    Fresnel zone plate telescopes for X-ray imaging II: numerical simulations with parallel and diverging beams

    We present the results of simulations of shadows cast by a zone plate telescope which may have one to four pairs of zone plates. From the shadows we reconstruct the images under various circumstances. We discuss physical basis of the resolution of the telescope and demonstrate this by our simulations. We allow the source to be at a finite distance (diverging beam) as well as at an infinite distance (parallel beam) and show that the resolution is worsened when the source is nearby. By reconstructing the zone plates in a way that both the zone plates subtend the same solid angles at the source, we obtain back high resolution even for sources at a finite distance. We present simulated results for the observation of the galactic center and show that the sources of varying intensities may be reconstructed with accuracy. Results of these simulations would be of immense use in interpreting the X-ray images from recently launched CORONAS-PHOTON satellite.Comment: 17 pages, 36 figures, Published in Experimental Astronom

    Instruments of RT-2 Experiment onboard CORONAS-PHOTON and their test and evaluation III: Coded Aperture Mask and Fresnel Zone Plates in RT-2/CZT Payload

    Imaging in hard X-rays of any astrophysical source with high angular resolution is a challenging job. Shadow-casting technique is one of the most viable options for imaging in hard X-rays. We have used two different types of shadow-casters, namely, Coded Aperture Mask (CAM) and Fresnel Zone Plate (FZP) pair and two types of pixellated solid-state detectors, namely, CZT and CMOS in RT-2/CZT payload, the hard X-ray imaging instrument onboard the CORONAS-PHOTON satellite. In this paper, we present the results of simulations with different combinations of coders (CAM & FZP) and detectors that are employed in the RT-2/CZT payload. We discuss the possibility of detecting transient Solar flares with good angular resolution for various combinations.     Correlated dynamics in egocentric communication networks

    We investigate the communication sequences of millions of people through two different channels and analyze the fine grained temporal structure of correlated event trains induced by single individuals. By focusing on correlations between the heterogeneous dynamics and the topology of egocentric networks we find that the bursty trains usually evolve for pairs of individuals rather than for the ego and his/her several neighbors thus burstiness is a property of the links rather than of the nodes. We compare the directional balance of calls and short messages within bursty trains to the average on the actual link and show that for the trains of voice calls the imbalance is significantly enhanced, while for short messages the balance within the trains increases. These effects can be partly traced back to the technological constrains (for short messages) and partly to the human behavioral features (voice calls). We define a model that is able to reproduce the empirical results and may help us to understand better the mechanisms driving technology mediated human communication dynamics.Comment: 7 pages, 6 figure

    Strong signature of natural selection within an FHIT intron implicated in prostate cancer risk

    Previously, a candidate gene linkage approach on brother pairs affected with prostate cancer identified a locus of prostate cancer susceptibility at D3S1234 within the fragile histidine triad gene (FHIT), a tumor suppressor that induces apoptosis. Subsequent association tests on 16 SNPs spanning approximately 381 kb surrounding D3S1234 in Americans of European descent revealed significant evidence of association for a single SNP within intron 5 of FHIT. In the current study, resequencing and genotyping within a 28.5 kb region surrounding this SNP further delineated the association with prostate cancer risk to a 15 kb region. Multiple SNPs in sequences under evolutionary constraint within intron 5 of FHIT defined several related haplotypes with an increased risk of prostate cancer in European-Americans. Strong associations were detected for a risk haplotype defined by SNPs 138543, 142413, and 152494 in all cases (Pearson's χ2 = 12.34, df 1, P = 0.00045) and for the homozygous risk haplotype defined by SNPs 144716, 142413, and 148444 in cases that shared 2 alleles identical by descent with their affected brothers (Pearson's χ2 = 11.50, df 1, P = 0.00070). In addition to highly conserved sequences encompassing SNPs 148444 and 152413, population studies revealed strong signatures of natural selection for a 1 kb window covering the SNP 144716 in two human populations, the European American (π = 0.0072, Tajima's D= 3.31, 14 SNPs) and the Japanese (π = 0.0049, Fay & Wu's H = 8.05, 14 SNPs), as well as in chimpanzees (Fay & Wu's H = 8.62, 12 SNPs). These results strongly support the involvement of the FHIT intronic region in an increased risk of prostate cancer. © 2008 Ding et al

    The Dichotomous Pattern of IL-12R and IL-23R Expression Elucidates the Role of IL-12 and IL-23 in Inflammation

    IL-12 and IL-23 cytokines respectively drive Th1 and Th17 type responses. Yet, little is known regarding the biology of these receptors. As the IL-12 and IL-23 receptors share a common subunit, it has been assumed that these receptors are co-expressed. Surprisingly, we find that the expression of each of these receptors is restricted to specific cell types, in both mouse and human. Indeed, although IL-12Rβ2 is expressed by NK cells and a subset of γδ T cells, the expression of IL-23R is restricted to specific T cell subsets, a small number of B cells and innate lymphoid cells. By exploiting an IL-12- and IL-23-dependent mouse model of innate inflammation, we demonstrate an intricate interplay between IL-12Rβ2 NK cells and IL-23R innate lymphoid cells with respectively dominant roles in the regulation of systemic versus local inflammatory responses. Together, these findings support an unforeseen lineage-specific dichotomy in the in vivo role of both the IL-12 and IL-23 pathways in pathological inflammatory states, which may allow more accurate dissection of the roles of these receptors in chronic inflammatory diseases in humans

