    A plasmid bearing a cDNA copy of the genome of bovine viral diarrhea virus (BVDV), chimeric derivatives of the plasmid and a method of producing an infectious bovine viral diarrhea virus using the plasmid are disclosed. The invention relates to a plasmid DNA molecule that replicates easily in E. coli and contains a sufficient portion of the genome of BVDV, cloned as cDNA, to be a suitable template to produce RNA in vitro which, upon transfection into bovine cells, gives rise to infectious BVDV. The BVDV created by the process of the invention can be engineered for use as a vector in many advantageous applications

    This paper presents a pre-amplified detector receiver based on a 250 nm InP/InGaAs/InP double heterojunction bipolar transistor (DHBT) process available from the Teledyne scientific. The front end consists of a double slot antenna followed by a five stage low noise amplifier and a detector, all integrated onto the same circuit. Results of measured responsivity and noise are presented. The receiver is characterized through measuring its response to hot (293) and cold (78) K terminations. Measurements of the voltage noise spectrum at the video output of the receiver are presented and can be used to derive the temperature resolution of the receiver for a specific video bandwidth

    Glutathione accelerates sodium channel inactivation in excised rat axonal membrane patches

    The effects of glutathione were studied on the gating behaviour of sodium channels in membrane patches of rat axons. Depolarizing pulses from –120 to –40 mV elicited sodium currents of up to 500 pA, indicating the simultaneous activation of up to 250 sodium channels. Inactivation of these channels in the excised, inside-out configuration was fitted by two time constants ( h1=0.81 ms; h2= 5.03 ms) and open time histograms at 0 mV revealed a biexponential distribution of channel openings ( short=0.28 ms; long=3.68 ms). Both, the slow time constant of inactivation and the long lasting single channel openings disappeared after addition of the reducing agent glutathione (2–5 mM) to the bathing solution. Sodium channels of excised patches with glutathione present on the cytoplasmatic face of the membrane had inactivation kinetics similar to channels recorded in the cell-attached configuration. These observations indicate that redox processes may contribute to the gating of axonal sodium channels

    Flux-free conductance modulation in a helical Aharonov-Bohm interferometer

    A novel conductance oscillation in a twisted quantum ring composed of a helical atomic configuration is theoretically predicted. Internal torsion of the ring is found to cause a quantum phase shift in the wavefunction that describes the electron's motion along the ring. The resulting conductance oscillation is free from magnetic flux penetrating inside the ring, which is in complete contrast with the ordinary Aharonov-Bohm effect observed in untwisted quantum rings.Comment: 10 pages, 4 figure

    Effect of superficial treatment with new natural antioxidant on salmon (Salmo salar) lipid oxidation

    Lipid oxidation is one of the main factors responsible for the quality loss in refrigerated and frozen stored fish products. The aim of the study was to evaluate the effect of superficial treatment of Atlantic salmon (Salmo salar) with dihydroquercetin (DHQ) solutions on the hydrolytic and oxidative changes in fish lipids during refrigerated storage. It was found that treatment with DHQ solution (1.0 g l−1) reduced approximately twice the free fatty acids content of chilled stored salmon. After 11 days of storage at 1 °С, the contents of hydroperoxides (HPO) and 2-thiobarbituric acid reactive substances (TBARS) of these samples decreased with 45.00 and 0.91 mg MDA/kg, respectively. The share of saturated fatty acids (SFA), monounsaturated fatty acids (MUFA), and polyunsaturated fatty acids (PUFA) in control and experimental samples did not differ significantly (P>0.05). Results obtained show that the superficial treatment of salmon with DHQ solution (1.0 g l−1) delayed the hydrolytic and oxidative changes in fish lipids significantly, thus preserving the salmon freshness up to 11 days of storage at 1 °С

    Detection of interstellar hydrogen peroxide

    The molecular species hydrogen peroxide, HOOH, is likely to be a key ingredient in the oxygen and water chemistry in the interstellar medium. Our aim with this investigation is to determine how abundant HOOH is in the cloud core {\rho} Oph A. By observing several transitions of HOOH in the (sub)millimeter regime we seek to identify the molecule and also to determine the excitation conditions through a multilevel excitation analysis. We have detected three spectral lines toward the SM1 position of {\rho} Oph A at velocity-corrected frequencies that coincide very closely with those measured from laboratory spectroscopy of HOOH. A fourth line was detected at the 4{\sigma} level. We also found through mapping observations that the HOOH emission extends (about 0.05 pc) over the densest part of the {\rho} Oph A cloud core. We derive an abundance of HOOH relative to that of H_2 in the SM1 core of about 1\times10^(-10).     This is a report on the findings of the dark matter science working group for the white paper on the status and future of TeV gamma-ray astronomy. The white paper was commissioned by the American Physical Society, and the full white paper can be found on astro-ph (arXiv:0810.0444). This detailed section discusses the prospects for dark matter detection with future gamma-ray experiments, and the complementarity of gamma-ray measurements with other indirect, direct or accelerator-based searches.     Detection of the hydroperoxyl radical HO2 toward \rho Oph A: Additional constraints on the water chemical network

