    Offspring subcutaneous adipose markers are sensitive to the timing of maternal gestational weight gain

    peer-reviewedBackground Excessive maternal weight gain during pregnancy impacts on offspring health. This study focused on the timing of maternal gestational weight gain, using a porcine model with mothers of normal pre-pregnancy weight. Methods Trial design ensured the trajectory of maternal gestational weight gain differed across treatments in early, mid and late gestation. Diet composition did not differ. On day 25 gestation, sows were assigned to one of five treatments: Control sows received a standard gestation diet of 2.3 kg/day (30 MJ DE/day) from early to late gestation (day 25–110 gestation). E sows received 4.6 kg food/day in early gestation (day 25–50 gestation). M sows doubled their food intake in mid gestation (day 50–80 gestation). EM sows doubled their food intake during both early and mid gestation (day 25–80 gestation). L sows consumed 3.5 kg food/day in late gestation (day 80–110 gestation). Offspring body weight and food intake levels were measured from birth to adolescence. Markers of lipid metabolism, hypertrophy and inflammation were investigated in subcutaneous adipose tissue of adolescent offspring. Results The trajectory of gestational weight gain differed across treatments. However total gestational weight gain did not differ except for EM sows who were the heaviest and fattest mothers at parturition. Offspring birth weight did not differ across treatments. Subcutaneous adipose tissue from EM offspring differed significantly from controls, with elevated mRNA levels of lipogenic (CD36, ACACB and LPL), nutrient transporters (FABP4 and GLUT4), lipolysis (HSL and ATGL), adipocyte size (MEST) and inflammation (PAI-1) indicators. The subcutaneous adipose depot from L offspring exhibited elevated levels of CD36, ACACB, LPL, GLUT4 and FABP4 mRNA transcripts compared to control offspring. Conclusions Increasing gestational weight gain in early gestation had the greatest impact on offspring postnatal growth rate. Increasing maternal food allowance in late gestation appeared to shift the offspring adipocyte focus towards accumulation of fat. Mothers who gained the most weight during gestation (EM mothers) gave birth to offspring whose subcutaneous adipose tissue, at adolescence, appeared hyperactive compared to controls. This study concluded that mothers, who gained more than the recommended weight gain in mid and late gestation, put their offspring adipose tissue at risk of dysfunction.This research was funded by Teagasc, under the National Development Plan.     Differences in intestinal size, structure, and function contributing to feed efficiency in broiler chickens reared at geographically distant locations

    The contribution of the intestinal tract to differences in residual feed intake (RFI) has been inconclusively studied in chickens so far. It is also not clear if RFI-related differences in intestinal function are similar in chickens raised in different environments. The objective was to investigate differences in nutrient retention, visceral organ size, intestinal morphology, jejunal permeability and expression of genes related to barrier function, and innate immune response in chickens of diverging RFI raised at 2 locations (L1: Austria; L2: UK). The experimental protocol was similar, and the same dietary formulation was fed at the 2 locations. Individual BW and feed intake (FI) of chickens (Cobb 500FF) were recorded from d 7 of life. At 5 wk of life, chickens (L1, n = 157; L2 = 192) were ranked according to their RFI, and low, medium, and high RFI chickens were selected (n = 9/RFI group, sex, and location). RFI values were similar between locations within the same RFI group and increased by 446 and 464 g from low to high RFI in females and males, respectively. Location, but not RFI rank, affected growth, nutrient retention, size of the intestine, and jejunal disaccharidase activity. Chickens from L2 had lower total body weight gain and mucosal enzyme activity but higher nutrient retention and longer intestines than chickens at L1. Parameters determined only at L1 showed increased crypt depth in the duodenum and jejunum and enhanced paracellular permeability in low vs. high RFI females. Jejunal expression of IL1B was lower in low vs. high RFI females at L2, whereas that of TLR4 at L1 and MCT1 at both locations was higher in low vs. high RFI males. Correlation analysis between intestinal parameters and feed efficiency metrics indicated that feed conversion ratio was more correlated to intestinal size and function than was RFI. In conclusion, the rearing environment greatly affected intestinal size and function, thereby contributing to the variation in chicken RFI observed across locations

    Birthweight and risk markers for type 2 diabetes and cardiovascular disease in childhood: the Child Heart and Health Study in England (CHASE).