    Comprehensive analysis of blood cells and plasma identifies tissue-specific miRNAs as potential novel circulating biomarkers in cattle

    Abstract Background The potential of circulating miRNAs as biomarkers of tissue function, both in health and disease, has been extensively demonstrated in humans. In addition, circulating miRNA biomarkers offer significant potential towards improving the productivity of livestock species, however, such potential has been hampered by the absence of information on the nature and source of circulating miRNA populations in these species. In addition, many miRNAs originally proposed as robust biomarkers of a particular tissue or disease in humans have been later shown not to be tissue specific and thus to actually have limited biomarker utility. In this study, we comprehensively analysed miRNA profiles in plasma and cell fractions of blood from cattle with the aim to identify tissue-derived miRNAs which may be useful as biomarkers of tissue function in this important food animal species. Results Using small RNA sequencing, we identified 92 miRNAs with significantly higher expression in plasma compared to paired blood cell samples (n = 4 cows). Differences in miRNA levels between plasma and cell fractions were validated for eight out of 10 miRNAs using RT-qPCR (n = 10 cows). Among miRNAs found to be enriched in plasma, we confirmed miR-122 (liver), miR-133a (muscle) and miR-215 (intestine) to be tissue-enriched, as reported for other species. Profiling of additional miRNAs across different tissues identified the human homologue, miR-802, as highly enriched specifically in liver. Conclusions These results provide novel information on the source of bovine circulating miRNAs and could significantly facilitate the identification of production-relevant tissue biomarkers in livestock. In particular, miR-802, a circulating miRNA not previously identified in cattle, can reportedly regulate insulin sensitivity and lipid metabolism, and thus could potentially provide a specific biomarker of liver function, a key parameter in the context of post-partum negative energy balance in dairy cows

    Role of PACAP and VIP Signalling in Regulation of Chondrogenesis and Osteogenesis

    Pituitary adenylate cyclase activating polypeptide (PACAP) and vasoactive intestinal peptide (VIP) are multifunctional proteins that can regulate diverse physiological processes. These are also regarded as neurotrophic and anti-inflammatory substances in the CNS, and PACAP is reported to prevent harmful effects of oxidative stress. In the last decade more and more data accumulated on the similar function of PACAP in various tissues, but its cartilage- and bone-related presence and functions have not been widely investigated yet. In this summary we plan to verify the presence and function of PACAP and VIP signalling tool kit during cartilage differentiation and bone formation. We give evidence about the protective function of PACAP in cartilage regeneration with oxidative or mechanically stress and also with the modulation of PACAP signalling in vitro in osteogenic cells. Our observations imply the therapeutic perspective that PACAP might be applicable as a natural agent exerting protecting effect during joint inflammation and/or may promote cartilage regeneration during degenerative diseases of articular cartilage

    Multivariate Analysis of Dopaminergic Gene Variants as Risk Factors of Heroin Dependence

    BACKGROUND: Heroin dependence is a debilitating psychiatric disorder with complex inheritance. Since the dopaminergic system has a key role in rewarding mechanism of the brain, which is directly or indirectly targeted by most drugs of abuse, we focus on the effects and interactions among dopaminergic gene variants. OBJECTIVE: To study the potential association between allelic variants of dopamine D2 receptor (DRD2), ANKK1 (ankyrin repeat and kinase domain containing 1), dopamine D4 receptor (DRD4), catechol-O-methyl transferase (COMT) and dopamine transporter (SLC6A3) genes and heroin dependence in Hungarian patients. METHODS: 303 heroin dependent subjects and 555 healthy controls were genotyped for 7 single nucleotide polymorphisms (SNPs) rs4680 of the COMT gene; rs1079597 and rs1800498 of the DRD2 gene; rs1800497 of the ANKK1 gene; rs1800955, rs936462 and rs747302 of the DRD4 gene. Four variable number of tandem repeats (VNTRs) were also genotyped: 120 bp duplication and 48 bp VNTR in exon 3 of DRD4 and 40 bp VNTR and intron 8 VNTR of SLC6A3. We also perform a multivariate analysis of associations using Bayesian networks in Bayesian multilevel analysis (BN-BMLA). FINDINGS AND CONCLUSIONS: In single marker analysis the TaqIA (rs1800497) and TaqIB (rs1079597) variants were associated with heroin dependence. Moreover, -521 C/T SNP (rs1800955) of the DRD4 gene showed nominal association with a possible protective effect of the C allele. After applying the Bonferroni correction TaqIB was still significant suggesting that the minor (A) allele of the TaqIB SNP is a risk component in the genetic background of heroin dependence. The findings of the additional multiple marker analysis are consistent with the results of the single marker analysis, but this method was able to reveal an indirect effect of a promoter polymorphism (rs936462) of the DRD4 gene and this effect is mediated through the -521 C/T (rs1800955) polymorphism in the promoter

    Characterization of Static- and Fatigue-Loaded Carbon Composites by X-Ray CT

    Computed Tomography methodologies were investigated to better understanding their possibility to improve the knowledge and a correct understanding of the behavior of thin Carbon-Polymer composites when static or fatigue loaded to failure. We applied CT to study a set of six aerospace grade carbon fiber/thermoplastic or fiber/thermoset matrix composites. The samples were subjected to either static or high-stress fatigue loading in tension. Both notched (central circular hole) and unnotched specimens were examined. We investigateed a high-temperature thermoplastic polyimide composite sample by acquiring CT data sets before, during (at set intervals), and after full-reversal (tension-compression), low-stress fatigue loading at the upper use temperature.</p