    Context: Hydrogen peroxide (HOOH) was recently detected toward \rho Oph A. Subsequent astrochemical modeling that included reactions in the gas phase and on the surface of dust grains was able to explain the observed abundance, and highlighted the importance of grain chemistry in the formation of HOOH as an intermediate product in water formation. This study also predicted that the hydroperoxyl radical HO2, the precursor of HOOH, should be detectable. Aims: We aim at detecting the hydroperoxyl radical HO2 in \rho Oph A. Methods: We used the IRAM 30m and the APEX telescopes to target the brightest HO2 lines at about 130 and 260 GHz. Results: We detect five lines of HO2 (comprising seven individual molecular transitions). The fractional abundance of HO2 is found to be about 1e-10, a value similar to the abundance of HOOH. This observational result is consistent with the prediction of the above mentioned astrochemical model, and thereby validates our current understanding of the water formation on dust grains. Conclusions: This detection, anticipated by a sophisticated gas-grain chemical model, demonstrates that models of grain chemistry have improved tremendously and that grain surface reactions now form a crucial part of the overall astrochemical network.Comment: 5 pages, 4 figures, accepted for publication as a Letter to the Editor in Astronomy and Astrophysic

    Squeezed between shells? On the origin of the Lupus I molecular cloud. - II. APEX CO and GASS HI observations

The Lupus I cloud is found between the Upper-Scorpius (USco) and the Upper-Centaurus-Lupus (UCL) sub-groups of the Scorpius-Centaurus OB-association, where the expanding USco H I shell appears to interact with a bubble currently driven by the winds of the remaining B-stars of UCL. Aims. We investigate if the Lupus I molecular could have formed in a colliding flow, and in particular, how the kinematics of the cloud might have been influenced by the larger scale gas dynamics. Methods. We performed APEX 13CO(2–1) and C 18O(2–1) line observations of three distinct parts of Lupus I that provide kinematic information on the cloud at high angular and spectral resolution. We compare those results to the atomic hydrogen data from the GASS H i survey and our dust emission results presented in the previous paper. Based on the velocity information, we present a geometric model for the interaction zone between the USco shell and the UCL wind bubble. Results. We present evidence that the molecular gas of Lupus I is tightly linked to the atomic material of the USco shell. The CO emission in Lupus I is found mainly at velocities between vLSR = 3–6 km s−1 which is in the same range as the H i velocities. Thus, the molecular cloud is co-moving with the expanding USco atomic H i shell. The gas in the cloud shows a complex kinematic structure with several line-of-sight components that overlay each other. The non-thermal velocity dispersion is in the transonic regime in all parts of the cloud and could be injected by external compression. Our observations and the derived geometric model agree with a scenario where Lupus I is located in the interaction zone between the USco shell and the UCL wind bubble. Conclusions. The kinematics observations are consistent with a scenario where the Lupus I cloud formed via shell instabilities. The particular location of Lupus I between USco and UCL suggests that counter-pressure from the UCL wind bubble and pre-existing density enhancements, perhaps left over from the gas stream that formed the stellar subgroups, may have played a role in its formation.Peer reviewedFinal Accepted Versio

    Why less may be more: a mixed methods study of the work and relatedness of 'weak ties' in supporting long-term condition self-management

    Background: The distribution of the roles and responsibilities of long-term condition management (LTCM) outside of formal health services implicates a wide set of relationships and activities of involvement. Yet, compared to studies of professional implementation, patient systems of implementation remain under-investigated. The aim of this paper is to explore the work, meaning and function attributed to ‘weaker’ ties relative to other more bonding relationships in order to identify the place of these within a context of systems of support for long-term conditions. Methods: This is a mixed methods survey with nested qualitative study. A total of 300 people from deprived areas in the North West of England with chronic illnesses took part in a survey conducted in 2010 to 2011. A concentric circles diagram was used as a research tool with which participants identified 2,544 network members who contributed to illness management. Notions of ‘work’ were used to describe activities associated with chronic illness and to identify how weaker ties are included and perceived to be involved through social network members (SNM) contributions. Results: The results provide an articulation of how SNMs are substantially involved in weak tie illness management. Weaker ties constituted 16.1% of network membership involved in illness work. The amount of work undertaken was similar but less than that of stronger ties. Weaker ties appeared more durable and less liable to loss over time than stronger ties. The qualitative accounts suggested that weak ties enabled the moral positioning of the self-managing ‘self’ and acted on the basis of a strong sense of reciprocity. Conclusions: Weak ties act as an acceptable bridge between a sense of personal agency and control and the need for external support because it is possible to construct a sense of moral acceptability through reciprocal exchange. Access to weak tie resources needs to be taken into account when considering the ways in which systems of health implementation for chronic illness are designed and delivered
