    A Cognitive Model of an Epistemic Community: Mapping the Dynamics of Shallow Lake Ecosystems

    We used fuzzy cognitive mapping (FCM) to develop a generic shallow lake ecosystem model by augmenting the individual cognitive maps drawn by 8 scientists working in the area of shallow lake ecology. We calculated graph theoretical indices of the individual cognitive maps and the collective cognitive map produced by augmentation. The graph theoretical indices revealed internal cycles showing non-linear dynamics in the shallow lake ecosystem. The ecological processes were organized democratically without a top-down hierarchical structure. The steady state condition of the generic model was a characteristic turbid shallow lake ecosystem since there were no dynamic environmental changes that could cause shifts between a turbid and a clearwater state, and the generic model indicated that only a dynamic disturbance regime could maintain the clearwater state. The model developed herein captured the empirical behavior of shallow lakes, and contained the basic model of the Alternative Stable States Theory. In addition, our model expanded the basic model by quantifying the relative effects of connections and by extending it. In our expanded model we ran 4 simulations: harvesting submerged plants, nutrient reduction, fish removal without nutrient reduction, and biomanipulation. Only biomanipulation, which included fish removal and nutrient reduction, had the potential to shift the turbid state into clearwater state. The structure and relationships in the generic model as well as the outcomes of the management simulations were supported by actual field studies in shallow lake ecosystems. Thus, fuzzy cognitive mapping methodology enabled us to understand the complex structure of shallow lake ecosystems as a whole and obtain a valid generic model based on tacit knowledge of experts in the field.Comment: 24 pages, 5 Figure

    The Structure of the Littoral Invertebrate Communities of the Kosciuszko Region Lakes

    The littoral macroinvertebrate assemblages of Lakes Albina, Blue, Club andCootapatamba in the Mt. Kosciuszko region were sampled by two methods: sweeps and cobble picks. Thirty-six species were collected with total abundance and species richness greatest in Lake Albina. Common species included themolluscs Pisidium kosciusko and Glacidorbis hedleyi, the crustaceans Metaphreatoicus australis and Neoniphragus n. sp., and an unidentified limnephidtrichoperan. Community structure was influenced by the nature of the substrate, with cobble sites having greater richness and abundance than boulder sites. The importance of the major taxonomic groups (crustaceans, insects and molluscs) varied with sampling method and among lakes. Crustaceans (isopods and amphipods) usually dominated in sweeps, with molluscs and insects varying in importance among lakes, while insects mostly dominated in cobble pick samples. Oligotrophic lakes are typically thought to be dominated by insects, however this study shows such lakes may appear to be dominated by insects or crustaceans depending on the sampling method used and the presence of fish

    Long-term responders on olaparib maintenance in high-grade serous ovarian cancer: Clinical and molecular characterization

    Purpose: Maintenance therapy with olaparib has improved progression-free survival in women with high-grade serous ovarian cancer (HGSOC), particularly those harboring BRCA1/2 mutations. The objective of this study was to characterize long-term (LT) versus short-term (ST) responders to olaparib. Experimental Design: A comparative molecular analysis of Study 19 (NCT00753545), a randomized phase II trial assessing olaparib maintenance after response to platinum-based chemotherapy in HGSOC, was conducted. LT response was defined as response to olaparib/placebo > 2 years, ST as < 3 months. Molecular analyses included germline BRCA1/2 status, three-biomarker homologous recombination deficiency (HRD) score, BRCA1 methylation, and mutational profiling. Another olaparib maintenance study (Study 41; NCT01081951) was used as an additional cohort. Results: Thirty-seven LT (32 olaparib) and 61 ST (21 olaparib) patients were identified. Treatment was significantly associated with outcome (P < 0.0001), with more LT patients on olaparib (60.4%) than placebo (11.1%). LT sensitivity to olaparib correlated with complete response to chemotherapy (P < 0.05). In the olaparib LT group, 244 genetic alterations were detected, with TP53, BRCA1, and BRCA2 mutations being most common (90%, 25%, and 35%, respectively). BRCA2 mutations were enriched among the LT responders. BRCA methylation was not associated with response duration. High myriad HRD score (>42) and/or BRCA1/2 mutation was associated with LT response to olaparib. Study 41 confirmed the correlation of LT response with olaparib and BRCA1/2 mutation. Conclusions: Findings show that LT response to olaparib may be multifactorial and related to homologous recombination repair deficiency, particularly BRCA1/2 defects. The type of BRCA1/2 mutation warrants further investigation. (C) 2017 AACR

    Patterns of genomic loss of heterozygosity predict homologous recombination repair defects in epithelial ovarian cancer

