    Infection with host-range mutant adenovirus 5 suppresses innate immunity and induces systemic CD4+ T cell activation in rhesus macaques.

    Ad5 is a common cause of respiratory disease and an occasional cause of gastroenteritis and conjunctivitis, and seroconversion before adolescence is common in humans. To gain some insight into how Ad5 infection affects the immune system of rhesus macaques (RM) 18 RM were infected with a host-range mutant Ad5 (Ad5hr) by 3 mucosal inoculations. There was a delay of 2 to 6 weeks after the first inoculation before plasmacytoid dendritic cell (pDC) frequency and function increased in peripheral blood. Primary Ad5hr infection suppressed IFN-γ mRNA expression, but the second Ad5hr exposure induced a rapid increase in IFN-gamma mRNA in peripheral blood mononuclear cells (PBMC). Primary Ad5hr infection suppressed CCL20, TNF and IL-1 mRNA expression in PBMC, and subsequent virus exposures further dampened expression of these pro-inflammatory cytokines. Primary, but not secondary, Ad5hr inoculation increased the frequency of CXCR3+ CD4+ T cells in blood, while secondary, but not primary, Ad5hr infection transiently increased the frequencies of Ki67+, HLADR+ and CD95+/CCR5+ CD4+ T cells in blood. Ad5hr infection induced polyfunctional CD4 and CD8+ T cells specific for the Ad5 hexon protein in all of the animals. Thus, infection with Ad5hr induced a complex pattern of innate and adaptive immunity in RM that included transient systemic CD4+ T cell activation and suppressed innate immunity on re-exposure to the virus. The complex effects of adenovirus infection on the immune system may help to explain the unexpected results of testing Ad5 vector expressing HIV antigens in Ad5 seropositive people

    In this paper, we look at a specific issue with practical Turing tests, namely the right of the machine to remain silent during interrogation. In particular, we consider the possibility of a machine passing the Turing test simply by not saying anything. We include a number of transcripts from practical Turing tests in which silence has actually occurred on the part of a hidden entity. Each of the transcripts considered here resulted in a judge being unable to make the ‘right identification’, i.e., they could not say for certain which hidden entity was the machine.     Machine humour: examples from Turing test experiments

    In this paper, we look at the possibility of a machine having a sense of humour. In particular, we focus on actual machine utterances in Turing test discourses. In doing so, we do not consider the Turing test in depth and what this might mean for humanity, rather we merely look at cases in conversations when the output from a machine can be considered to be humorous. We link such outpourings with Turing’s “arguments from various disabilities” used against the concept of a machine being able to think, taken from his seminal work of 1950. Finally we consider the role that humour might play in adding to the deception, integral to the Turing test, that a machine in practice appears to be a human

    An intersectional analysis of early-career young cisgender women teachers’ experiences of learner-on-teacher violence

    Background: Learner-on-teacher violence is a growing global concern, and South African schools are not exempt from this issue. However, the specific experiences of early-career young cisgender women teachers remain underexplored, despite their heightened vulnerability at the intersection of sex, gender, age, physical appearance and structural factors such as inadequate institutional support. Objectives: This article presents an intersectional analysis of how intersecting identity markers, such as sex, gender and age, alongside physical appearance and structural vulnerabilities, shape the heightened susceptibility of early-career young cisgender women teachers to violence from male learners. Method: The study used qualitative research, conducting eight interviews with early-career young cisgender women teachers from secondary schools in Tshwane, Gauteng. Results: The findings show that young cisgender women teachers are especially vulnerable to male learner violence, influenced by a small age gap and patriarchal gender ideologies. This vulnerability impacts their psychological well-being, affecting their interactions with learners and the school environment. Conclusion: The intersection of sex, gender, age, physical appearance and structural vulnerabilities produces a distinct form of learner-on-teacher violence, underscoring the specific risks faced by early-career cisgender women teachers. This vulnerability requires targeted attention and intervention to protect them from male learner violence. Collaborative efforts from the Department of Education and schools are necessary to support and safeguard these teachers, especially in schools with a history of violence. Contribution: This study enhances our understanding of how male learner violence affects the teaching experiences and well-being of early-career young cisgender women teachers in secondary schools

    First molecular documented case of a rarely reported parasite:Plasmodium knowlesi infection in Denmark in a traveller returning from Malaysian Borneo

