Characterization of the transport of lysine-containing dipeptides by PepT1orthologs expressed in Xenopus laevis oocytes.
During digestion, dietary proteins cleaved in di and tri-peptides are translocated from the intestinal lumen into the enterocytes via PepT1 (SLC15A1) using an inwardly directed proton electrochemical gradient. The kinetic properties in various PepT1 orthologs (Dicentrarchus labrax, Oryctolagus cuniculus, Danio rerio) have been explored to determine the transport efficiency of different combinations of lysine, methionine, and glycine. Species-specific differences were observed. Lys-Met resulted the best substrate at all tested potentials in sea bass and rabbit PepT1, whereas in the zebrafish transporter all tested dipeptides (except Gly-Lys) elicited similar currents independently on the charge position or amino acid composition. For the sea bass and rabbit PepT1, kinetic parameters, K0.5 and Imax and their ratio, show the importance of the position of the charged lysine in the peptide. The PepT1 transporter of these species has very low affinity for Lys-Lys and Gly-Lys; this reduces the transport efficiency which is instead higher for Lys-Met and Lys-Gly. PepT1 from zebrafish showed relatively high affinity and excellent transport efficiency for Met-Lys and Lys-Met. These data led
us to speculate about the structural determinants involved in substrate interaction according to the model proposed for this transporter
Pasta consumption and connected dietary habits: Associations with glucose control, adiposity measures, and cardiovascular risk factors in people with type 2 diabetes—TOSCA.IT study
Background: Pasta is a refined carbohydrate with a low glycemic index. Whether pasta shares the metabolic advantages of other low glycemic index foods has not really been investigated. The aim of this study is to document, in people with type-2 diabetes, the consumption of pasta, the connected dietary habits, and the association with glucose control, measures of adiposity, and major cardiovascular risk factors. Methods: We studied 2562 participants. The dietary habits were assessed with the European Prospective Investigation into Cancer and Nutrition (EPIC) questionnaire. Sex-specific quartiles of pasta consumption were created in order to explore the study aims. Results: A higher pasta consumption was associated with a lower intake of proteins, total and saturated fat, cholesterol, added sugar, and fiber. Glucose control, body mass index, prevalence of obesity, and visceral obesity were not significantly different across the quartiles of pasta intake. No relation was found with LDL cholesterol and triglycerides, but there was an inverse relation with HDL-cholesterol. Systolic blood pressure increased with pasta consumption; but this relation was not confirmed after correction for confounders. Conclusions: In people with type-2 diabetes, the consumption of pasta, within the limits recommended for total carbohydrates intake, is not associated with worsening of glucose control, measures of adiposity, and major cardiovascular risk factors
Proteomic analysis of dopamine and \u3b1-synuclein interplay in a cellular model of Parkinson's disease pathogenesis
Altered dopamine homeostasis is an accepted mechanism in the pathogenesis
of Parkinson\u2019s disease. a-Synuclein overexpression and impaired disposal
contribute to this mechanism. However, biochemical alterations
associated with the interplay of cytosolic dopamine and increased a-synuclein
are still unclear. Catecholaminergic SH-SY5Y human neuroblastoma
cells are a suitable model for investigating dopamine toxicity. In the present
study, we report the proteomic pattern of SH-SY5Y cells overexpressing
a-synuclein (1.6-fold induction) after dopamine exposure. Dopamine
itself is able to upregulate a-synuclein expression. However, the effect is
not observed in cells that already overexpress a-synuclein as a consequence
of transfection. The proteomic analysis highlights significant changes in 23
proteins linked to specific cellular processes, such as cytoskeleton structure
and regulation, mitochondrial function, energetic metabolism, protein synthesis,
and neuronal plasticity. A bioinformatic network enrichment procedure
generates a significant model encompassing all proteins and allows us
to enrich functional categories associated with the combination of factors
analyzed in the present study (i.e. dopamine together with a-synuclein). In
particular, the model suggests a potential involvement of the nuclear factor kappa B pathway that is experimentally confirmed. Indeed, a-synuclein significantly

Precision Measurement of KS Meson Lifetime with the KLOE detector
Using a large sample of pure, slow, short lived K0 mesons collected with KLOE
detector at DaFne, we have measured the KS lifetime. From a fit to the proper
time distribution we find tau = (89.562 +- 0.029_stat +- 0.043_syst) ps. This
is the most precise measurement today in good agreement with the world average
derived from previous measurements. We observe no dependence of the lifetime on

Precision measurement of the Dalitz plot distribution with the KLOE detector
Using fb of data collected with
the KLOE detector at DANE, the Dalitz plot distribution for the decay is studied with the world's largest sample of events. The Dalitz plot density is parametrized as a polynomial
expansion up to cubic terms in the normalized dimensionless variables and
. The experiment is sensitive to all charge conjugation conserving terms of
the expansion, including a term. The statistical uncertainty of all

Measurement of the slope parameter with the KLOE detector
We present a measurement of the slope parameter for the decay, with the KLOE experiment at the DANE -factory,
based on a background free sample of 17 millions mesons produced
in radiative decays. By fitting the event density in the Dalitz plot we
determine .
The result is in agreement with recent measurements from hadro- and