    AIMS/HYPOTHESIS: Lower birthweight (a marker of fetal undernutrition) is associated with higher risks of type 2 diabetes and cardiovascular disease (CVD) and could explain ethnic differences in these diseases. We examined associations between birthweight and risk markers for diabetes and CVD in UK-resident white European, South Asian and black African-Caribbean children. METHODS: In a cross-sectional study of risk markers for diabetes and CVD in 9- to 10-year-old children of different ethnic origins, birthweight was obtained from health records and/or parental recall. Associations between birthweight and risk markers were estimated using multilevel linear regression to account for clustering in children from the same school. RESULTS: Key data were available for 3,744 (66%) singleton study participants. In analyses adjusted for age, sex and ethnicity, birthweight was inversely associated with serum urate and positively associated with systolic BP. After additional height adjustment, lower birthweight (per 100 g) was associated with higher serum urate (0.52%; 95% CI 0.38, 0.66), fasting serum insulin (0.41%; 95% CI 0.08, 0.74), HbA1c (0.04%; 95% CI 0.00, 0.08), plasma glucose (0.06%; 95% CI 0.02, 0.10) and serum triacylglycerol (0.30%; 95% CI 0.09, 0.51) but not with BP or blood cholesterol. Birthweight was lower among children of South Asian (231 g lower; 95% CI 183, 280) and black African-Caribbean origin (81 g lower; 95% CI 30, 132). However, adjustment for birthweight had no effect on ethnic differences in risk markers. CONCLUSIONS/INTERPRETATION: Birthweight was inversely associated with urate and with insulin and glycaemia after adjustment for current height. Lower birthweight does not appear to explain emerging ethnic difference in risk markers for diabetes

    Taming the Invisible Monster: System Parameter Constraints for Epsilon Aurigae from the Far-Ultraviolet to the Mid-Infrared

    We have assembled new Spitzer Space Telescope Infrared Array Camera observations of the mysterious binary star Epsilon Aurigae, along with archival far-ultraviolet to mid-infrared data, to form an unprecedented spectral energy distribution spanning three orders of magnitude in wavelength from 0.1 microns to 100 microns. The observed spectral energy distribution can be reproduced using a three component model consisting of a 2.2+0.9/-0.8 Msun F type post-asymptotic giant branch star, and a 5.9+/-0.8 Msun B5+/-1 type main sequence star that is surrounded by a geometrically thick, but partially transparent, disk of gas and dust. At the nominal HIPPARCOS parallax distance of 625 pc, the model normalization yields a radius of 135+/-5 Rsun for the F star, consistent with published interferometric observations. The dusty disk is constrained to be viewed at an inclination of i > 87 deg, and has effective temperature of 550+/-50 K with an outer radius of 3.8 AU and a thickness of 0.95 AU. The dust content of the disk must be largely confined to grains larger than ~10 microns in order to produce the observed gray optical-infrared eclipses and the lack of broad dust emission features in the archival Spitzer mid-infrared spectra. The total mass of the disk, even considering a potential gaseous contribution in addition to the dust that produces the observed infrared excess, is << 1 Msun. We discuss evolutionary scenarios for this system that could lead to the current status of the stellar components and suggests possibilities for its future evolution, as well as potential observational tests of our model.Comment: 13 pages, 3 figures.     Inflationary Scenarios from Branes at Angles

    We describe a simple mechanism that can lead to inflation within string-based brane-world scenarios. The idea is to start from a supersymmetric configuration with two parallel static Dp-branes, and slightly break the supersymmetry conditions to produce a very flat potential for the field that parametrises the distance between the branes, i.e. the inflaton field. This breaking can be achieved in various ways: by slight relative rotations of the branes with small angles, by considering small relative velocities between the branes, etc. If the breaking parameter is sufficiently small, a large number of e-folds can be produced within the D-brane, for small changes of the configuration in the compactified directions. Such a process is local, i.e. it does not depend very strongly on the compactification space nor on the initial conditions. Moreover, the breaking induces a very small velocity and acceleration, which ensures very small slow-roll parameters and thus an almost scale invariant spectrum of metric fluctuations, responsible for the observed temperature anisotropies in the microwave background. Inflation ends as in hybrid inflation, triggered by the negative curvature of the string tachyon potential.     Improving contemporary approaches to the master planning process.

    Master-planning has had a strong revival in recent years. However, significant demographic and social changes are on-going amidst the constraints of the current economic stagnation, the policy of reduced public spending and the drive to respond to environmental imperatives. These conditions challenge the feasibility of the application of past master-planning practice. The way we conceive of master-planning now requires re-visiting. The traditional perspective of master-planning as a design-led activity concerned with the architectural form of buildings, spaces and infrastructures is out-dated and inadequate to coordinating the plural processes of negotiating sustainable place development which, in addition to realising a visually pleasing townscape, critically satisfies social, functional, economic and environmental requirements. Masterplanning requires both a business planning component, without which there is no delivery, and a governance component, without which the physical strategy has no legitimacy. A more adaptive master-planning approach is required. The paper proposes how a flexible master-planning process can provide a basis of a suitable approach for the development of sustainable settlements.
