlier and faster drops in virus level. These patterns of virus decline were consistent with predictions from a mathematical model based on single-passage behavior of cells co-infected with virus and DI particles. By contrast, the production of virus during fixed-volume passages exhibited irregular fluctuations that could not be described by either the steady-state or regular oscillatory dynamics of the model. However, these irregularities were, to a significant degree, reproduced when measured host-cell levels were incorporated into the model, revealing a high sensitivity of virus and DI particle populations to fluctuations in available cell resources.</p> <p>Conclusions</p> <p>This study shows how the development of mathematical models, when guided by quantitative experiments, can provide new insight into the dynamic behavior of virus populations.</p
Structural and functional studies reveal the molecular basis of substrate promiscuity of a glycosyltransferase originating from a major agricultural pest
The two-spotted spider mite, Tetranychus urticae, is a major cosmopolitan pest that feeds on more than 1100 plant species. Its genome contains an unprecedentedly large number of genes involved in detoxifying and transporting xenobiotics, including 80 genes that code for UDP glycosyltransferases (UGTs). These enzymes were acquired via horizontal gene transfer from bacteria after loss in the Chelicerata lineage. UGTs are well-known for their role in phase II metabolism; however, their contribution to host adaptation and acaricide resistance in arthropods, such as T. urticae, is not yet resolved. TuUGT202A2 (Tetur22g00270) has been linked to the ability of this pest to adapt to tomato plants. Moreover, it was shown that this enzyme can glycosylate a wide range of flavonoids. To understand this relationship at the molecular level, structural, functional, and computational studies were performed. Structural studies provided specific snapshots of the enzyme in different catalytically relevant stages. The crystal structure of TuUGT202A2 in complex with UDP-glucose was obtained and site-directed mutagenesis paired with molecular dynamic simulations revealed a novel lid-like mechanism involved in the binding of the activated sugar donor. Two additional TuUGT202A2 crystal complexes, UDP-(S)-naringenin and UDP-naringin, demonstrated that this enzyme has a highly plastic and open-ended acceptor-binding site. Overall, this work reveals the molecular basis of substrate promiscuity of TuUGT202A2 and provides novel insights into the structural mechanism of UGTs catalysis
The Roles of Standardization, Certification and Assurance Services in Global Commerce
In this article we examine the rapid emergence and expansion of standardized product and process frameworks and a private-sector compliance and enforcement infrastructure that we believe may increasingly be providing a substitute for public and legal regulatory infrastructure in global commerce. This infrastructure is provided by a proliferation of performance codes and standards, many of which define acceptable social and environmental behavior, and a rapidly-growing number of privately-trained and authorized inspectors and certifiers that we call the third-party assurance industry. We offer reasons for this development, evidence of its scope and scale, and then describe the phenomenon in more detail by examining supply chain arrangements in two industries, food products and apparel, where the use of third-party standards and assurance services has expanded especially rapidly. We conclude with a discussion of the implications for the make or buy decision at the core of the theory of the firm. We argue that as quasi-regulatory standards are developed within various industries, and as performance to these standards can be systematically evaluated using third-party inspectors and certifiers, the costs of moving production outside of vertical firm hierarchies drop. We believe this may be an important factor in accelerating the shift to outsourcing that has been observed over the last two decades

Taphonomic Criteria for Identifying Iberian Lynx Dens in Quaternary Deposits
For decades, taphonomists have dedicated their efforts to assessing the nature of the massive leporid accumulations recovered at archaeological sites in the northwestern Mediterranean region. Their interest lying in the fact that the European rabbit constituted a critical part of human subsistence during the late Pleistocene and early Holocene. However, rabbits are also a key prey in the food webs of Mediterranean ecosystems and the base of the diet for several specialist predators, including the Iberian lynx (Lynx pardinus). For this reason, the origin of rabbit accumulations in northwestern Mediterranean sites has proved a veritable conundrum. Here, we present the zooarchaeological and taphonomic study of more than 3000 faunal and 140 coprolite remains recovered in layer IIIa of Cova del Gegant (Catalonia, Spain). Our analysis indicates that this layer served primarily as a den for the Iberian lynx. The lynxes modified and accumulated rabbit remains and also died at the site creating an accumulation dominated by the two taxa. However, other agents and processes, including human, intervened in the final configuration of the assemblage. Our study contributes to characterizing the Iberian lynx fossil accumulation differentiating between the faunal assemblages accumulated by lynxes and hominins