    Background: Defects in BRCA1, BRCA2, and other members of the homologous recombination pathway have potential therapeutic relevance when used to support agents that introduce or exploit double-stranded DNA breaks. This study examines the association between homologous recombination defects and genomic patterns of loss of heterozygosity (LOH). Methods: Ovarian tumours from two independent data sets were characterised for defects in BRCA1,BRCA2, and RAD51C, and LOH profiles were generated. Publically available data were downloaded for a third independent data set. The same analyses were performed on 57 cancer cell lines. Results: Loss of heterozygosity regions of intermediate size were observed more frequently in tumours with defective BRCA1 or BRCA2 (P=10−11). The homologous recombination deficiency (HRD) score was defined as the number of these regions observed in a tumour sample. The association between HRD score and BRCA deficiency was validated in two independent ovarian cancer data sets (P=10−5 and 10−29), and identified breast and pancreatic cell lines with BRCA defects. Conclusion: The HRD score appears capable of detecting homologous recombination defects regardless of aetiology or mechanism. This score could facilitate the use of PARP inhibitors and platinum in breast, ovarian, and other cancers

    Lithium-ion battery degradation: how to model it

    Predicting lithium-ion battery degradation is worth billions to the global automotive, aviation and energy storage industries, to improve performance and safety and reduce warranty liabilities. However, very few published models of battery degradation explicitly consider the interactions between more than two degradation mechanisms, and none do so within a single electrode. In this paper, the first published attempt to directly couple more than two degradation mechanisms in the negative electrode is reported. The results are used to map different pathways through the complicated path dependent and non-linear degradation space. Four degradation mechanisms are coupled in PyBaMM, an open source modelling environment uniquely developed to allow new physics to be implemented and explored quickly and easily. Crucially it is possible to see 'inside' the model and observe the consequences of the different patterns of degradation, such as loss of lithium inventory and loss of active material. For the same cell, five different pathways that can result in end-of-life have already been found, depending on how the cell is used. Such information would enable a product designer to either extend life or predict life based upon the usage pattern. However, parameterization of the degradation models remains as a major challenge, and requires the attention of the international battery community

    ANTIBACTERIAL PROPERTIES OF ZINC OXIDE NANOPARTICLES SYNTHESIZED BY THE SUPERNATANT OF WEISSELLA CONFUSA UPM22MT04

    This study was aimed to produce zinc oxide nanoparticles (ZnO-NPs) using the supernatant of Weissella confusa UPM22MT04 and assess their effectiveness in inhibiting methicillin-resistant Staphylococcus aureus (MRSA). An isolate of Weissella confusa UPM22MT04 was isolated from a wastewater treatment plant in Johor, Malaysia, and was utilized to synthesize ZnO-NPs. The synthesized ZnO-NPs were characterized through several techniques, including UV-visible spectroscopy, Fourier-transform infrared spectroscopy, transmission electron microscopy, energy-dispersive X-ray spectroscopy, and dynamic light scattering. Monodisperse spherical ZnO-NPs of 1.7 - 7.9 nm were obtained with 0.1 M zinc nitrate at 80°C. The biosynthesized ZnO-NPs exhibited vigorous inhibitory activity against MRSA. Results found that ZnO-NPs inhibited MRSA at a minimum concentration of 0.625 mg/mL and were bactericidal at a minimum concentration of 1.25 mg/mL. In MTT assays, ZnO-NPs showed no toxicity to HS-27 fibroblasts. The supernatant of Weissella confusa UPM22MT04 could be used to synthesize ZnO-NPs, which are an antibacterial agent, eco-friendly and non-toxic

    Early crustal processes revealed by the ejection site of the oldest martian meteorite

    The formation and differentiation of the crust of Mars in the first tens of millions of years after its accretion can only be deciphered from incredibly limited records. The martian breccia NWA 7034 and its paired stones is one of them. This meteorite contains the oldest martian igneous material ever dated: ~4.5 Ga old. However, its source and geological context have so far remained unknown. Here, we show that the meteorite was ejected 5–10 Ma ago from the north-east of the Terra Cimmeria—Sirenum province, in the southern hemisphere of Mars. More specifically, the breccia belongs to the ejecta deposits of the Khujirt crater formed 1.5 Ga ago, and it was ejected as a result of the formation of the Karratha crater 5–10 Ma ago. Our findings demonstrate that the Terra Cimmeria—Sirenum province is a relic of the differentiated primordial martian crust, formed shortly after the accretion of the planet, and that it constitutes a unique record of early crustal processes. This province is an ideal landing site for future missions aiming to unravel the first tens of millions of years of the history of Mars and, by extension, of all terrestrial planets, including the Earth