    Plasmodium knowlesi has been reported as an emerging infection throughout the Southeast Asian region, especially in the Malaysian state of Sabah, where it accounts for the majority of the malaria cases reported. This is in contrast to Europe, where imported P. knowlesi is a rarely reported infection. We present a case of P. knowlesi infection in a Danish woman returning from a short trip to Malaysian Borneo. Microscopy of blood smears revealed 0.8% infected erythrocytes, but due to the atypical morphological presentation, a conclusive species identification was made by molecular methods. Plasmodium knowlesi is a potentially fatal infection and taking the increasing travel activity into consideration after the coronavirus disease 2019 (COVID-19) pandemic, P. knowlesi should be a differential diagnosis in patients with travel-associated illness returning from highly endemic Southeast Asian areas.</p

    The experience of maternal mental distress in The Gambia: A qualitative study identifying idioms of distress, perceptions of contributing factors and the supporting role of existing cultural practices

    Maternal mental health problems are experienced frequently in the perinatal period and can be up to twice as common in women from low- and middle-income countries. The terms used to describe mental illness symptoms and the factors that contribute to developing these symptoms are affected by culture and context. Based on 14 focus group discussions held with pregnant women, health professionals and cultural leaders, this qualitative study aimed to understand women’s experiences of mental distress during the perinatal period in The Gambia. To do this it aimed to 1) identify the most commonly used idioms of distress, 2) identify the factors believed to affect women’s perinatal mental health and 3) explore the role of existing cultural practices, including musical practices, that were identified as locally significant in supporting maternal and mental health. Sondomoo tenkung baliyaa (Mandinka) and xel bu dalut (Wolof) were identified as the most commonly used idioms of distress which roughly translate to lack of a steady mind/heart. Using thematic analysis, six themes (Poverty of the healthcare system, Shifting cultural context, Economic factors, Social factors, Spiritual factors, and Cultural practices involving music) were identified to describe the factors that shape women’s experience of mental health during the perinatal period. Lack of economic resources, the prevailing poverty of the health system, an unsupportive husband and spiritual attack by evil spirits or witches were common reasons given for a woman experiencing maternal mental distress. Various existing cultural practices involving music, such as fertility societies, naming ceremonies and community music-making, were recognised as valuable for supporting women’s mental health during the perinatal period. This work emphasises that initiatives to support perinatal mental health should be grounded in an understanding of local community cultural practices, knowledge, and experiences

    Community psychosocial music intervention (CHIME) to reduce antenatal common mental disorder symptoms in The Gambia: a feasibility trial

    Objectives: Examine the feasibility of a Community Health Intervention through Musical Engagement (CHIME) in The Gambia to reduce common mental disorder (CMD) symptoms in pregnant women. Design: Feasibility trial testing a randomised stepped-wedge cluster design. Setting: Four local antenatal clinics. Participants: Women who were 14–24 weeks pregnant and spoke Mandinka or Wolof were recruited into the intervention (n=50) or control group (n=74). Intervention: Music-based psychosocial support sessions designed and delivered by all-female fertility societies. Sessions lasted 1 hour and were held weekly for 6 weeks. Delivered to groups of women with no preselection. Sessions were designed to lift mood, build social connection and provide health messaging through participatory music making. The control group received standard antenatal care. Outcomes: Demographic, feasibility, acceptability outcomes and the appropriateness of the study design were assessed. Translated measurement tools (Self-Reporting Questionnaire (SRQ-20); Edinburgh Postnatal Depression Scale (EPDS)) were used to assess CMD symptoms at baseline, post-intervention and 4-week follow-up. Results: All clinics and 82% of women approached consented to take part. A 33% attrition rate across all time points was observed. 72% in the intervention group attended at least three sessions. Audio and video analysis confirmed fidelity of the intervention and a thematic analysis of participant interviews demonstrated acceptability and positive evaluation. Results showed a potential beneficial effect with a reduction of 2.13 points (95% CI (0.89 to 3.38), p<0.01, n=99) on the SRQ-20 and 1.98 points (95% CI (1.06 to 2.90), p<0.01, n=99) on the EPDS at the post-intervention time point for the intervention group compared with standard care. Conclusion: Results demonstrate that CHIME is acceptable and feasible in The Gambia. To our knowledge, CHIME is the first example of a music-based psychosocial intervention to be applied to perinatal mental health in a low- and middle-income country context
