    Distinguishing Posed and Spontaneous Smiles by Facial Dynamics

    Smile is one of the key elements in identifying emotions and present state of mind of an individual. In this work, we propose a cluster of approaches to classify posed and spontaneous smiles using deep convolutional neural network (CNN) face features, local phase quantization (LPQ), dense optical flow and histogram of gradient (HOG). Eulerian Video Magnification (EVM) is used for micro-expression smile amplification along with three normalization procedures for distinguishing posed and spontaneous smiles. Although the deep CNN face model is trained with large number of face images, HOG features outperforms this model for overall face smile classification task. Using EVM to amplify micro-expressions did not have a significant impact on classification accuracy, while the normalizing facial features improved classification accuracy. Unlike many manual or semi-automatic methodologies, our approach aims to automatically classify all smiles into either `spontaneous' or `posed' categories, by using support vector machines (SVM).     Validating child vaccination status in a demographic surveillance system using data from a clinical cohort study: evidence from rural South Africa

    <p><b>Background:</b> Childhood vaccination coverage can be estimated from a range of sources. This study aims to validate vaccination data from a longitudinal population-based demographic surveillance system (DSS) against data from a clinical cohort study.</p> <p><b>Methods:</b> The sample includes 821 children in the Vertical Transmission cohort Study (VTS), who were born between December 2001 and April 2005, and were matched to the Africa Centre DSS, in northern KwaZulu-Natal. Vaccination information in the surveillance was collected retrospectively, using standardized questionnaires during bi-annual household visits, when the child was 12 to 23 months of age. DSS vaccination information was based on extraction from a vaccination card or, if the card was not available, on maternal recall. In the VTS, vaccination data was collected at scheduled maternal and child clinic visits when a study nurse administered child vaccinations. We estimated the sensitivity of the surveillance in detecting vaccinations conducted as part of the VTS during these clinic visits.</p> <p><b>Results:</b> Vaccination data in matched children in the DSS was based on the vaccination card in about two-thirds of the cases and on maternal recall in about one-third. The sensitivity of the vaccination variables in the surveillance was high for all vaccines based on either information from a South African Road-to-Health (RTH) card (0.94-0.97) or maternal recall (0.94-0.98). Addition of maternal recall to the RTH card information had little effect on the sensitivity of the surveillance variable (0.95-0.97). The estimates of sensitivity did not vary significantly, when we stratified the analyses by maternal antenatal HIV status. Addition of maternal recall of vaccination status of the child to the RTH card information significantly increased the proportion of children known to be vaccinated across all vaccines in the DSS.</p> <p><b>Conclusion:</b> Maternal recall performs well in identifying vaccinated children aged 12-23 months (both in HIV-infected and HIV-uninfected mothers), with sensitivity similar to information extracted from vaccination cards. Information based on both maternal recall and vaccination cards should be used if the aim is to use surveillance data to identify children who received a vaccination.</p&gt

    Effect of photoperiod and temperature on ovarian cycle of the frog Rana tigrina (Daud.)

    The effect of varying photoperiod regimes (LD: 20,4; 4,20; 6,18; 18,6 and 12,12) on ovarian follicular development was analysed in the frogRana tigrina maintained at ambient and constant 30° ± l° C for 3 months. The experiments were conducted in early recrudescent and quiescent phases. The frogs were fed guppiesad libitum on alternate day. None of the photoperiod regimes had any effect on the ovaries or the fat bodies, whereas exposure to constant high temperature (regardless of photoperiod) during recrudescent phase induced production of greater number of eggs (~ 18000 vs 13000 in controls) of ovulatory sizes (> 1400µm) compared to the corresponding controls maintained at ambient temperature. Hence, ovarian mass also increased in these frogs. In the quiescent phase, high temperature merely enhanced growth of previtellogenic oocytes. In both the phases high temperature caused a reduction in the fat bodies over the respective controls, possibly due to increased metabolic activity. The above findings indicate that temperature plays a key role in the regulation of ovarian cycle ofRana tigrina and that the photoperiodic mechanisms may not govern the annual recrudescence of ovaries in the frog. The study also shows that the frog exhibits the phenomenon of "phenotypic plasticity" in its reproductive behaviour by producing significantly greater number of eggs in response to elevated temperature

    Coupling Computational Fluid Dynamics Simulations and Statistical Moments for Designing Healthy Indoor Spaces

