    Low molecular weight proteins of barley related to food allergy

    As protein composition and modification are critical for malt and beer quality, proteomic approaches have the potential to improve malting and brewing processes, as well as to monitor and characterize important low molecular weight proteins related to food allergy. New product compositions and industrial processes create additional needs that require much greater technological development. The detection and quantification of allergenic proteins by mass spectrometry is promising and contributes to greater accuracy, thereby significantly improving consumer information. In the case of allergenic proteins, a wide range of isoforms, post-translational modifications and other structural changes during the technological process can increase or decrease their allergenicity. In this context, we focused on tracking barley proteins related to pathogens, a large proportion of which are allergy-related. These mainly include a group of protease/amylase inhibitors such as α-amylase/trypsin inhibitor CMa, CMb, CMe, α-amylase inhibitor BDAI-1.     Identification of a Protein with Antioxidant Activity that is Important for the Protection against Beer Ageing

    This study was carried out with fresh Australian lager beer which was sampled directly off the production line, the same samples aged for 12 weeks at 30 °C, and the vintage beer which was kept at 20 °C for 5 years. Characteristic Australian lager flavour was maintained in the fresh and vintage beers but was lost in the aged beer. Sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) and free thiol group labelling analyses of beer proteins found that this flavour stability correlated with the presence of an unknown 10 kilodaltons (kDa) protein with a higher level of free thiols. The protein was purified by size-exclusion chromatography, then peptide sequencing and database matching identified it as the barley lipid transfer protein (LTP1). Further characterisation using diphenylpicrylhydrazyl (DPPH) free radical scavenging and a Saccharomyces cerevisiae-based antioxidant screening assay demonstrated that the LTP1 protein was active in DPPH reduction and antioxidant activity. The absence of free thiol in the aged beer indicates that the thiol functional groups within the LTP1 protein were saturated and suggests that it is important in the flavour stability of beer by maintaining reduction capacity during the ageing process

    An efficient reduction of double bonds conjugated with nitrile groups and acid or base sensitive furan rings with 2-phenylbenzimidazoline generated in situ has been successfully accomplished with high yields and excellent selectivity. The employed reducing agent was prepared in one step from ordinary chemicals. The other advantages of the presented method include mild and convenient reaction conditions, a benign and cost effective reagent, simple work-up and separation of the products. As this process does neither affect cyano and nitro groups nor furan rings, it is a valuable alternative when metal-catalyzed hydrogenations or borohydride reductions have failed