    Cross-contamination between occupants in an indoor space may occur due to transfer of infectious aerosols. Computational fluid dynamics (CFD) provides detailed insight into particle transport in indoor spaces. However, such simulations are site-specific. This study couples CFD with statistical moments and establishes a framework that transitions site-specific results to generating guidelines for designing “healthy” indoor spaces. Eighteen cases were simulated, and three parameters were assessed: inlet/outlet location, air changes per hour, and the presence/absence of desks. Aerosol release due to a simulated “sneeze” in a two-dimensional ventilated space was applied as a test case. Mean, standard deviation, and skewness of the velocity profiles and particle locations gave an overall picture of the spread and movement of the air flow in the domain. A parameter or configuration did not dominate the values, confirming the significance of considering the combined influence of multiple parameters for determining localized air-flow characteristics. Particle clustering occurred more when the inlet was positioned above the outlet. The particle dispersion pattern could be classified into two time zones: “near time”, \u3c60 \u3es, and “far time”, \u3e120 s.     Dynamics of oogenesis in the tropical anuran Rana tigrina (Amphibia: Ranidae) with special reference to vitellogenic cycles in wild-caught and captive flogs

    The ovarian cycle of Rana tigrina was analysed by quantifying the developing oocytes (classified into stages on the basis of diameter) and atretic ones at monthly intervals. Stages I to IV represent oocytes in the first growth phase and the remaining ones the vitellogenic or second growth phase. Stages I-III occurred year round but exhibited significant variation in their number. The number of stage II oocytes always dominated the other stages. Recruitment of oocytes to stages IV and V in April marked the initiation of vitellogenic growth in all specimens. Of the 30 to 35% second growth phase oocytes, 25 to 28% reached ovulatory sizes by June. After spawning the ovarian mass declined drastically from 15 to 0.2% of body mass in July. Atresia was maximal (5%) in August. In other months, it was less than 1.5% of the total oocytes. Oogenic episodes occurred in March and July yielding new oocytes. The number of first growth phase oocytes fluctuated from 65 to 95%. The fluctuation was inversely correlated with the second growth phase oocytes indicating a 30 to 35% annual turnover rate of oocytes in the frog. The final egg number/ovarian mass is positively correlated with the snout-vent length as well as body mass of the frogs. R. tigrina produces about 4000 eggs/100g body mass. Further, the mean number of yolky eggs/100 g body mass and the total volume (V) of eggs/frog were highly correlated. Frogs living in captivity produced fewer eggs compared to the wild ones (3594 ± 227 in captive vs 4704 ± 317 in wild frogs). Also, these frogs failed to breed though they showed amplexus with breeding males. Injection of desoxycorticosterone acetate however induced spawning in 4 out of 5 frogs. They released about 3000 eggs each. Captivity seems to mainly impair breeding and to a little extent the vitellogenic growth of oocytes in R. tigrina

    Bisulfite-free analysis of 5MeC-binding proteins and locus-specific methylation density using a microparticle-based flow cytometry assay

    DNA methylation analysis is emerging as a new technique with potential capabilities for early cancer detection. However, current state-of-the-art techniques are not easily translatable into routine clinical methods. Herein we describe a bead-based flow cytometry assay which combines DNA hybridization to microparticles with 5MeC-specific proteins/antibodies. These assays can be used to study the binding properties of current and emerging 5MeC-binding proteins and may also have potential in the measurement of 5MeC density in clinical samples for cancer detection

    Microsatellite alteration and immunohistochemical expression profile of chromosome 9p21 in patients with sporadic renal cell carcinoma following surgical resection.

    BACKGROUND: Long-term prognostic significance of loss of heterozygosity on chromosome 9p21 for localized renal cell carcinoma following surgery remains unreported. The study assessed the frequency of deletions of different loci of chromosome 9p along with immunohistochemical profile of proteins in surgically resected renal cancer tissue and correlated this with long-term outcomes. METHODS: DNA was extracted from renal tumours and corresponding normal kidney tissues in prospectively collected samples of 108 patients who underwent surgical resection for clinically localized disease between January 2001 and December 2005, providing a minimum of 9 years follow-up for each participant. After checking quality of DNA, amplified by PCR, loss of heterozygosity (LOH) on chromosome 9p was assessed using 6 microsatellite markers in 77 clear cell carcinoma. Only 5 of the markers showed LOH (D9S1814, D9S916, D9S974, D9S942, and D9S171). Protein expression of p15(INK4b), p16(INK4a), p14(ARF), CAIX, and adipose related protein (ADFP) were demonstrated by immunostaining in normal and cancer tissues. Loss of heterozygosity for microsatellite analysis was correlated with tumour characteristics, recurrence free, cancer specific, and overall survival, including significance of immunohistochemical profile of protein expressions. RESULTS: The main deletion was found at loci telomeric to CDKN2A region at D9S916. There was a significant correlation between frequency of LOH stage (p = 0.005) and metastases (p = 0.006) suggesting a higher LOH for advanced and aggressive renal cell carcinoma. Most commonly observed LOH in the 3 markers: D9S916, D9S974, and D9S942 were associated with poor survival, and were statistically significant on multivariate analysis. Immunohistochemical expression of p14, p15, and p16 proteins were either low or absent in cancer tissue compared to normal. CONCLUSIONS: Loss of heterozygosity of p921 chromosome is associated with aggressive tumours, and predicts cancer specific or recurrence free survival on long-term follow-up.
